甘蔗组培亚无性系农艺性状及同工酶变异的分析

摘    要:供试材料为无性繁殖第2代的4个甘蔗组培亚无性系群体.仙游79-83组培亚无性系的性状平均表现为茎变细,茎数/丛增加,株高减少,锤度与供体的无显著差异.新台糖10号组培亚无性系的茎变细,茎数/丛无明显变化,株高增加,锤度比对照有所下降.同一供体在MS和N_6培养基培养出来的亚无性系,4个性状变异的平均表现趋剪一致,但变异幅度有所不同.培养基对不同供体及其不同性状变异的影响有一定差异.同工酶分析表明,不同供体及同一供体在不同培养基下培养出来的亚无性系,其POD和Est的变异均有差异.仙游79-83亚无性系的POD变异率较Est的变异率高,但不同培养基培养出来的亚无性系之间差异明显.新台糖10号的亚无性系则表现为POD的变异率比Est的低,而培养基之间的差异不明显.就供体而言,仙游79-83亚无性系POD的变异率高于新台糖lO号亚无性系的变异率,而Est则因培养基而异.供试亚无性系农艺性状的变异与同工酶的变异有一定的联系,并表明组培对甘蔗无性系变异的影响是有限的.

Abstract:Four populations of sugarcane subclone derived from donor varieties Xianyou 79—83 and ROC10 tissue culture under MS and N_6 medium were analysed on their variations of agronomic character, and POD and Est isozymes. The results showed that the subclones derived from Xianyou 79-83 possessed thinner stalk diameter, shorter plant height, more number of stalk per clump and approximate brix in comparison with donor variety. And the subclones derived from ROC10 had the character displays with thinner stalk diameter, higher plant height, lower brix value and no significant changable number of stalk per clump when compared with donor variety. The variation tendencies of different characters were similar between the subclones derived from a same donor but cultured under different medium. However, the difference of variation range was observed between the subclones cultured under various medium for a particular character. The analysis of isozymes indicated that there were higher variation rates for POD in subclones derived from Xianyou 79—83 than in those from ROC10, cultured under both mediums. For Est, the variation rate was dependent on donor and medium. At the present study, the association of stalk diameter and stalk number per clump with the POD variation was observed in the subclone populations. The results also showed that the range of POD and Est variations was limited.
