The aim of the present study was to evaluate the expression and localization of lymphangiogenic factors (VEGF-C and VEGF-D), their receptor (VEGFR3) and lymphatic endothelial marker (LYVE1) in buffalo placenta during early pregnancy [EP], and to investigate the functional role of lymphangiogenic growth factors in placental lymphangiogenesis. The mRNA and protein expression of VEGF-C, VEGF-D, their receptor VEGFR3 and LYVE1 showed significant expression in EP1 (29–42 days) and EP2 stages (51–82 days) both in caruncle (maternal part) and cotyledon (foetal part) of the buffalo placenta. Immunoreactivity of VEGF-C, VEGF-D and LYVE1 was observed around the endometrial gland, in lymphatics and trophoblast cells, whereas VEGFR3 mainly localized in lymphatics of the caruncle and cotyledons. Cultured trophoblast cells were treated with VEGF-C/VEGF-D (50, 100 and 150 ng/ml) and combined doses of VEGF-C and VEGF-D (150 ng/ml) each for different time durations (24, 48 and 72 h). The mRNA expression of LYVE1 and PCNA was significantly (p < .001) upregulated with VEGF-C and VEGF-D and combined treatment (@150 ng/ml), as well as significantly downregulating Caspase-3 at 48 and 72 h. Thus, the present study provides evidence that lymphangiogenic factors are expressed in buffalo placental compartments and they may play a significant role in the regulation of placental function in water buffaloes.  相似文献

In vegetable brassicas, cytoplasmic male sterility (CMS) system is the most preferred mechanism for hybrid seed production and Ogura cybrid cytoplasm is the only source used widely. Effects of the alien cybrid cytoplasm on important quality traits need to be understood for their effective use in breeding programme. In analysing 38 Ogura cybrid cytoplasm‐based cauliflower CMS lines, it was revealed that introgression of Ogura cytoplasm resulted in significant alteration in important quality traits. Cupric reducing antioxidant activity and ferric reducing ability of plasma values were increased up to 5–10 times in different genotypes. Among 38 analysed CMS lines, concentration of plant pigments such as anthocyanin, total chlorophylls and ascorbic acid was reduced in 21, 17 and 32 genotypes, respectively. However, the concentration of total carotenoids and β‐carotene was elevated in most of the CMS lines and total carotenoid was increased up to 20 times in the CMS line, Ogu13‐85‐2A. The results indicate the role of nuclear–cytoplasmic interaction and mitochondrial genome in determining concentration of different quality traits.  相似文献

The problem of real-time irrigation scheduling under limited water supply is considered. The goal is to develop an irrigation operation policy which maximizes crop yields and is responsive to current season changes in weather and other variables. Because irrigation decisions are sequential and dependent on crop and soil water status, and also because crop yields can only be known at the end of the season, the decisions are arrived at by a two-stage process. In the first or the design stage, irrigations are planned for the entire season at weekly intervals using historical data and an optimal irrigation scheduling model. In the second stage, the decisions for the subsequent weeks are revised each week after updating the status of the system with real time data up to that week and solving the irrigation optimization model once again for the new conditions. Thus, each week an irrigation decision is made, the entire planning horizon is kept in view. The procedure is illustrated by application to a case study.  相似文献

The tetracycline resistance gene, tet(B), has been described previously in Gram negative bacteria. Among cereals, only maize has not only a high amount of carotenoids, tocopherols, and oil content but also is rich in starch and protein content compared with other major food crops, such as rice and wheat. The present investigation was made primarily to assess the genetic variability for nutritionally important traits in 87 elite maize inbreds representing major heterotic groups in China. Carotenoid and tocopherol fractions were measured by high-performance liquid chromatography (HPLC), whereas oil, starch, and protein contents were detected by a VECTER22/N near-infrared analyzer. Significant interactions between genotypes and years were observed for all the traits. The pooled mean values of beta-carotene, beta-cryptoxanthin, alpha-carotene, lutein, zeaxanthin, and total carotenoids were 0.449, 0.876, 0.121, 5.803, 3.048, and 10.298 microg g (-1), respectively, whereas the combined mean performance of alpha-tocopherol, gamma-tocopherol, delta-tocopherol, and total tocopherols were 23.98, 32.90, 2.189, and 59.55 microg g (-1), respectively. The average protein, starch, and oil contents were observed to be 12.28, 64.51, and 3.55%, respectively. High level of heritability estimates were observed for all the traits and ranged from 65.6% (protein content) to 92.5% (alpha/gamma-tocopherol ratio). Most of the traits studied in this experiment were either significantly positive correlated or independent. The present finding exhibits substantial opportunities to the breeders for improvement of these traits in maize cultivars and also suggests further exploration of a new source of elite breeding stocks containing a high level of these nutritionally important compounds. Finally, these findings may also help in biofortification of maize.  相似文献

Apple scar skin viroid (ASSVd) infection is a major limitation to apple fruit quality and causes huge economic losses. In surveys of apple orchards in the northern Indian state of Himachal Pradesh, fruits with dappling symptoms were noticed. ASSVd was detected from these fruits and molecularly characterized. Ten clones from three isolates were sequenced, of which seven were new sequence variants of ASSVd. The clones had significant sequence variability (94–100%) with each other. Variability was more common in the pathogenic domain of the viroid genome. Four of the clones were 330 nucleotides (nt) long, and the other six had an additional nucleotide. Phylogenetic analysis showed close affinity of the present isolates with some Chinese and Korean isolates. The study reports seven new variants of ASSVd and also provides the first molecular evidence of viroid infection (ASSVd) in apple in India.  相似文献

Black gram (Vigna mungo L.) var. Shyam plants were raised in refined sand at four levels of molybdenum (Mo), i.e., low (0.002 µM) to excess (2 µM) for 70 days. The molybdenum deficiency symptoms appeared as interveinal chlorosis of young and middle leaves. Compared to control (0.2 µM Mo), total dry matter, seed yield and seed protein decreased at low and excess Mo. The concentration of Mo in leaves and seed as well as activity of nitrate reductase increased with an increase in Mo supply. Low and excess Mo deteriorated the quality of seeds by lowering the content of starch, sugars, protein, and nitrogen and increasing electrical conductivity of seed leachate. Molybdenum deficiency and excess both resulted in production of lightweight immature seeds, poor in vigor and germination potential. The values of sufficiency and threshold of toxicity in leaves were 0.078 and 2.15 µg Mo g?1 dry matter of black gram.  相似文献

Comparative studies of two cultivated and sixteen wild species of the genus Oryza were carried out using one- and two-dimensional gel electrophoresis for variation in their seed proteins for interrelationships of these species. A number of polypeptides in the range of molecular weight 13–110 kDa were seen to vary. Under reducing conditions, polypeptides spread over the regions of mol. wt. 33–40.5, 25–27 and 19–21.5 kDa exhibited maximum variation in their patterns. Two-dimensional gel electrophoresis revealed the occurrence of disulphide-linked glutelin polypeptide pairs of mol. wt. 60, 58, 52 and 25 kDa breaking into a large and a small subunit each in the range of mol. wt. 18–40.5 and 16–25 kDa respectively in Oryza sativa. The number of such polypeptide pairs varied from 2 to 6 in different species and also in O. sativa showed variation in mol. wts. of their constituent subunits. The UPGMA dendrogram revealed that most of the Oryza species occurred in different clusters and subclusters and thus did not share very close relationships. The undisputed and closest relationship observed was that of cultivated rice O. sativa with the O. rufipogon followed by that with O. nivara. The African cultivated O. glaberrima occurring on the nearest branch of the same subcluster, thereby, supporting the phylogenetic of these species suggested in earlier studies. Eight diploid species and seven tetraploid species were included in one part of the dendrogram while the remaining two species with AA genome i.e. O. glumaepatula and O. meridionalis and one with FF i.e. O. brachyantha stood separately from these as scattered in the group of seven tetraploid species with BBCC, CCDD and HHJJ genomes. The tetraploids O. alta, O. latifolia and O. grandiglumis with CCDD genomes which occurred on the farthest part were distantly related with O. sativa. The cyanogen bromide peptide maps and two dimensional gel electrophoresis also supported the closest relationship between O. sativa and O. rufipogon.  相似文献
