摘    要:以垂花蕙兰种子为试验材料,采用种子→原球茎→完整植株的途径,研究基本培养基、植物生长调节剂、有机添加物和活性炭等关键因素对垂花蕙兰种子萌发、原球茎增殖、分化和生根等各培养阶段的影响,探讨垂花蕙兰无菌播种和快速繁殖的关键技术。结果表明,种子在1/2MS + 6-BA 1.0 mg·L-1 + NAA 1.0 mg·L-1 + AC 0.5 g·L-1 +蔗糖20 g·L-1 培养中培养60 d可形成原球茎,种子萌发数可达90粒;原球茎在1/2MS + 6-BA 1.0 mg·L-1 + NAA 0.5 mg·L-1 + KT 1.0 mg·L-1 + AC 0.5 g·L-1 + 蔗糖20 g·L-1 培养基上增殖效果好,增殖系数为6.8;原球茎在1/2MS + 6-BA 0.5 mg·L-1 + NAA 0.1 mg·L-1 + AC 0.5 g·L-1 + 蔗糖20 g·L-1 上培养分化效果好,分化率高达89.2%;幼苗在1/2MS + IBA 1.0 mg·L-1 +50 g·L-1 土豆泥+蔗糖20 g·L-1 上培养30 d可生根,生根率为94.5%,苗高可达5.6 cm。

Abstract:To established a system of aseptic sowing and rapid propagation technology for pendent Cymbidium, the effects of basic culture media, hormone, organic additive and AC on pendent Cymbidium seeds germination, multiplication, differentiation and rooting was investigated by applying the seed→protocorm→the whole plant model.Our results showed that 90 seeds germinated and developed into protocorm in 1/2MS medium containing 1.0 mg·L-16-BA+1.0 mg·L-1NAA+0.5 g·L-1AC+20 g·L-1sugar after 60d.The best multiplication of protocorm (multiplication coefficient 6.8) was reached in 1/2MS medium containing 1.0 mg·L-16-BA+0.5 mg·L-1NAA+1.0 mg·L-1KT+0.5 g·L-1AC+20 g·L-1sugar.Meanwhile, the best differentiation (differentiation rate 89.2%) of protocorm was reached in 1/2MS medium containing 0.5 mg·L-16-BA+0.1 mg·L-1 NAA+0.5 g·L-1AC+20 g·L-1sugar.Furthermore, the best rooting (rooting rate 94.5%, plant length up to 5.6cm) was reached in 1/2MS medium containing 1.0 mg·L-1 NAA +50 g·L-1mashed potato+0.5 g·L-1AC+20g·L-1 sugar after 30 d.
