摘    要:以新牧1号杂花苜蓿和新疆大叶紫花苜蓿的子叶作为外植体,研究激素配比、培养基种类、添加物等对苜蓿愈伤组织诱导和分化的影响。结果表明:新牧1号杂花苜蓿愈伤组织诱导的较优激素配比是2.00 mg/L2,4-D+0.05 mg/L KT,其诱导率达100.0%,三周愈伤直径大小达6.10 mm;较优分化培养基种类是SH培养基,分化率可达58.3%;在MS培养基中添加低浓度KT可使新牧1号杂花苜蓿获得较优的分化效果。新疆大叶紫花苜蓿较优分化培养基种类是SH培养基,分化率可达54.2%,以MSO为基本培养基,添加2.00 mg/LAgNO_3,可使新疆大叶杂花苜蓿的分化率从41.1%增加到63.3%。

Abstract:The experiment was designed to study the effect of hormone combination,medium types, and additives concentration on alfalfa callus induction and differentiation with cotyledon of Medicago varia Martyn.cv.Xinmu No.1 and Medicago sativa L.cv.Daye as explants.The results showed that the better hormone combination for callus induction of Medicago varia Martyn.cv.Xinmu No.1 was 2.00 mg/L 2,4 - D + 0. 05 mg/L KT,the induction rate was 100 percent,three weeks later,the callus diameter reached 6.10 mm;the optimum medium for differentiation was SH medium,its differentiation rate was 58.3%,the optimum hormone combination for differentiation of Medicago.varia Martyn.cv.Xinmu No.1 was MS added with low concentration KT.As for Medicago sativa L.cv.Daye,the optimum medium for differentiation also was SH medium,the differentiation rate was 54.2%,with MSO as the basal medium,adding 2.00 mg/L AgNO_3 could increase differentiation rate of Medicago sativa L.cv.Daye from 41.1%to 63.3%.
