A total of 318 Escherichia coli isolates obtained from different food-producing animals affected with colibacillosis between 2001 and 2006 were subjected to phylogenetic analysis: 72 bovine isolates, 89 poultry isolates and 157 porcine isolates. Overall, the phylogenetic group A was predominant in isolates from cattle (36/72, 50%) and pigs (101/157, 64.3%) whereas groups A (44/89, 49.4%) and D (40/89, 44.9%) were predominant in isolates from poultry. In addition, group B2 was not found among diseased food-producing animals except for a poultry isolate. Thus, the phylogenetic group distribution of E. coli from diseased animals was different by animal species. Among the 318 isolates, cefazolin resistance (minimum inhibitory concentrations: ≥32 μg/ml) was found in six bovine isolates, 29 poultry isolates and three porcine isolates. Of them, 11 isolates (nine from poultry and two from cattle) produced extended spectrum β-lactamase (ESBL). The two bovine isolates produced blaCTX-M-2, while the nine poultry isolates produced blaCTX-M-25 (4), blaSHV-2 (3), blaCTX-M-15 (1) and blaCTX-M-2 (1). Thus, our results showed that several types of ESBL were identified and three types of β-lactamase (SHV-2, CTX-M-25 and CTX-M-15) were observed for the first time in E. coli from diseased animals in Japan.  相似文献

Purpose

The objective of this study was to determine the changes in the main soil chemical properties including pH, electrical conductivity (EC), available phosphorus (P), soil organic carbon (SOC) and total nitrogen (TN) stocks after long-term (31 years) additions of two types of organic matters—rice straw and rice straw compost, combined with NPK fertilizers in single rice paddy in a cold temperate region of Japan.

Materials and methods

A long-term experiment on combined inorganic fertilizers and organic matters in paddy rice cultivation began in May 1982 in Yamagata, northeastern Japan. After the 31st harvest, soil samples were collected from five treatments [(1) PK, (2) NPK, (3) NPK + 6 Mg ha?1 rice straw (RS), (4) NPK + 10 Mg ha?1 rice straw compost (CM1), and (5) NPK + 30 Mg ha?1 rice straw compost (CM3)] at five soil depths (0–5, 5–10, 10–15, 15–20, and 20–25 cm). Soil chemical properties of pH, EC, available P, SOC, and TN were analyzed.

Results and discussion

The pH decreased significantly only at the higher compost rate of 30 Mg ha?1, while EC increased in all the organic matter treatments. Available P significantly increased in the CM1 and CM3 treatments by 55.1 and 86.4 %. The amounts of SOC stock increased by 67.2, 21.4, and 8.6 %, and soil TN stock by 64.1, 20.2, and 8.5 % in CM3, RS, and CM1, respectively, compared to NPK treatment.

Conclusions

Significant changes in soil properties were observed after 31 years of organic matter applications with reference to PK- and NPK-fertilized rice paddy soils. A significant decrease in pH was observed with the application of a high rate (30 Mg ha?1) of rice straw compost but not with the conventional rate of 10 Mg ha?1. However, EC increased significantly relative to that of the PK- and NPK-fertilized plots in all the organic matter treatments. Available P significantly increased in the CM1 and CM3 treatments by 55.1 and 86.4 %. The amounts of SOC stock expressed as a percentage of total C applied to the soil were higher from 10 Mg ha?1 compost (28.7 %) than that from 6 Mg ha?1 rice straw (17.4 %), indicating a more effective soil organic C accumulation from rice straw compost than that from original rice straw.

Newborn calves received a low dose of bovine interferon-tau (boIFN-tau) orally for 4 weeks and calves that had developed diarrhea received a low dose of boIFN-tau orally for 5 days. No effects of boIFN-tau were seen in the duration of the diarrhea, or in daily weight gain. Calves received a high dose of boIFN-tau subcutaneously 3 times and they were then stimulated with bovine herpesvirus type 1 vaccine. No adverse effects were observed after the administration of boIFN-tau and lymphocyte subsets from calves did not change after the stimulation. Our results suggest that boIFN-tau does not seem protecting for preventing calves from diarrhea, recovering the health of calves with diarrhea or immunomodulation, although the treatment itself is not toxic.  相似文献

The effects of grazing and supplemental protein concentrations, provided during the grazing period, on subsequent finishing performance and carcass quality were investigated. This experiment was carried out using 15 Japanese Black cattle (Wagyu) steers. The steers were fed as follows: (i) grazing supplemented with moderate protein concentrate (GMP) (18% crude protein (CP); dry matter (DM) basis) before finishing, followed by feeding in a barn until slaughter; (ii) grazing supplemented with low protein concentrate (GLP) (14% CP; DM basis) before finishing, followed by feeding in the barn until slaughter; and (iii) no grazing before finishing (NG). From the end of the grazing season to the end of the growing period, the GMP and GLP steers were managed in the same way as the NG steers. All of the animals were fed the same diet in the finishing period (9 months to 28 months). None of the three treatments affected the average daily gains. For all treatments, chilled carcass weights were more than 450 kg and did not differ significantly among the groups. The longissimus muscle areas in the grazed steers were numerically larger than in those receiving the NG treatment. Fat thickness was significantly greater (P < 0.05) in the NG steers than in the grazed steers. The intramuscular fat and marbling scores were not affected in the grazed steers. In conclusion, the carcass quality of grazed steers was similar to that of the NG steers at a similar final age, and the quantity of meat in the NG steers would be less than that of the GMP and GLP steers. Therefore, spring‐born Wagyu steers should be grazed using the GLP supplement before fattening.  相似文献

Abstract

Taxonomic characteristics in accumulating Co and Ni was investigated with 60 plant species (37 genera, 27 families) grown on granite derived soils containing slightly low level of available Co. Considerable variations in Co and Ni content were observed among plant !species with small differences with regard to sampling sites. Exceptionally high Co content (25 ppm on D.W. basis) was found in the leaves of Clethra barbinervis. The value was about 48 times higher than the lowest Co content species. Reasonably high Co content was found with some species in genera: Hydrangea and Viburunum. Ni was highly accumulated in the leaves of Sasa borealis (16 ppm) and in fruits of Clethra barbinervis (65 ppm). [Other Nirich species were especially found in genera: Struthiopteris, Hydrangea, Stewartia, Clethra, and Symplocos. High positive correlation (P<0.01) was found between the concentration of Co and Ni in plant species.  相似文献

A long-term experiment on combined inorganic fertilizers and organic matter in paddy rice (Oryza sativa L.) cultivation began in May 1982 in Yamagata, northeastern Japan. In 2012, after the 31st harvest, soil samples were collected from five fertilizer treatments [(1) PK, (2) NPK, (3) NPK + 6 Mg ha?1 rice straw (RS), (4) NPK + 10 Mg ha?1 rice straw compost (CM1), and (5) NPK + 30 Mg ha?1 rice straw compost (CM3)], at five soil depths (0–5, 5–10, 10–15, 15–20 and 20–25 cm), to assess the changes in soil organic carbon (SOC) content and carbon (C) decomposition potential, total nitrogen (TN) content and nitrogen (N) mineralization potential resulting from long-term organic matter addition. The C decomposition potential was assessed based on the methane (CH4) and carbon dioxide (CO2) produced, while the N mineralization potential was determined from the potassium chloride (KCl)-extractable ammonium-nitrogen (NH4+-N), after 2, 4, 6 and 8 weeks of anaerobic incubation at 30°C in the laboratory. Compared to NPK treatment, SOC in the total 0–25 cm layer increased by 67.3, 21.0 and10.8%, and TN increased by 64.2, 19.7 and 10.6%, in CM3, RS and CM1, respectively, and SOC and TN showed a slight reduction in the PK treatment by 5.2 and 5.7%, respectively. Applying rice straw compost (10 Mg ha?1) instead of rice straw (6 Mg ha?1) to rice paddies reduced methane production by about 19% after the soils were measured under 8 weeks of anaerobic incubation at 30°C. Soil carbon decomposition potential (Co) and nitrogen mineralization potential (No) were highly correlated with the SOC and TN contents. The mean ratio of Co/No was 4.49, lower than the mean ratio of SOC/TN (13.49) for all treatments, which indicated that the easily decomposed organic matter was from soil microbial biomass and soil proteins.  相似文献

We investigated the effects of grazing and supplemental protein sources during the grazing period on growth performance, ruminal characteristics and blood constituents in 15 growing Wagyu steer calves weaned at 3 months of age. Each animal was allotted one of three treatments: (1) GSBM, soybean meal (SBM) supplemented as a protein source during a grazing period; (2) GCGM, corn gluten meal (CGM) supplemented as a protein source during a grazing period; or (3) NG, no grazing, in which steers were fed hay and GSBM concentrate only. The estimated rumen undegradable protein (RUP) concentration of CGM was higher than that of SBM. High RUP supplementation during the grazing period did not influence feed intake or growth performance. Compared with the NG treatment, the average daily gain in GSBM and GCGM calves was significantly depressed (P < 0.05). Bodyweight/withers height ratio in the NG calves tended to be greater than in the GSBM and GCGM calves. Final paunch girth/heart girth ratio in the GSBM and GCGM calves was significantly greater (P < 0.05) than in the NG calves. Supplementation with CGM did not influence ruminal ammonia nitrogen and blood constituents. These results indicate that CGM supplementation during the grazing period did not enhance the growth of Wagyu calves because the CGM diet probably did not elevate the metabolizable protein supply to the grazing calves above that of the SBM diet in our conditions.  相似文献

Despite its explosive properties and toxicity to both animals and humans, diethyl ether is an agent long used in Japan in the anaesthesia jar method of rat anaesthetises. However, in response to a recent report from the Science Council of Japan condemning diethyl ether as acceptable practice, we searched for an alternative rat anaesthesia method that provided data continuous with pre-existing regular toxicology studies already conducted under diethyl ether anaesthesia. For this, we examined two candidates; 30% isoflurane diluted with propylene glycol and pentobarbitone. Whereas isoflurane is considered to be one of the representatives of modern volatile anaesthetics, the method of propylene glycol-diluted 30% isoflurane used in this study was our modification of a recently reported method revealed to have several advantages as an inhalation anaesthesia. Intraperitoneal pentobarbitone has long been accepted as a humane method in laboratory animal anaesthesiology. These 2 modalities were scrutinized in terms of consistency of haematology and blood chemistry with previous results using ether. We found that pentobarbitone required a much longer induction time than diethyl ether, which is suspected to be the cause of fluctuations in several haematological and blood chemical results. Conversely, only calcium ion concentration showed a slight difference from traditional results in the case of 30% isoflurane. Additionally, serum prolactin and corticosterone levels indicated that 30% isoflurane induced less stress than ether, confirming that 30% isoflurane can both provide results consistent with diethyl ether, while at the same time remove its disadvantages. As such 30% isoflurane appears to be a strong alternative anaesthetic agent for future regular toxicology studies in Japan.  相似文献

Asian cultivated rice Oryza sativa L. was domesticated from its wild ancestor, O. rufipogon. During domestication, the cultivated rice lost its seed-shattering behaviour. Previous studies have shown that two major quantitative trait loci (QTLs; qSH1 and sh4) are responsible for the seed-shattering degree. Here, we produced introgression lines carrying non-functional alleles from O. sativa ‘Nipponbare’ at the two major QTLs in the genetic background of wild rice O. rufipogon W630, and examined the effects of the two QTLs on seed shattering and abscission layer formation. The introgression lines, with Nipponbare alleles at either or both loci, showed complete or partial abscission layer formation, respectively, indicating that other unknown loci might be involved in enhancing seed shattering in wild rice. We detected a single QTL named qSH3 regulating seed-shattering degree using an F2 population between Nipponbare and the introgression line carrying Nipponbare alleles at the two QTLs. Although we generated an introgression line for qSH3 alone, no effects on seed shattering were observed. However, a significant effect on seed-shattering degree was observed for the introgression line carrying Nipponbare alleles at qSH3 and the two QTLs, suggesting an important role of qSH3 on seed shattering in coordination with the two QTLs.  相似文献
