川牛膝多糖的提取及含量测定

摘    要:目的]为开发利用川牛膝多糖提供依据。[方法]以川牛膝为材料,采用传统水提醇沉法提取其中的多糖,通过L9(3.4)正交试验对提取工艺进行优化,并考察提取温度、提取时间、料液比、乙醇浓度对多糖得率和提取液中蛋白质含量的影响。[结果]各因素对川牛膝多糖得率的影响由大到小依次为:乙醇浓度〉提取温度〉提取时间〉料液比,对多糖提取液中蛋白质含量的影响由大到小依次为:提取温度〉料液比〉乙醇浓度〉提取时间;川牛膝多糖的优化提取工艺为:提取温度100 ℃,提取时间8 h,料液比1∶20,乙醇浓度95%。[结论]在优化提取工艺条件下,提取液中川牛膝多糖的含量高达68.8 μg/μl。

Extraction and Content Measurement of Polysaccharide in Radix cyathulae

Abstract:Objective] The study was to provide the basis for the exploration and utilization of polysaccharide in Radix cyathulae.Method] With R.cyathulae as the material,the polysaccharide in it was extracted with traditional water extraction and alcohol precipitation method.The extraction process was optimized through orthogonal test and the effects of extraction temperature,extraction time,solid-liquid ratio,ethanol concn.on polysaccharide yield rate and protein content in extracts were investigated.Result] The effects of 4 factors on yield rate of polysaccharide from R.cyathulae from big to small in order was ethanol concn.>extraction temperature > extraction time> solid-liquid ratio and the effects of 4 factors on protein content in extracts in order was extraction temperature>solid-liquid ratio>ethanol concn.>extraction time.The optimized extraction process of polysaccharide from R.cyathulae was that extraction temperature of 100 ℃,extraction time of 8 h,solid-liquid ratio of 1∶ 20 and ethanol concn.of 95%.Conclusion] With the optimized extraction process,the polysaccharide content in extracts of R.cyathulae reached 68.8 μg/μl.
