摘    要:以36份石榴品种(类型)为研究对象,用RAPD技术对其亲缘关系进行分析。从128个随机引物中筛选出12个重复性好、条带清晰的多态性引物,并用这些引物对36份材料进行RAPD扩增,共扩增出111条谱带,平均每个引物9.25条。12个引物扩增出不同的多态性条带,多态性程度达到72.07%。用Statistic分析软件计算出的遗传距离在0.027~0.441之间,并对36份材料进行了UPGA法聚类,RAPD结果的亲缘关系树状图显示,36个品种或类型基因背景较复杂,说明了我国的石榴栽培品种有丰富的种质资源。

Abstract:36 materials of Punica granatum L. were analyzed using random amplified polymorphic DNA markers in this paper.12 primers which had good repeat and clear bands were screened from 128 arbitrary 10-mer primers. 12 random primers were used to amplify 36 pomegranate samples. The total 111 bands were obtained and the average bands per random primer were 9.25. These bands displayed polymorphism. The genetic diversity degree was 72.07%. The genetic distance of 36 materials was between 0.027 and 0.441 by the account of statistic analysis software. A tree diagram of phylogenetic relationship was constructed using UPGA according to these data. The results showed that the genetic background of Punica granatum L. germplasm resources was too complex and abundant to be classified.
