温敏型亚麻雄性不育系育性相关基因的RAPD初探

摘    要:通过优化组合,建立了适合本试验的亚麻基因组DNA最佳RAPD反应体系;应用分离群体分组分析法(BSA),利用1S×Argus杂交组合F2分离群体所构建的可育基因池(BF)和不育基因池(BS)进行育性基因的RAPD标记分析.经多次重复试验,在所用的240条10 bp随机引物中筛选出1条多态性引物S1113,其序列为:CACGGCACAA.S1113在可育基因池中扩增出大小约为450 bp的多态性DNA片段,暂命名为S1113450,可能与亚麻温敏雄性不育系可育基因连锁.

Abstract:With bulked segregant analysis(BSA),bulked male-fertile DNA pool and bulked male-sterile DNA pool were used for RAPD analysis which were respectively consisted of 10 fertile plant individuals and 10 sterile plants from F2 generations of 1S ×Argus. PCR amplification was carried out with 240 10 by primers. After many experiments,the primer 51113(CACGGCACAA)was found that produced polymorphism DNA fragment between two bulked DNA pools. The polymorphism amplificanon fragment (450 bp) appeared in bulked male-sterile DNA pool andpossibly linked to male-fertility gene for the thermo-sensitivity male-sterile line of flax.
