南瓜离体培养及植株再生的研究

摘    要:研究了不同外植体、消毒方法、激素水平等对南瓜离体培养及植株再生的影响。结果表明:以子叶作为外植体可以通过愈伤组织途径之后形成再生植株;而以子叶节作为外植体可以直接诱导不定芽形成再生植株。诱导不定芽的最佳培养基组合为MS+1.0 mg/L6-BA+0.5 mg/L NAA;诱导愈伤组织的最佳培养基组合为MS+0.5mg/L 6-BA+0.5mg/L 2,4-D;诱导南瓜不定根的最佳培养基组合为MS+1.0 mg/L6-BA+0.5 mg/LIAA。

Abstract:Adventitious shoots were regenerated from cotyledon and cotyledon nodes of pumpkin.The result showed that cotyledon being used as explants,the plant regeneration can obtain from the way of growing callus,and cotyledon nodes being used as explants,plant regeneration can obtain direct from inducing adventitious bud.At the same time,the result showed that the optimum medium for adventitious bud was MS medium supplement with 1 mg/L 6-BA 0.5 mg/L NAA.The callus induced in the medium of MS medium supplement with 0.5 mg/L 6-BA 0.5 mg/L 2,4-D was best;and the adventitious root was induced on MS medium supplemented with 1 mg/L 6-BA 0.5 mg/L IAA.
