The purpose of this study was to evaluate oxidative stress and neurotoxic potential of organophosphorus (OP) insecticide diazinon in the sentinel freshwater fish, Oreochromis niloticus. Antioxidant and acetylcholinesterase (AChE) enzyme activities and malondialdehyde (MDA) and protein levels were measured spectrophotometrically in gill, kidney, alimentary tract, and muscle tissues of fish treated with sub-lethal diazinon concentrations for 1, 7, 15, and 30 days. Dose-dependent inhibitions of AChE were observed in all the experimental fish. On the contrary of alimentary tract, MDA levels were elevated in kidney and muscle and gill was not affected. AChE and MDA levels intercorrelated in kidney and muscle tissues. Diazinon had increased superoxide dismutase (SOD) activities in all the tissues, while kidney was the most affected tissue. Tissue-specific alterations were observed on catalase (CAT) and glutathione peroxidase (GPx) activities; however, the activities were not changed in gill and muscle tissues for GPx and in gill, muscle, and kidney tissues for CAT. Protein levels decreased in kidney, muscle, and alimentary tract, while increased in gill and alimentary tract in 15 days. With respect to these results, diazinon has oxidative and neurotoxic potentials in O. niloticus. Observed changes with diazinon treatment were generally tissue-specific and dose-dependent.  相似文献

Ecotourism is a rapidly growing industry with unknown impacts on viewed wildlife that may require novel management action. We examined the impact of viewing activities on the behaviour of brown bears (Ursus arctos) in coastal British Columbia.Domination of the best feeding sites and human avoidance by large male bears has consistently been reported. We, however, saw displacement in time rather than space - during the viewing day large males were less active than at other times, while females with cubs tended to be more active.In each year, females with cubs spent similarly high proportions of their time fishing when people were present. In years with large male activity, less time was spent fishing when people were absent. When freed from the potential threat of large male bears, females with cubs showed no measurable impact of controlled human activity.Human presence at a feeding site impacts the behaviour of brown bears, but not as expected. Temporal avoidance of human activity by large males was observed; indications that they departed upon satiation, before the arrival of morning tours, however, suggests that there was little energetic impact. By displacing large males, viewing activities created a temporal refuge, enhancing feeding opportunities for subordinate age/sex classes. With the strong positive relationships between mean female mass and litter size, this may in turn increase population productivity.  相似文献

A culture system was developed for the in vitro inoculation of bovine splenic lymphocytes with sheep RBC and trinitrophenylated proteins. The addition of pokeweed mitogen to the cultures was necessary for the induction of plaque-forming cells to the antigen. Several studies have evaluated the spatial distribution of cool- and warm-season grasses across different topographic positions in the Nebraska Sandhills, but limited research has explored topographic differences in total plant production or production of plant functional groups in relation to variable amounts of precipitation. This study evaluated how spring and growing season precipitation influenced plant production at four topographic positions common in the eastern Nebraska Sandhills. Plant production data were collected from annually moved grazing exclosures in mid-June (peak cool-season grass production) and mid-August (peak warm-season grass production) during a 17-yr period from 2001 to 2017. Total plant production and precipitation use efficiency were 35 ? 58% greater on interdune positions, and precipitation marginal response for total plant production was more sensitive to increases in spring and growing season precipitation on interdune compared with dune positions in both mid-June and mid-August. The greater precipitation marginal response of total plant production on interdune positions was driven primarily by greater increases in cool-season grass production with increasing spring or growing season precipitation. Warm-season grass precipitation marginal response was not different among the topographic positions, but production was 23 ? 70% greater on interdune compared with dune topographic positions in mid-August. When differences in the amount of each topographic position at the study location were accounted for, growing season precipitation explained 49% of the variation for total plant production in mid-August, but spring precipitation only explained 23% of the variation for total plant production in mid-June. Because of the differential response of plant production at dune and interdune positions, incorporating the relative amount of each topographic position into estimates of plant production at the pasture or ranch scale will provide better information for adjusting stocking rates to more accurately match animal demand with forage availability.  相似文献

A full-length cDNA of a glutathione S-transferase (GST) was cloned from a cDNA library of the local Egyptian cattle tick Boophilus annulatus. The 672 bp cloned fragment was sequenced and showed an open reading frame encoding a protein of 223 amino acids. Comparison of the deduced amino acid sequence with GSTs from other species revealed that the sequence is closely related to the mammalian mu-class GST. The cloned gene was expressed in E. coli under T7 promotor of pET-30b vector, and purified under native conditions. The purified enzyme appeared as a single band on 12% SDS-PAGE and has a molecular weight of 30.8 kDa including the histidine tag of the vector. The purified enzyme was assayed upon the chromogenic substrate 1-chloro-2,4-dinitrobenzene (CDNB) and the recombinant enzyme showed high level of activity even in the presence of the beta-galactosidase region on its 5' end and showed maximum activity at pH 7.5. The Km values for CDNB and GSH were 0.57 and 0.79 mM, respectively. The over expressed rBaGST showed high activity toward CDNB (121 units/mg protein) and less toward DCNB (29.3 units/mg protein). rBaGST exhibited peroxidatic activity on cumene hydroperoxide sharing this property with GSTs belonging to the GST alpha class. I50 values for cibacron blue and bromosulfophthalein were 0.22 and 8.45 microM, respectively, sharing this property with the mammalian GSTmu class. Immunoblotting revealed the presence of the GST molecule in B. annulatus protein extracts; whole tick, larvae, gut, salivary gland and ovary. Homologues to the GSTmu were also detected in other tick species as Hyalomma dromedarii and Rhipicephalus sp. while in Ornithodoros moubata, GSTmu homologue could not be detected.  相似文献

The objective of this study was to determine the cost to female brown bears with cubs of avoiding risk, or perceived risk, from both large adult male bears and ecotourists. We measured salmon consumption by brown bears in British Columbia under different risk scenarios (the presence and absence of large male bears and people). Declines occurred despite superabundant food where consumption was never limited by salmon abundance.While there were significant changes in the proportion of time spent foraging and in the consumption of captured fish between periods with and without ecotourists present, there was no change in foraging effectiveness, catch per unit effort.Selection of sub-optimal habitats to reduce predation risk incurs energy costs. Risk avoidance reduced daily salmon consumption by females with cubs by more than one third. By associating with people, females with cubs were able to avoid encounters with large, potentially aggressive males, which avoided people in both time and space. Controlled human activity at feeding sites can provide a temporal feeding refuge for vulnerable age/sex classes. This is the first study to quantify the energetic cost of intra-specific risk-avoidance for a large carnivore.  相似文献

Over a 19‐year period, 51 horses showing chronic lameness or gait abnormality that was not fully responsive to veterinary treatment at other referral clinics were referred to us for further treatment. All had either failed to have a diagnosis made, or treatment for the diagnosed conditions had shown only partial response. After further examination, we concluded that they showed abnormal function of the neck or back, but there was no obvious pathological cause. A diagnosis of ‘somatic dysfunction’ was made, characterised by altered muscle tone, tenderness or subtle changes in gait, similar to the condition recognised in human medicine. These cases subsequently underwent osteopathic treatment under sedation. Forty‐six cases (90.2%) responded to treatment in the short term (6 months after treatment). Seventeen (53.1%) were working at the same level or better than previously at least a year after treatment. Ten (31.2%) worked at a reduced level for between 1.5 and 10 years. Nineteen cases were lost to long‐term follow‐up. The findings of this study suggest that neck and back problems may be overlooked when investigating chronic lameness. When no pathology can be pinpointed as a cause of lameness, a diagnosis of somatic dysfunction as a primary problem should be considered. The presence of somatic dysfunction as a secondary complicating factor should not be overlooked. Stiffness and/or low grade pain in the spine of horses can result in persistent abnormal posture, gait, or behaviour, similar to ‘somatic dysfunction’ seen in the human patient. The cause is neuromuscular in origin, rather than resulting from tissue pathology. The diagnosis made clinically can be confirmed with thermography. This study confirms that osteopathic manipulation of the spine of horses can be a valuable treatment for cases of lameness that do not respond fully to standard veterinary treatments.  相似文献
