半夏RAPD分子标记反应体系优化

摘    要:从半夏中提取基因组DNA作为模板进行RAPD反应的优化试验,获得RAPD-PCR扩增反应的最佳条件:25μL体系中Mg2+1.5 mmol/L;dNTPs各0.2 mmol/L;模板DNA100 ng;引物0.25μmol/L;Taq酶活性值1.0 U。

Abstract:Genomic DNAs Pinellia ternata was used as template to optimize the random amplified polymorphic DNA(RAPD) reaction system,and some essential factors which might take effects on the results of RAPD were investigated.The optimization of a 25 u1 RAPD-PCR system was determined as followed: 1.5 mmo1/L Mg2+,0.2 mmol/L dNTPs for each,100 ng template DNA,0.25 μmol/L primer and 1.0 U Taq DNA polymerase.
