Wilt disease caused by Fusarium solani is a serious constraint to Dalbergia sissoo (shisham) plantations in northern India. In this study, the antagonistic potential of 40 bacterial isolates recovered from rhizophere soil of healthy shisham trees, and a well‐characterized Trichoderma species (Trichoderma virens) were tested for their possibility as biocontrol agents for F. solani. Two promising isolates (S1 and S15) were identified which inhibited pathogen growth, caused chitin degradation, produced siderophores and solubilized phosphate in vitro. Isolate S15 scored highest for hydrogen cyanide (HCN) production while isolate S1 was a non‐HCN producer. These two isolates were identified as Serratia marcescens (S1) and Pseudomonas azotoformans (S15) following sequence analysis of 16S rDNA. In dual culture assays, T. virens caused 80% inhibition of mycelial growth of the test fungus. The three selected antagonists when tested in planta in the glasshouse completely suppressed production of wilt symptoms on 12‐month‐old shisham plants. Further work is needed to ascertain the potential of these isolates to be used as biocontrol agents to manage shisham wilt under field conditions.  相似文献

Vermicomposting of fly ash has been attempted, using red earthworm, Eisenia foetida. Fly ash, which was obtained from thermal power station, was mixed with cowdung in different proportions (20, 40, 60 and 80%). These mixtures were used as feed for earthworms, and after 30 days, vermicast recovery, worm zoomass and numbers of juveniles produced were recorded. A total of six runs each of 30 days were conducted during the whole study. Concentrations of heavy metals in different mixtures of fly ash–cowdung, before and after vermicomposting and in the earthworms used in the study were also estimated. Results show maximum output of vermicasts and maximum number of juveniles produced was in reactors with 40% fly ash while maximum weight gain by earthworm was in 20% fly ash vermireactors. Performance of vermireactors up to 60% fly ash was more or less similar but at 80% fly ash, there is a marked reduction in overall performance of the reactors. Chemical analysis of different samples of fly ash–cowdung mixtures prior to vermicomposting revealed high concentrations of zinc, chromium, lead, nickel and copper. Chemical analysis of vermicomposted samples showed 30–50% reduction in heavy metals up to 60% fly ash and 10–30% reduction in 80% fly ash. Metal analysis of earthworms revealed considerable bioaccumulation of heavy metals in their body. The Present study indicates the feasibility of E. foetida for mitigating the toxicity of metals and up to 60% fly ash–cowdung mixtures can be used for sustainable and efficient vermicomposting.  相似文献

Eremostachys superba Royle ex Benth. (Lamiaceae) has undergone a severe decline in population size since its discovery in the North-western Himalayas in late 19th century. One hundred and seventy-two plants from six populations in the Indian states of Uttar Pradesh and Jammu & Kashmir, located between 0.45 km and 455.72 km apart from each other were evaluated for RAPD polymorphism. Sixteen random primers generated 92 bands overall, 77 of which were polymorphic. Shannon’s index of genetic diversity within populations (H o) ranged between 0.305 and 0.421; the average within-population diversity (H pop) was 0.389; and the total species diversity (H sp) was 0.478. The population from Mohand (representing the type locality) had the fewest plants, at 18, and was genetically the most depauperate. Among the other populations, ranging in size between 52 and 1,022 individuals, no relation between population size and genetic diversity was evident. It is suggested that these six populations represent relics of a larger, extended population, in which the presence of perennating rootstocks has helped preserve historic patterns of genetic diversity. AMOVA revealed that 83.01% of the variation exists within populations, which was consistent with earlier studies on the reproductive biology of E. superba, which indicated this species is predominantly allogamous. FST distances between all populations were significant, indicating geographic differentiation despite some of them being closely separated. Habitat restoration and protection from indiscriminate harvesting are proposed as primary strategies for conserving E. superba. Rejuvenation of the Mohand population through intrapopulation crossing between plants bearing diverse molecular phenotypes is also suggested.  相似文献

Eucalyptus spp. and their hybrids are frequently cloned and mass planted across farmland tracts and commercial plantations in northern India. It is a viable feeder species to the paper and pulp industries in this region. In 2018 and 2019, during field surveys conducted in northern India, a serious leaf blight disease was frequently observed in E. tereticornis plantations. Isolation from the blighted leaf samples consistently yielded fungal isolates having Calonectria‐like morphology. Morphological features coupled with sequence analysis of partial β‐tubulin (TUB2) and partial translation elongation factor‐alpha (TEF1) gene regions of two fungal isolates confirmed the species as Ca. cerciana. In detached leaf assays and glasshouse inoculation experiments, both isolates produced symptoms similar to those observed on the naturally infected leaves. Koch's postulates were fulfilled by re‐isolating Ca. cerciana from the inoculated leaves. This work is the first to confirm that Ca. cerciana is associated with a serious leaf blight disease of Eucalyptus in northern India and is an important addition to the taxonomy of Calonectria fungi in India.  相似文献

A field experiment was conducted to study the effect of planting material and plant density on stevia (Stevia rebaudiana Bertoni) under western Himalayan conditions during 2011 and 2012. The experiment conducted in a split plot design consisted of two types of planting material (rooted slips and fresh seedlings) in the main plot and five inter- and intra-row spacing in subplots with three replications. Yield attributes and dry leaf biomass yield of stevia were not affected by the type of planting material; however, plant density significantly influenced the yield attributes and leaf and stem dry biomass. Although the wider spacing (60 × 45 cm) gave more leaves, higher leaf area index, higher leaf dry mass per plant as compared to closer spacing, it resulted in lower values of these attributes per unit area. Plants spaced in 30 × 30 cm accumulated 41.2% and 42.8% more total biomass than 60 × 45 cm. Steviol glycoside content did not change due to different planting materials and plant densities; however, closer plant spacing (30 × 30 cm) recorded 114.8% and 70.0% higher steviol glycoside accumulation compared to wider row spacing (60 × 45 cm) in 2011 and 2012, respectively.  相似文献

β‐glucan binding protein (βGBP), a pattern recognition protein was purified from the haemolymph of freshwater prawn Macrobrachium rosenbergii by heparin affinity chromatography that showed a single band in native gradient PAGE. The β‐glucan binding property of the purified protein was confirmed in a phenoloxidase (PO) assay, where addition of βGBP along with β‐glucan increased the specific PO activity compared with that of β‐glucan alone. The molecular weight of the βGBP was found to be ~316 kDa on gel filtration chromatography. In SDS‐PAGE, βGBP molecule was reduced to one polypeptide chain of molecular weight ~113 kDa. Thus the βGBP in M. rosenbergii is possibly a homotrimeric molecule. The purified sample run on unreduced condition in SDS‐PAGE also revealed a similar size band (~113 kDa) and hence, the polypeptide chains of βGBP are held by non‐covalent interactions. The purified βGBP samples run in native PAGE was stained positively with alcian blue for carbohydrates and Sudan black for lipids indicating the βGBP to be a glycolipoprotein. With rabbit polyclonal anti‐βGBP serum developed, an indirect ELISA was standardized and the normal βGBP concentration in adult M. rosenbergii serum was quantified to be ~2 mg mL?1. Furthermore, the applicability of the developed ELISA is discussed.  相似文献

Chickpea (Cicer arietinum L.) is a dry season food legume largely grown on residual soil moisture after the rainy season. The crop often experiences moisture stress towards end of the crop season (terminal drought). The crop may also face heat stress at the reproductive stage if sowing is delayed. The breeding approaches for improving adaptation to these stresses include the development of varieties with early maturity and enhanced abiotic stress tolerance. Several varieties with improved drought tolerance have been developed by selecting for grain yield under moisture stress conditions. Similarly, selection for pod set in the crop subjected to heat stress during reproductive stage has helped in the development of heat‐tolerant varieties. A genomic region, called QTL‐hotspot, controlling several drought tolerance‐related traits has been introgressed into several popular cultivars using marker‐assisted backcrossing (MABC), and introgression lines giving significantly higher yield than the popular cultivars have been identified. Multiparent advanced generation intercross (MAGIC) approach has been found promising in enhancing genetic recombination and developing lines with enhanced tolerance to terminal drought and heat stresses.  相似文献

To analyze the relationship between the management of three forest stand plantations and soil quality in the Dimapur district of the Northeastern Himalayan region,India,three forest soil profiles,covered by different species stands,at three depths were tested for 13 physical and chemical variables.Only four of these variables(electric conductivity,bulk density exchangeable Mg and available P)were included in a minimum data set,after using a varimax rotation algorithm in a principal component analysis,and subsequently used to calculate a soil quality index(SQI).Results showed higher SQIs in the surface layers(0–20 cm depth)than in the deeper ones.Average weighed SQI varied significantly(P<0.05)through the three considered forest sites,with the lowest value at site FS3.These findings reveal that the approach used here is suitable for preliminary screening of the impact of a forestry species on soil,to aid in species selection and improve soil health for afforestation and reforestation projects.  相似文献

Cold-adapted bioinoculants are considered as harbingers of sustainable hill agriculture. Therefore, two previously characterized psychrotolerant diazotrophs, Pseudomonas jesenii MP1 and Rhodococcus qingshengii S10107, were evaluated for their plant growthpromoting potential for chickpea (Cicer arietinum L.) grown under natural field conditions. Comparative analysis of agronomical and biochemical crop parameters revealed the irrelevance of chemical fertilizers for chickpea production; the diazotrophs alone were sufficient to fulfil the crop''s nutritional requirement. However, the integrated use of bacterial strains in combination with urea at 20 kg N ha-1 as urea was being recommended for higher crop yield and better soil nitrogen status. Quantitative polymerase chain reaction (qPCR) and denaturing gradient gel electrophoresis (DGGE)-based soil bacterial dynamics unveiled the persistence of both diazotrophs until the end of the crop maturation period without affecting the native micro-flora. Therefore, these bioinoculants can be explored as natural nitrogen resource, and an additional incentive in their bio-formulation will be a step towards agricultural sustainability.  相似文献
