福建柑桔种质的RAPD分析

摘    要:从100条随机引物中筛选出21条能扩增出清晰的具有多态性DNA谱带的引物,通过RAPD分析,在分子水平上对福建54份柑桔种质进行鉴定、分类和亲缘关系分析。结果表明:这些种质的相异系数为0.01493-0.48172,说明它们之间的亲缘关系较近;在遗传相似系数为0.45的水平上,54份柑桔种质被分为三大类,其中,太田芦柑(C itrus reticulataB lanoocv.Tiantian lugan)和晚熟芦柑(C itrus reticulataB lanoo cv.W anshu lugan)的亲缘关系最近,柠檬(C itrus lim onBurm.F.)与枳(Poncirus trifoliateRaf.)的亲缘关系最远,与形态学观察及传统分类结果相吻合。此外,还对福建引进的一些杂柑品种的遗传多样性进行了探讨。

Abstract:Identification,classfication and genetic relationship of 54 citrus germplasms in Fujian Province were detected at molecular level by random amplified polymorphic DNA(RAPD) with 21 primers screened from 100 random primers.The results revealed that discordant coefficient of citrus germplasms was 0.01493-0.48172,which indicated that the genetic relationships between them were close.The 54 citrus germplasms were divided into three groups when genetic similarity coefficient was 0.45.The closest relationship was between Citrus reticulata Blanoo cv.Tiantianlugan and Wanshulugan.Citrus limon Burm.F.is highly divergent from Poncirus trifoliate Raf..The results were in consistence with the morphological observation and traditional classification system.Additionally,the genetic diversity of the citrus hybrid varieties in Fujian was researched based on the RAPD analysis.
