ABSTRACT

Calcareous soils typically suffer from zinc deficiency and zinc sulfate is incorporated in many cultivated soils. Utilization of ZnSO4 has some kinds of interaction with soil particles and organic matter. In this study, the efficacy of two znic(Zn)-amino acid chelates (Zn-ACs) i.e., Zn-alanine (Zn-Ala) and Zn-glycine (Zn-Gly) on wheat (Triticum aestivum, cv. N91-8) growth characteristics and zinc concentration in wheat was examined under greenhouse conditions and compared to the a commercial ZnSO4. Results showed that Zn-Ala and Zn-Gly significantly increased the dry weight and shoot length of wheat in comparison to ZnSO4 treatment. Soil application of Zn-Amino acid chelates proved to be the most influential source of zinc in increasing wheat growth and yield indices. Number of fertile spikelet and grain yield increased significantly respectively compared to ZnSO4 treatment. Zn concentration and protein content of wheat grain in Zn-ACs treatment was significantly higher than the ZnSO4 treatment. Soil application of Zn-ACs caused a significant decrease in the grain phytic acid (PA) concentration and also phytic acid to zinc molar ratio in comparison with ZnSO4 treatment. According to the results, Zn-ACs could be utilized as a zinc fertilizer source for improving the zinc bioavailability in wheat.  相似文献

Dicathais orbita is a large Australian marine gastropod known to produce bioactive compounds with anticancer properties. In this research, we used bioassay guided fractionation from the egg mass extract of D. orbita using flash column chromatography and identified fractions containing tyrindoleninone and 6-bromoisatin as the most active against colon cancer cells HT29 and Caco-2. Liquid chromatography coupled with mass spectrometry (LCMS) and 1H NMR were used to characterize the purity and chemical composition of the isolated compounds. An MTT assay was used to determine effects on cell viability. Necrosis and apoptosis induction using caspase/LDH assay and flow cytometry (PI/Annexin-V) and cell cycle analysis were also investigated. Our results show that semi-purified 6-bromoisatin had the highest anti-cancer activity by inhibiting cell viability (IC50 = ~100 µM) and increasing caspase 3/7 activity in both of the cell lines at low concentration. The fraction containing 6-bromoisatin induced 77.6% apoptosis and arrested 25.7% of the cells in G2/M phase of cell cycle in HT29 cells. Tyrindoleninone was less potent but significantly decreased the viability of HT29 cells at IC50 = 390 µM and induced apoptosis at 195 µM by increasing caspase 3/7 activity in these cells. This research will facilitate the development of these molluscan natural products as novel complementary medicines for colorectal cancer.  相似文献

Identification of genetic factors controlling traits associated with seed germination under drought stress conditions, leads to identification and development of drought tolerant varieties. Present study by using a population of F2:, derived from a cross between a drought tolerant variety, Gharib (indica) and a drought sensitive variety, Sepidroud (indica), is to identify and compare QTLs associated with germination traits under drought stress and non-stress conditions. Through QTL analysis, using composite interval mapping, regarding traits such as germination rate (GR), germination percentage (GP), radicle length (RL), plumule length (PL), coleorhiza length (COL) and coleoptile length (CL), totally 13 QTLs were detected under pole drought stress (-8 MPa poly ethylene glycol 6000) and 9 QTLs under non-stress conditions. Of the QTLs identified under non-stress conditions, QTLs associated with COL (qCOL-5) and GR (qGR-1) explained 21.28% and 19.73% of the total phenotypic variations, respectively Under drought stress conditions, QTLs associated with COL (qCOL-3) and PL (qPL-5) explained 18.34% and 18.22% of the total phenotypic variations, respectively. A few drought-tolerance-related QTLs identified in previous studies are near the QTLs detected in this study, and several QTLs in this study are novel alleles. The major QTLs like qGR-1, qGP-4, qRL-12 and qCL-4 identified in both conditions for traits GR, GP, RL and CL, respectively, should be considered as the important and stable trait-controlling QTLs in rice seed germination. Those major or minor QTLs could be used to significantly improve drought tolerance by marker-assisted selection in rice.  相似文献

Prunus scoparia is a wild deciduous shrub, usually living on dry calcareous soils of the rocky mountains and has been used as a grafting rootstock for domesticated almonds to provide drought resistance. In the current study, micropropagation ability of P. scoparia was investigated using cytokinin and auxin. Uniform nodal shoot pieces (3–5 cm in length) of seedlings were used as explants. The explants were disinfected with 10% sodium hypochlorite solution. For adventitious shoot induction and proliferation, Murashige and Skoog (MS) media containing 7.00 g/l agar and 30.00 g/l sucrose containing five concentrations of benzyl adenine (BA) (0.00, 0.50, 1.00, 2.00, and 4.00 mg/1) and also containing six concentrations of Thidiazuron (TDZ) (0.00, 0.50, 1.00, 2.00, 5.00, and 7.00 mg/1) were compared. For rooting, in vitro shoots (2–3 cm) were transferred into ½ MS medium supplemented with 30 g/l sucrose, 7.50 g/l agar, and different concentrations of IBA (0.00, 0.25, 0.50, and 1.00 mg/l) and NAA (0.00, 0.25, 0.50, and 1.00 mg/l). Based on the results obtained for shoot proliferation, only 2.00 and 4.00 mg/l BA and 2.00 mg/l TDZ concentrations generated shoots, while other treatments did not show shoot proliferation. Among the three treatments that generated shoots, the best results for shoot number, leaf number, and leaf color quality were observed in media containing 2.00 mg/l TDZ. Based on the results obtained for rooting, the effect of IBA concentrations on the rooting percentage, root number, and root length was significant. Among IBA concentrations, only 0.50 mg/l IBA induced rooting, while there was no rooting in the media containing other IBA concentrations. None of the NAA concentrations showed rooting. In conclusion, MS culture medium supplemented with 2.00 mg/l TDZ and ½ MS culture medium supplemented with 0.50 mg/l IBA are suggested for in vitro shoot proliferation and rooting of P. scoparia, respectively. The results presented herein could be used for in vitro selection and micropropagation of P. scoparia.  相似文献

Although the effects caused in plants by the calcium (Ca)–phosphorus (P) interaction in calcareous soils are well documented, very few studies report on such effects in nutritive solutions or hydroponic cultivation. In a sand and perlite (1:1 volume) hydroponic system, effects of various P (21, 42, and 63 ppm) and Ca (120, 180, and 240 ppm) concentrations on potato tuberization were studied. A factorial experiment based on a completely randomized design with three replications was conducted. For maximum tuber yield and tuber number production, 21 and 42 ppm P was sufficient, respectively. Increase in P and Ca concentrations in nutrient solutions in early growing season resulted in an increase in shoot and root weight, leaf area, and shoot length linearly, but tuber yield and tuber number did not follow this trend. The maximum tuber specific gravity and total solid percent were also observed in 42 ppm P. The highest tuber number and tuber yield were observed in 120 ppm Ca concentration.  相似文献

A greenhouse pot experiment was conducted to investigate the effect of application of coal gangue (CG) at different rates (0, 5, 10, 20, and 50%) and inoculation with two arbuscular mycorrhizal (AM) fungi Glomus intraradices and Glomus mosseae, as mediating plant adaptation to soil amended with CG, on the nutrient content of forage maize. The results showed CG amendment at all levels and both AM fungi significantly improved the nutrient content of the plant as compared to control. In general, the highest shoot dry weight and nutrient phosphorus, iron, and zinc (P, Fe, and Zn) were obtained with 10% CG and G. intraradice treatments, which were 49.68, 30.49, 16.72, and 75.71% higher than those of the control plants, respectively. Therefore, 10% dose of CG may be considered as a suitable dose for amendment in the corn cultivation bed in terms of providing nutrient contents for this plant as well as AM fungi root colonization.  相似文献
