大花蕙兰试管苗玻璃化发生原因及防止技术研究

摘    要:试验结果表明,大花蕙兰试管苗玻璃化的原因主要有3个:一是培养温度的突然升高,造成高温伤害的后遗症,引发玻璃化;二是经过多次继代培养,原球茎过多吸收细胞分裂素6-BA,使体内激素比例严重失调,导致玻璃化;三是长期使用氨、锰、铁、锌等含量较低的培养基,引起玻璃化苗的产生。采取相应的技术措施,可以完全控制玻璃化苗的发生。

Abstract:The present experiments showed that vitrifiable genetic reasons ofcymbidium grandifiorium of tube plantlet had three. Firstly, the culture temperature sudden rise and the high temperature damage caused sequelae of tube plantlet vitrifiation. Secondly,after more subculture, the protocorm derived from Cymbidium grandiflorium absorbed overabundance cytokinin 6-BA (6-benzyladenine),which aroused hormone proportion inbalanc in plant body and generated vitrification. Thirdly, the culture medium which contained lower ammonia, manganese, iron, zinc etc, were longtermly used, and this probably caused vitrification.It concluded that when technical measures were taken,a thorough control of vitrification is possible.
