摘    要:以鄂豆6号成熟种子无菌苗下胚轴1/2切段、1/3切段为外植体,按顺插、反插、平躺3种方式接种在添加植物生长调节剂NAA、6-BA、2,4-D、KT配伍的1/2MS基本培养基中诱导器官发生。结果表明,添加NAA0.1mg.L-1 KT1.0mg.L-1培养基中下胚轴生根率较高,添加NAA0.1mg.L-1 KT1.0mg.L-1、2,4-D0.1mg.L-1 6-BA1.5mg.L-1、2,4-D0.1mg.L-1 KT1.5mg.L-1可诱导芽的发生;1/2切段生根和发芽比1/3切段的好;反插根的发生较多,顺插有利于芽的形成。

Abstract:The 1/2 and 1/3 segments of asepsis hypocotyls from the mature seed of soybean E-Dou No.6 were selected as explants, and then the explants were inoculated on 1/2MS medium in which the plant growth regulator, NAA, 6-BA,2,4-D and KT, were added to induce organogenesis. In addition, the explants were planted as three inoculating manners, cis-insertion, trans- insertion and lying. The results showed that the induced rate of hypocotyls radication was quite high in medium containing NAA0.1 mgL-1 KT1.0 mgL-1,germination could be induced by medium containing NAA 0.1 mgL-1 KT1.0 mgL-1,2,4-D0.1 mgL-1 6-BA1.5 mgL-1 and 2,4-D0.1 mgL-1 KT1.5 mgL-1 respectively. The radication and germination from 1/2 segments were better than that from 1/3 segments, the planted manner of trans-insertion was more effective for organogenesis, and cis-insertion was good for germination.
