马尾藻多糖佐剂对猪PRRS疫苗免疫效果影响的研究

摘    要:分别以马尾藻多糖的不同剂量与猪PRRS灭活抗原配制成完全佐剂疫苗,免疫5组非猪PRRSV免疫猪,并于首免的第0、20、40、50d采血,测定其体液免疫和细胞免疫水平。结果表明:(1)各马尾藻多糖佐剂组T淋巴细胞转化率(61.4%、65.0%、63.4%)均高于空白组1(52.8%)及普通佐剂疫苗组(59.6%),各马尾藻多糖佐剂组与空白对照组间差异显著(P〈0.05);(2)各马尾藻多糖佐剂组的T淋巴细胞亚群数量均高于空白组和普通佐剂疫苗组,其中以CD3的数量差异最明显(P〈0.05),但马尾藻多糖组与普通佐剂疫苗组的CD8和NK细胞亚群的测定值差异不显著;(3)间接ELISA法测定各马尾藻多糖佐剂组血清中抗体的活性(0.631、1.015、0.688)均高于两对照组(0.518、0.568),其中100mg/头份马尾藻多糖佐剂组与对照组间的差异显著(P〈0.05)。可见,以马尾藻多糖作为佐剂即可以提高体液免疫水平,又能增强细胞免疫功能,其中100mg/头份马尾藻多糖剂量的总体免疫效果最佳。

Abstract:In the present study, different dosages (50, 100,200 mg) of Sargassum polysaccharides were used for making complete adjuvant vaccine with PRRS inactivated antigen, and pig groups without PRRS were vaccinated with normal adjuvant vaccine and three kinds of complete adjuvant vaccines, respectively. The check group was not treated with adjuvant vaccine. Every group had five non-immune pigs. Blood of different pigs were sampled to determine the levels of cellimmunity and humoral-immunity at 0, 20, 40 and 50 days after primary vaccination. The results showed that transformation rate of T lymphocyte in polysaccharides groups was 61.4%, 65.0%, and 63.4%, respectively, which was higher than that of the control group (52.8%) and normal adjuvant group (59.6%). And there was significant difference between pclysaccharides groups and control group (P〈0.05). The qualities of T lymphocyte subsets in polysaccharides groups were all higher than that of the other groups, and the increase of CD3 was found to be significant. But there was no significant difference in CD8 and NK cell subsets between polysaccharides groups and normal adjuvant vaccine group. The results of indirect ELISA showed that the antibody activity in serum from polysaccharides groups was 0.631,1.015 and 0.688, respectively, which was higher than that of the control group (0.518) and normal group (0.568), and the difference between 100mg Sargassum polysaccharides group and the control groups was significant. These results demonstrated that Sargassum polysaccharides could improve humoral-imnmnity level as adjuvant and enhance cell-immunity function. The effect of 100 mg Sargassum polysaccharides among all, was found to be the best.
