用不同剂量60Co-γ射线对益母草、蒲公英、车前、紫苏种子进行辐照处理,测定这4种药用植物种子的发芽率、发芽势、发芽指数等指标,了解60Co-γ射线辐照对其种子萌发的影响,为这4种药用植物的新品种选育提供参考。结果表明,60Co-γ射线对4种药用植物种子具有较强的诱发突变能力,辐照剂量与种子发芽率、发芽势、发芽指数、活力指数、苗高、苗鲜重均呈负相关,其中与苗高的相关性达极显著水平。益母草和蒲公英种子的半致死剂量大于400 Gy,车前种子的半致死剂量在300~400 Gy之间,紫苏种子的半致死剂量在50~100 Gy之间。  相似文献

测定并比较了60Co辐射诱变姬松茸突变株J3与原菌株J1子实体不同部位的氨基酸含量.结果表明,除菌柄和盖皮外,姬松茸突变株J3子实体其它部位的必需氨基酸、儿童氨基酸、硫氨酸、支链氨基酸、鲜味氨基酸、芳香族氨基酸和甜味氨基酸均高于原菌株J1.姬松茸突变株J3子实体中谷氨酸含量占氨基酸总量的比例分别比原菌株J1、α-酪蛋白和卵清蛋白提高34.88%、56.68%和122.32%.  相似文献

AIM:To investigate whether the bcl-2 antisense oligonucleotide increases the sensitivity of HL60 and K562 cell lines to daunorubicin.METHODS:IC50 for HL60 and K562 was determined with MTT method, the expression levels of Bcl-2 protein were assayed by immunofluorescence using fluoresce isothiocyanate labeling. In addition, apoptosis was detected by morphological observation and flow cytometric analysis of DNA fragmentation.RESULTS:It was found that the two oligonucleotides directed against the coding region and the translation initiation of bcl-2 mRNA, combined respectively with daunorubicin, inhibited expression of bcl-2 protein, increased apoptosis in HL60 and K562 cells, and decreased IC50 of daunorubicin significantly (P<0.05). Compared to the antisense oligonucleotide directed against the translation initiation of bcl-2 mRNA, the antisense oligonucleotide directed against the coding region showed stronger effects in the aspects of increasing the sensitivity of HL60 cells to daunorubicin (P<0.05).CONCLUSIONS:These two antisense sequences in the translation initiation and the coding region of bcl-2 mRNA increased the sensitivity of HL60 and K562 cell lines to daunorubicin in a sequence-specific manner.  相似文献

AIM: To study the signal transduction pathway of apoptosis initiation induced by homoharringtonine in HL-60 cells. METHODS: After establishing the model of apoptosis initiation induced by homoharringtonine in HL-60 cells, at the point of apoptosis initiation, molecular caspase-3, Bcl-2, Bax and Fas/FasL were measured with flow cytometry and transmission electron microscope. ERK2 and P38 expression in HL-60 cells were detected by using immunohistochemistry. RESULTS: The model of apoptosis initiation induced by homoharringtonine was established in HL-60 cells. At the point of apoptosis initiation, upregulation of caspase-3 and decrease in Bcl-2/Bax were observed. However, the expression of Fas/FasL did not significantly change. ERK2 expression decreased and P38 expression increased. CONCLUSIONS: Caspase-3, Bcl-2, Bax and mitogen activated protein kinase pathways were involved in signal transduction of apoptosis initiation induced by homoharringtonine in HL-60 cells.  相似文献

为探究辐射对睡莲的生物学效应,对2个睡莲品种弗吉尼亚(Nymphaea Virginia)和奥毛斯特(N.Almost black)的块茎进行不同剂量的60Co-γ射线辐射处理。结果表明,2个睡莲品种的存活率均随辐射剂量的增加而下降,半致死剂量分别为24.342和27.671 Gy。5~10 Gy 剂量60Co-γ辐射处理使睡莲叶面积和浮叶数显著增加,20~40 Gy剂量60Co-γ辐射处理使叶面积和浮叶数显著减少。2个睡莲品种的开花时间均随辐射剂量的增加而延迟,但整个花期长度无显著变化。10~40 Gy剂量60Co-γ辐射处理显著降低了睡莲花径,但对睡莲开花率无显著影响。辐射处理导致2个睡莲品种的叶片均出现红棕色斑块、锯齿、皱缩、小孔、卷曲、黄化等变异,且红棕色斑块的面积随着辐射剂量的增加而增大。辐射处理使黑红色花品种奥毛斯特出现普遍的褪色现象,而白色花品种弗吉尼亚的花色未发生变异。此外,经60Co-γ射线辐射处理后2个睡莲品种均出现了花型变异的现象。本研究结果为利用辐射诱变技术选育睡莲新品种奠定了基础。  相似文献

为评价60Co-γ射线和电子束辐照灭菌对葛根提取物品质的影响效应及其差异性,采用不同剂量的60Co-γ射线(0、2.8、5.7、8.3、10.6 kGy)和电子束(0、2.4、5.1、7.7、10.3 kGy)辐照处理葛根提取物,研究辐照对其微生物总数、抗氧化活性及指纹图谱的影响。结果表明,60Co-γ射线和电子束辐照均能有效降低葛根提取物中需氧菌总数、霉菌和酵母菌总数,随着吸收剂量增大,抑制作用增强。60Co-γ射线和电子束辐照吸收剂量分别为5.7 kGy和5.1 kGy时,葛根提取物均达到《中国药典》所规定的微生物限度标准(需氧菌总数为103 CFU·g-1、霉菌和酵母菌总数为102 CFU·g-1);吸收剂量分别为5.7 kGy和7.7 kGy时,需氧菌总数、霉菌和酵母菌总数均降至10 CFU·g-1以下。此外,大肠埃希氏菌(Escherichia coli)与沙门氏菌(Salmonella)均未检出。2种辐照方式均对葛根提取物的DPPH自由基(DPPH·)清除率、羟自由基(·OH)清除率以及总还原能力无显著影响(P>0.05)。不同剂量60Co-γ射线和电子束辐照处理葛根提取物特征峰的相似度达到1.0,且相对保留时间的相对标准差(RSD)均小于0.5%,表明2种辐照方式对葛根提取物指纹图谱均无显著影响(P>0.05)。综上所述,60Co-γ射线和电子束辐照对葛根提取物微生物具有较强的抑制作用,2种辐照方式均不会对葛根提取物的抗氧化活性及指纹图谱产生显著影响,不影响葛根提取物的有效性及质量稳定性。该研究为2种辐照技术,特别是电子束辐照在葛根药材、饮片及其中成药制剂灭菌中的合理应用提供了科学参考。  相似文献

为探究60Co-γ射线辐射处理种子对大花紫薇生长发育中光合效率的影响,以大花紫薇种子为试验材料,以不同60Co-γ射线辐射剂量(0、20、60、100、140、180 Gy)进行辐射处理,测量不同处理生长发育过程中叶绿素及光合指标的变化,研究其对紫薇光合系统的影响。结果表明:20、60 Gy处理较对照和其他处理叶绿素质量分数更高,更有利于叶片进行光合作用;60、100 Gy处理较对照和其他处理潜在光合速率和最大光合效率更好;各时期光合性能指数与综合性能指数均呈现先上升后下降的趋势,但随时间的增加整体呈现下降的趋势,60 Gy较其他处理性能指数高。由此得出,60 Gy辐射处理有利于大花紫薇光合色素的产生与积累,较好的增加电子传递效率,有利于光合系统的运转和能量的积累。  相似文献

Hyperthermia is a form of a cancer treatment which is frequently applied in combination with radiotherapy (RT) to improve therapy responses and radiosensitivity. The mode of action of hyperthermia is multifactorial; the one hand by altering the amount of the blood circulation in the treated tissue, on the other hand by modulating molecular pathways involved in cell survival processes and immunogenic interactions. One of the most dominant proteins induced by hyperthermia is the major stress-inducible heat shock protein 70 (Hsp70). Hsp70 can be found in the blood either as a free-protein (free HSP70) derived from necrotic cells, or lipid-bound (liposomal Hsp70) when it is actively released in extracellular vesicles (EVs) by living cells. The aim of the study was to evaluate the levels of free and liposomal Hsp70 before and after treatment with RT alone or hyperthermia combined with radiotherapy (HTRT) in dogs and cats to evaluate therapy responses. Peripheral blood was collected from feline and canine patients before and at 2, 4, 6 and 24 h after treatment with RT or HTRT. Hsp70 enzyme-linked immunosorbent assays (ELISAs) were performed to determine the free and liposomal Hsp70 concentrations in the serum. The levels were analysed after the first fraction of radiation to study immediate effects and after all applied fractions to study cumulative effects. The levels of free and liposomal Hsp70 levels in the circulation were not affected by the first singular treatment and cumulative effects of RT in cats however, after finalizing all treatment cycles with HTRT free and liposomal Hsp70 levels significantly increased. In dogs, HTRT, but not treatment with RT alone, significantly affected liposomal Hsp70 levels during the first fraction. Free Hsp70 levels were significantly increased after RT, but not HTRT, during the first fraction in dogs. In dogs, on the other hand, RT alone resulted in a significant increase in liposomal Hsp70, but HTRT did not significantly affect the liposomal Hsp70 when cumulative effects were analysed. Free Hsp70 was significantly induced in dogs after both, RT and HTRT when cumulative effects were analysed. RT and HTRT treatments differentially affect the levels of free and liposomal Hsp70 in dogs and cats. Both forms of Hsp70 could potentially be further investigated as potential liquid biopsy markers to study responses to RT and HTRT treatment in companion animals.  相似文献
