Immunocytochemical localization of the Bacillus sphaericus binary toxin components in Culex quinquefasciatus (Diptera: Culicidae) larvae midgut

This study describes the immunocytochemical localization of the Bacillus sphaericus 2362 binary toxin components, BinA and BinB, in Culex quinquefasciatus larvae that had been intoxicated with this entomopathogen. Ultrathin sections of C. quinquefasciatus larvae midgut embedded in the hydrophilic resin L.R. White were incubated with the antibodies anti-BinA or anti-BinB and then revealed with goat anti-rabbit IgG coupled to gold particles. Immunocytochemical detection demonstrated the presence of specific labeling in ultra-thin sections that had been incubated with the BinA antiserum. Gold particles were detected on the apical areas of cell membranes and inside the epithelial cell cytoplasm, particularly the mitochondria, of cells from the gastric caeca and posterior stomach in larvae exposed during 2 or 24 h to the entomopathogen. A similar labeling pattern was observed in ultrathin sections from both midgut regions when incubated with BinB antiserum.     

稻麦两熟制条件下钵苗机插方式对不同类型水稻品种米质的影响

为探明钵苗机插方式在稻麦两熟地区对水稻米质性状的影响及其品种类型间差异,本文以常规粳稻、杂交籼稻和籼粳交水稻为材料,比较了钵苗机插、毯苗机插和机械直播3种种植方式在稻麦两熟制高产栽培条件下的稻米加工品质、外观品质、营养品质、蒸煮食味品质特性变化及其品种类型间差异。结果表明,钵苗机插的整精米率比毯苗机插和机械直播分别提高1.6%和4.9%(P0.05),整精米产量分别提高7.8%和25.9%(P0.05),改善了稻米加工品质;与毯苗机插和机械直播相比,钵苗机插的稻米垩白米率、垩白面积和垩白度显著增加,对稻米外观品质有不利影响,但钵苗机插精米蛋白质含量较高,有利于稻米营养品质的改良,其精米蛋白质产量分别比毯苗机插和机械直播提高8.7%和28.0%;3种种植方式相比,钵苗机插稻米蒸煮食味品质表现出一定程度的变优趋势,其中,稻米直链淀粉含量、消减值和糊化温度呈钵苗机插毯苗机插机械直播的趋势,胶稠度、峰值黏度、热浆黏度、最终黏度和崩解值呈钵苗机插毯苗机插机械直播的趋势;食味值于种植方式间无显著差异,呈籼粳交水稻和常规粳稻钵苗机插毯苗机插机械直播,杂交籼稻毯苗机插钵苗机插机械直播的趋势。由此得出,钵苗机插不仅能增加稻谷产量,还能改善部分稻米品质特征,是稻麦两熟地区实现高产高效优质协调水稻生产的潜力型机械化种植方式。

弹性轴-轴承系统动力学行为研究

以弹性轴的多柔体动力学分析为基础,通过ADAMS与Matlab联合仿真,研究了弹性轴-轴承系统在径向正弦载荷作用下的动力学特性.研究表明,在所研究的频率范围内,弹性轴-轴承系统存在两阶自然频率.其一阶频率与轴承的动力学特性有关,而二阶频率则取决于弹性轴的动力学特性.在二阶频率的正弦载荷作用下,弹性轴-轴承系统的最大油膜压力远大于刚性轴-轴承系统,而其最小油膜厚度则远小于刚性轴-轴承系统.     

基于委托培养的大学生村官选聘模式研究

大学生村官政策的实施有效推进了社会主义新农村建设中基层干部队伍建设。但是，目前的选聘机制较为单一，主要以统一考录为主，导致了部分大学生村官乡村工作中技能不适用、情感待融合等现象。委托培养是一种有效的补充选聘方式，可以在农村当年高考生中择优挑选．由县级主管部门与其本人和培养学校签订委托培养协议，学成后派回本村担任大学生村官。     

The carboxyterminal processing protease of D1 protein: Herbicidal activity of novel inhibitors of the recombinant and native spinach enzymes

The carboxyterminal processing protease of D1 protein (CtpA) is predicted to be an excellent target for the discovery of a general broad-spectrum herbicide. Directed and random screening of compounds against recombinant spinach CtpA (rCtpA) has led to the discovery of five different chemical classes of inhibitors. Lead compounds from each inhibitor class were investigated for their in vitro effects on the activities of both recombinant and native spinach CtpA. All of the lead compounds have K_i values of less than 50 μM when tested against rCtpA, and all except one showed competitive inhibition. Results from partially purified native CtpA from spinach were similar to those from the recombinant form of the enzyme, thus validating the use of rCtpA in the inhibitor screen. Compounds from three of the classes of CtpA inhibitors show in vivo herbicidal activity against Arabidopsis thaliana when applied either by addition to growth media or by spraying the leaves. Transgenic Arabidopsis plants which over-express CtpA showed greater resistance to the compounds than wild-type plants providing evidence that these inhibitors are directly acting against CtpA.

现阶段,我国农村信用合作社是整个农村金融体系中最重要组成部分之一。新中国成立至今,特别是经过改革开放以来多年的发展,农村信用合作社已成为我国广大农村地区最庞大的金融组织,但其经营模式有待进一步的完善,服务功能尚需加强。所以,借鉴国外农村合作金融的成功模式,对促进我国农村合作金融的发展具有重要的理论和现实意义。
