铜对牛卵母细胞体外成熟及体外受精胚胎发育的影响

摘    要:研究了氯化铜对牛卵母细胞体外成熟及体外受精胚胎发育潜力的影响。培养液中铜的浓度以原子吸收光谱法测定。培养液中铜的浓度为0(对照组)、0.46和0.68 mg·L-1。结果表明,试验组在卵母细胞成熟和受精卵发育至2细胞率与对照组差异不显著(P>0.05),但试验组较对照组显著提高了桑椹胚和囊胚的发育率(P<0.05)。在卵母细胞体外成熟培养的0~22 h、受精卵培养的0~48 h、48~96 h、96~144 h和144~192 h,0.46 mg·L-1试验组培养液中铜浓度分别下降了4.3%、8.7%、26.0%、28.3%和30.4%,0.68 mg·L-1试验组培养液中铜浓度分别下降了2.9%、7.4%、23.5%、25.0%和27.9%。培养液中铜对牛早期胚胎培养至桑椹胚和囊胚有重要的作用,早期胚胎发育48 h后对培养液中微量元素铜的需求明显增加。

Abstract:This study was to investigate the effects of chloratum culture-medium on bovine oocytes maturation and embryo development by in vitro fertilization. The concentrations of copper were mensurated by plasma atomic spectrometric techniques. Copper concentrations in culture medium were 0 mg/l (control), 0.46 mg/l and 0.68 mg·L-1. The results were as follows: there were no significant differences in oocytes maturation and 2-cell zygotes between copper and control, and copper increased the rate of morulae and blastocysts (P>0.05). At 0-22 h of oocytes maturation, 0-48 h, 48-96 h, 96-144 h and 144-192 h of zygotes culture, decreased percentage of copper concentrations in 0.46 mg/l treatment were 4.3%, 8.7%, 26.0%, 28.3% and 30.4%, respectively. Decreased percentage of copper concentrations in 0.68 mg·L-1 treatment were 2.9%, 7.4%, 23.5%, 25.0% and 27.9%, respectively. In conclusion, copper played an important role in the success of culture of morulae and blastocysts, and there had adscendent demand for copper utilized by zygotes after 48 h of in vitro zygotes culture.
