摘    要:采用16条10 bp随机引物对采自浙江、江苏、安徽、云南、福建、海南和山东7省茶区的假眼小绿叶蝉种群进行RAPD标记,探讨不同地域种群的亲缘关系。结果表明:16个随机引物从供试的叶蝉种群中共得到121条稳定清晰的片段,片段长度约为200-2 000 bp,其中78条多态性条带,多态性为64.46%。根据RAPD图谱上扩增片段的共享度计算出Nei’s遗传距离指数,运用NTSYS软件中的UPGMA方法对这些谱带进行聚类分析,构建分子系统树,确定7个种群间的亲缘关系。系统树显示:安徽省十字铺和福建省福安两种群首先聚为一支,山东临沂种群和云南西双版纳种群与最后其他5种群聚类。表明假眼小绿叶蝉亲缘关系的远近与地理距离有一定的相关性。

Abstract:Random amplified polymorphic DNA(RAPD) techniques were utilized to assess the genetic polymorphism and relationships among seven populations of tea green leafhopper,Empoasca vitis(Gothe) in seven tea gardens from seven provinces of China,respectively.Genomic DNA of tested leafhoppers was amplified by 16 random primers with 10 bp,then PCR products were separated by agarose gel electrophoresis.One hundred and twenty-one reproducible fragments ranging from 200-2000 bp were obtained,in which 78 were population-specific bands with the DNA polymorphism of 64.46 %.The amplified bands were scored as present(1) or absent(0) for each leafhopper population DNA to calculate genetic similarity(F) and genetic distance(D).The cluster analysis was done and dendrogram based on the value of D or F was constructed used NTSYS software via UPGMA method.The dendrogram showed strong similarities within populations.Anhui Shizipu population and Fujian Fu'an population were the most similar,clustered first;however Shandong Linyi population and Yunnan Xishuangbanna population had far genetic distance with other five populations and clustered lastly.This study that the genetic distance among Empoasca vitis(Gothe) populations correlates and geographical distance to some degree.
