紫花苜蓿高效遗传转化受体系统的建立

摘    要:以新疆大叶和新牧一号苜蓿下胚轴和子叶为材料,研究了不同基因型、外植体、培养基及激素配比对其愈伤组织诱导、芽分化和根诱导的影响,建立了适用于这2种苜蓿的再生体系。结果表明,下胚轴是一种较好的外植体材料,其再生能力高于子叶,最高再生率为54.96%;0.2 mg/L KT能辅助2,4-D对愈伤组织的诱导发挥更好的作用;SH培养基相对于MS培养基对愈伤组织的诱导更有效,而MS培养基则有利于愈伤组织的分化;适用于2种苜蓿愈伤组织诱导、分化及生根培养的培养基分别为SH(附加2 mg/L 2,4-D和0.2 mg/L KT),MS(附加1.0 mg/LBAP和0.3 mg/L NAA)和不含激素的1/2 MS(扩繁时附加1 mg/L IBA)。

Abstract:Using hypocotyls and cotyledon of Xinjiang Daye and Xinmu No.1 as materials,the effect of genotypes,explants,medium and hormones on callus induction,embryos,differentiation and root induction were studied.The results showed that in callus induction and in vitro regeneration by hypocotyls,the highest regeneration rate was 54.96%,stronger than the cotyledons and 2,4-D combined with 0.2 mg/L kinetin(KT)could act better for callus induction.The SH medium was a very effective medium for the callus induction compared to MS medium,whereas the latter was more advantageous to embryo induction than the former medium.As to XinJiang Daye and Xinmu No.1,the applicable media for induction of callus,embryos and roots were SH containing 2 mg/L 2,4-D and 0.2 mg/L KT,MS containing 1.0 mg/L BAP and 0.3 mg/L NAA,and hormone-free 1/2 MS containing 1.0 mg/L IBA when propagated respectively.
