The release of tumor necrosis factor-alpha (TNF-) from cultured bovine alveolar macrophages (BAM) was evaluated following stimulation of BAM with bovine herpesvirus-1 (BHV-1), parainfluenza-3 (PI-3) virus, bovine respiratory syncytial virus (BRSV), Escherichia coli 0111:B4 endotoxin, Pasteurella haemolytica type 1 endotoxin, Pasteurella multocida endotoxin, and virus/endotoxin combinations. A cytotoxic assay system using Georgia bovine kidney cells as targets was used to measure TNF- activity. The cytotoxic activity was neutralized by an anti-human TNF- monoclonal antibody.

Stimulation of BAM with 1 median tissue culture infectious dose (TCID50) of live or ultraviolet (UV)-inactivated PI-3 virus/cell resulted in release of TNF- in significantly (P<0.05) higher amounts than sham-induced BAM. The quantities of TNF- released after live or UV-inactivated BHV-1 or BRSV induction were not significantly higher than sham-induced BAM. E. coli 0111:B4, P. haemolytica type 1 and P. multocida endotoxins stimulated TNF- release in a dose-dependent manner. Sequential exposure of BAM to 1 TCID50 per cell of either live BHV-1, PI-3 virus or BRSV and then 5 μg ml−1 of either E. coli 0111:B4, P. haemolytica type 1 or P. multocida endotoxin caused a significant (P<0.05) reduction in detectable TNF- in seven of nine virus/endotoxin combinations tested, when compared with 5 μg ml−1 of endotoxin alone. Parainfluenza-3 virus/endotoxin combinations stimulated higher TNF- release when compared with other virus/endotoxin combinations. Five out of six test animals had serum-neutralizing antibodies to PI-3 virus, one out of six had serum-neutralizing antibodies to BHV-1, and two out of six had serum-neutralizing antibodies to BRSV, suggesting a possible relationship between serum neutralizing antibodies and TNF- release from in vitro cultivated BAM.  相似文献

An old dog had an expansile lesion affecting the ulnar diaphysis. The lesion had clinical and radiographic features typical of a bone cyst; however, computed X-ray tomography indicated that the lesion had a tissue content incompatible with a true cyst. The histological diagnosis was osteosarcoma. This report emphasises the highly variable radiological appearance of canine osteosarcoma; biopsy is required to establish the diagnosis because the radiological signs may mimic a lesion of different aetiology.  相似文献

Totally 13575 ewes of two different breeds, Dala and Spel, were inseminated with semen, frozen in straws and thawed at 70°C for 8 sec. An insemination dose of 0.2 ml containing approx. 150 × 106 spermatozoa with at least 45 to 50% progressive motility was imerted 5 to 12 mm into the cervix. The insemination was performed once between 12 and 30 h after the onset of heat. The NR rate of the Dala ewes increased significantly during the season. The NR rate of the ewes inseminated before 15. November was 44.3%, from 15. to 20. November 52.2%. from 20. to 25. November 55.3% and from 25. November and later 61.4%. The corresponding values for the ewes of the Spel breed were 57.3, 58.7, 61.5 and 71.0% respectively, and only the difference between the two last values was statistically significant. The difference between the fertility of the two breeds was significant within each of the periods .  相似文献

Adult populations ofMaladera matrida Argaman (Coleoptera: Scarabaeidae), a beetle highly injurious to agricultural crops, were studied in the coastal plain of Israel from 1985 to 1988. There were two generations annually. Adults emerged in March, April or May and disappeared in October or November. The beetles are on the wing, mate, and feed at twilight and in the dark. Feeding and mating behavior, sex ratio, and preferred food plants were studied.  相似文献

In this two year study we investigated the effect that the level of concentrate in feedlot diet had on the occurrence of health problems in bull calves belonging to two multibreed groups.

In each of the two years, the 168 day feedlot period was divided into two equal periods of 77 days with an intervening 14 day adjustment period. During the first period, the animals in half of the pens within each breed-group received a high concentrate (85% grain) diet (H) while the rest of the pens received a diet (L) of hay ad libitum and half as much concentrate as the bulls on high concentrate. The L diet contained an average of 48% grain. In the second period, diets of bulls in half of the pens within each breed-group were switched from L to H and vice versa. During the feedlot period, the incidence of sickness among bulls was recorded. Disease incidence was higher in the second year compared to the first. In the second year, 49 out of 56 (87.5%) calves on the high concentrate diet required treatment for respiratory infections compared to only four (7.14%) in bulls on the low concentrate diet. The results suggest that a high concentrate diet for animals in feedlots may be more stressful to calves coming directly to the feedlot following weaning compared to a low concentrate diet, thereby acting as an additional predisposing factor to respiratory infections.

In carnations grown on rockwool disease incidence of fusarium wilt caused byFusarium oxysporum f.sp.dianthi (Fod) was reduced when Fe-EDDHA instead of Fe-DTPA was used as iron source in the nutrient solution. Addition ofPseudomonas sp. strain WSC417r intensified this reduction in the cultivar Pallas, moderately resistant to Fusarium, but not in the susceptible cultivar Lena. Treatment of plants with Fe-EDDHA instead of Fe-DTPA as iron source resulted in higher numbers and percentages on the roots, ofin vitro antagonistic fluorescent pseudomonads. However, differences were only significant at 56 days after planting for cv. Lena and at 14 and 28 days after planting for cv. Palas. Both chelators, at different concentrations, had no effect on root colonization by eitherPseudomonas sp. strain WCS417r orFod strain WCS816. However, when coinoculated, reduced numbers of propagules ofFusarium were found at concentrations of Fe-EDDHA lower than 10–5 M.Higher concentrations of the siderophore fusarine produced byFod strain WCS816 were demonstrated when Fe-EDDHA instead of Fe-DTPA was used as iron source in culture media. At equal concentrations, no such differences were found in the amount of siderophore produced by WCS417r. Germ tube length ofFod was less with Fe-EDDHA than with Fe-DTPA. The reduction of germ tube length was stronger when the purified siderophore of WCS417r was added in excess to the culture media with Fe-EDDHA than those with Fe-DTPA. Therefore, the observed reduction of germ tube growth can not completely be explained by iron deprivation. It appeared that EDDHA exhibited a toxic effect for conidia ofFod strain WCS816 as well.we conclude that the observed disease reduction by Fe-EDDHA is a consequence of a limitation of iron availability forFod. This limitation is possibly intensified by the increase in number or percentage of antagonistic fluorescent pseudomonads that strongly compete for iron. The additional effect after bacterization withPseudomonas strain WCS417r in Fe-EDDHA treated carnations of cv. Pallas is likely to be due, at least partly, to a direct competition for iron between the siderophores ofFod strain WCS816 and ofPesudomonas sp. strain WCS417r.Samenvatting Verwelkingsziekte in anjers op steenwol, veroorzaakt doorFusarium oxysporum f. sp.dianthi (Fod), werd gereduceerd indien het ijzer-chelaat Fe-EDDHA in plaats van Fe-DTPA werd toegevoegd aan de nutriëntenvloeistof. Bacterisatie metPseudomonas sp. stam WCS417r had een additioneel effect bij de matig resistence cultivar Pallas maar niet bij de vatbare cultivar Lena. Toevoeging van Fe-EDDHA in plaats van Fe-DTPA aan planten als ijzerbron resulteerde op de wortels in hogere aantallen en percentages fluorescerende pseudomonaden, diein vitro antagonistisch waren ten opzichte vanFod. De verschillen waren echter alleen significant 56 dagen na planten voor de cultivar Lena en 14 en 28 dagen na planten voor de cultivar Pallas. Beide chelaten vertoonden bij verschillende concentraties geen effect op de kolonisatie van de wortel door beide microorganismen. Echter, wanneer beide micro-organismen gezamelijk werden toegevoegd nam de wortelkolonisatie doorFod stam WCS816 af bij concentraties lager dan 10–5 M Fe-EDDHA. Er werd meer van het siderofoor fusarine doorFod stam WCS816 geproduceerd bij concentraties lager dan 10–4 M Fe indien Fe-EDDHA in plaats van Fe-DTPA als ijzerbron aan het cultuurmedium was toegevoegd. Er werd geen effect van beide chelaten gevonden op de siderofoorproduktie door WCS417r. Indien een overmaat van het gezuiverde siderofoor van WCS417r werd toegevoegd aan Fe-EDDHA werden een sterkere afname van de kiembuislengte gevonden dan toevoeging aan Fe-DTPA. De reductie van de kiembuislengte bleek niet volledig verklaard te kunnen worden door een afname van de ijzerbeschikbaarheid. Het chelaat EDDHA heeft ook een toxisch effect op conidiën van fusarium.Wij concluderen, dat de waargenomen reductie van de verwelkingziekte door Fe-EDDHA een gevolg is van de afname van de ijzerbeschikbaarheid voorFod. Dit wordt waarschijnlijk versterkt door de ontwikkeling van een antagonistische, fluorescerendePseudomonas-populatie die sterk concurreren om ijzer. Het additioneel effect dat door bacterisatie metPseudomonas sp. WCS417r van de met Fe-EDDHA behandelde matig resistante anjers (Pallas) werd verkregen is voor een deel het gevolg van een directe concurrentie om ijzer tussen de sideroforen vanFod stam WCS816 en vanPseudomonas sp. stam WCS417r.  相似文献

To improve genetic gain of breeding programs for village poultry production, breeding schemes with observations obtained in village production systems using individual (VIO) and group recording (VGO) were examined under different levels of genotype-by-environment-interactions (GxE). GxE was modeled by varying the correlation between traits measured in the breeding station and village environments for bodyweight (rg_BW) and egg production (rg_EP). Relative and absolute genetic gains obtained from VIO and VGO were used for comparison between the schemes. Results showed that village observations significantly improved genetic gains compared to the scheme without birds tested in the village. The improvement was only slightly larger with individual observations than with group observations. Higher rg_BW and rg_EP led to lower relative genetic gain, but a higher absolute gain of VIO and VGO. It is recommended to apply a breeding scheme using group recording of village performance when strong GxE in breeding for village poultry is expected.  相似文献

Contemporary genetic diversity is the product of both historical and contemporary forces, such as climatic and geological processes affecting range distribution and continuously moulded by evolutionary forces selection, gene flow and genetic drift. Predatory freshwater fishes, such as Northern Pike Esox lucius, commonly exhibit small population sizes, and several local populations are considered endangered. Pike inhabit diverse habitat types, including lakes, rivers and brackish marine waters, thus spanning from small isolated patches to large open marine systems. However, pike population structure from local to regional scales is relatively poorly described, in spite of its significance to developing conservation measures. We analysed microsatellite variation in a total of 1185 North European pike from 46 samples collected across both local and regional scales, as well as over time, to address two overarching questions: Is pike population structure associated with local and/or regional connectivity patterns, and which factors likely have the main influence on the contemporary distribution of genetic diversity? To answer this, we combined estimators of population diversity and structure to assess evidence of whether populations within (i) habitats, (ii) drainage systems and (iii) geographical regions are closer related than among these ranges, and whether patterns are temporally stable. Contrasting previous predictions that genetic drift obscures signals of postglacial colonisation history, we identified clear regional differences in population genetic signatures, suggesting a major effect of drainage divides on colonisation history and connectivity. However, several populations deviated from the general pattern, showing that local processes may be complex and need to be examined case‐by‐case.  相似文献

OBJECTIVE: The aim of this study was to characterize the clinical and morphologic features of neuronal ceroid lipofuscinosis (NCL) in the Polish Owczarek Nizinny (PON) breed of dog. ANIMALS: Nine Swedish PON dogs of both sexes were included in the study. PROCEDURE: All dogs underwent a detailed clinical evaluation, with emphasis on ophthalmic exams. Histopathology and electron microscopy were performed on the eyes, brain and various internal organs. Immunohistochemical staining for detection of sphingolipid activator proteins (SAPs) and mitochondrial ATP synthase (SCMAS) was performed on the eyes and brain. RESULTS: The dogs showed behavioral abnormalities, motor disturbances and visual impairment or blindness. Pupillary responses were abnormal while fundus changes varied from normal to severe retinal atrophy. Electroretinography (ERG) showed variable changes, from slight alterations in the process of dark adaptation to severely reduced or nonrecordable ERG a- and b-wave amplitudes. Histopathology revealed intracytoplasmic storage bodies within neurons of the brain and in retinal cells, especially the retinal pigment epithelium (RPE). Round to oval granular type of inclusion bodies, known as granular osmiophilic dense deposits (GRODS), were found in neuronal cells in the brain and in the retina. Immunohistochemistry identified the storage material in the brain and retina as consisting of SAPs. CONCLUSION: The presently described NCL disease in PON dogs shows similarities to previously recorded cases in the Miniature Schnauzer. The closest human equivalent to this disease is infantile NCL (CLN1), in which the major stored proteins are SAPs and the ultrastructure of the inclusion bodies of neuronal cells is granular.  相似文献
