Excessive use of nitrogen(N) fertilizers in agricultural systems increases the cost of production and risk of environmental pollution. Therefore, determination of optimum N requirements for plant growth is necessary. Previous studies mostly established critical N dilution curves based on aboveground dry matter(DM) or leaf dry matter(LDM) and stem dry matter(SDM), to diagnose the N nutrition status of the whole plant. As these methods are time consuming, we investigated the more rapidly determined leaf area index(LAI) method to establish the critical nitrogen(N_c) dilution curve, and the curve was used to diagnose plant N status for winter wheat in Guanzhong Plain in Northwest China. Field experiments were conducted using four N fertilization levels(0, 105, 210 and 315 kg ha-1) applied to six wheat cultivars in the 2013–2014 and 2014–2015 growing seasons. LAI, DM, plant N concentration(PNC) and grain yield were determined. Data points from four cultivars were used for establishing the N_c curve and data points from the remaining two cultivars were used for validating the curve. The N_c dilution curve was validated for N-limiting and non-N-limiting growth conditions and there was good agreement between estimated and observed values. The N nutrition index(NNI) ranged from 0.41 to 1.25 and the accumulated plant N deficit(N_(and)) ranged from 60.38 to –17.92 kg ha~(-1) during the growing season. The relative grain yield was significantly affected by NNI and was adequately described with a parabolic function. The N_c curve based on LAI can be adopted as an alternative and more rapid approach to diagnose plant N status to support N fertilization decisions during the vegetative growth of winter wheat in Guanzhong Plain in Northwest China.  相似文献

Methylene chloride and methanol extracts of 20 Indonesian plants with ethnomedical uses have been assessed for in vitro antibacterial and antifungal properties by disk diffusion method. Extracts of the six plants: Terminalia catappa, Swietenia mahagoni Jacq., Phyllanthus acuminatus, Ipomoea spp., Tylophora asthmatica and Hyptis brevipes demonstrated high activity in this bioassay system. These findings should stimulate the search for novel, natural product such as new antibacterial and antifungal agents.  相似文献

Oak mortality is often associated with a complex of decline factors. We describe the morphological and physiological responses of coast live oak, Quercus agrifolia Née, in California to an invasive insect, the goldspotted oak borer (GSOB), Agrilus auroguttatus Schaeffer (Coleoptera: Buprestidae), and evaluate drought as a potential inciting factor. Morphological traits of 356 trees were assessed and physiological traits of 70 of these were monitored intensively over one growing season. Morphological characteristics of tree health included crown thinning and dieback; bole staining resulting from larval feeding; density of GSOB adult exit holes; and holes caused by woodpecker feeding. These characteristics were used to rank GSOB infestation/injury into four classes, and taken together, they explained 87% of the variation in a principal component analysis. Drought stress on various size/age and infestation classes of Q. agrifolia was measured by assessing branchlet pre-dawn and solar noon xylem water potential, leaf cell turgor potential, and water use efficiency over one growing season. Both morphological and physiological traits were highly variable in mature and old growth trees. Early summer plant water status (branchlet xylem water potential and water use efficiency) was similar between uninfested and newly colonized trees, suggesting that GSOB are not pre-selecting drought-stressed Q. agrifolia for oviposition. By late summer, leaf water and cell turgor potentials were lower in infested than in uninfested mature trees, suggesting that GSOB infestation causes drought stress in these trees. Among the tree size/age classes, infested old growth trees exhibited the greatest change in water use efficiency over the growing season, and showed greater morphological injury symptoms of decline than infested mature trees. Morphological attributes of decline in Q. agrifolia associated with GSOB were correlated weakly with increasing physiological drought stress among infestation classes of trees. We propose that the collection of morphological responses of Q. agrifolia to GSOB described here can be used to monitor the future expansion of the GSOB distribution as well as the GSOB-induced decline of Q. agrifolia in California.  相似文献

Accumulated soil P in agricultural soils is a major source of soluble and particulate forms of P entering water resources and degrading water quality. However, few research sites are currently available to evaluate the long-term effects of different cropping systems and fertility practices on soil inorganic and organic P accumulation. The objectives of this study were: (1) to compare the forms and quantity of different inorganic and organic soil P fractions in plots on Sanborn Field, which has been cultivated for 111 years; and (2) to assess the use of standard soil test P extractants for determining changes in soil P dynamics over time. A modified sequential P extraction procedure was used to separate labile and stable inorganic and organic P pools from surface soils collected on Sanborn Field in 1915, 1938, 1962, and 1999 from plots in continuous corn, continuous wheat, continuous timothy, and a corn-wheat-clover rotation amended with either manufactured fertilizers, horse or dairy manure or receiving no fertilization since 1888. Additional samples were collected from a native grass prairie site of a similar soil series to estimate soil characteristics at Sanborn Field before initial cultivation in 1888. Observed accumulation of Bray-1 P among fertilizer and manure treatments was attributed to over-application of P due to unrealistically high yield goals for each cropping system. Long-term cultivation of Sanborn Field increased soil bulk density and lowered soil pH and total organic C compared with native prairie. Fertilization either by addition of manufactured fertilizer or manure significantly increased inorganic resin-P and inorganic NaOH-extractable P. Applications of animal manure also significantly increased most organic P fractions compared with the unfertilized treatment. The native prairie had a larger proportion of total P in organic forms compared with cultivated plots, especially in organic NaOH-extractable P, but no significant decreases in either residual or total P were observed due to cultivation. This study confirms that soil P availability in cropping systems that are amended with predominantly organic P amendments may differ from conventional cropping systems relying on manufactured P fertilizers. However, no direct evidence was found to support the hypothesis that any individual inorganic or organic soil P fraction has a better relationship than conventional soil test P extractants with plant P uptake under contrasting organic and conventional fertility practices.  相似文献

Yessotoxins from a large-scale culture (226 L) of Protoceratium reticulatum strain CAWD129 were harvested by filtration followed by solid-phase extraction. The extract was purified by column chromatography over basic alumina and reverse-phase flash chromatography to afford pure yessotoxin (193 mg). Isolation of yessotoxin was greatly facilitated by selection of a strain which did not produce analogues that interfered with yessotoxin isolation. In addition to yessotoxin, numerous minor yessotoxins were detected by LC-MS in other fractions. From one of these, an early eluting minor analogue with the same molecular weight as yessotoxin and a similar mass spectrometric fragmentation pattern was isolated. This analogue was identified by NMR and mass spectrometry as a novel yessotoxin analogue containing a furan ring in the side chain. This finding reveals biosynthetic flexibility of the yessotoxin pathway in P. reticulatum and confirms earlier findings of production of many minor yessotoxin analogues by this alga. Production of these analogues appeared to be a constitutive trait of P. reticulatum CAWD129.  相似文献

OBJECTIVE: Previous research on nutrition labelling has mainly used subjective measures. This study examines the effectiveness of two types of nutrition label using two objective measures: eye movements and healthiness ratings. DESIGN: Eye movements were recorded while participants made healthiness ratings for two types of nutrition label: standard and standard plus the Food Standards Agency's 'traffic light' concept. SETTING: University of Derby, UK. SUBJECTS: A total of 92 participants (mean age 31.5 years) were paid for their participation. None of the participants worked in the areas of food or nutrition. RESULTS: For the standard nutrition label, participant eye movements lacked focus and their healthiness ratings lacked accuracy. The traffic light system helped to guide the attention of the consumer to the important nutrients and improved the accuracy of the healthiness ratings of nutrition labels. CONCLUSIONS: Consumers have a lack of knowledge regarding how to interpret nutrition information for standard labels. The traffic light concept helps to ameliorate this problem by indicating important nutrients to which to pay attention.  相似文献

Sea urchins are commercially harvested for their edible gonads, also known as roe or uni. Most species, particularly those from the Northern hemisphere are reported to exhibit limited variation in gonad colour, which is proposed to be due to carotenoid pigments. The commercially harvested species from New Zealand, Evechinus chloroticus, and from Australia, Heliocidaris erythrogramma, however, exhibit considerable gonad colour variation, ranging from a culinary desirable light yellow/orange to an undesirable brown. Gonads from E. chloroticus and H. erythrogramma were characterized spectrophotometrically and the carotenoid content extracted and analysed by reversed‐phase high‐performance liquid chromatography. The main carotenoid found in the gonads of both species was 9′‐cis‐echinenone, with lower levels of all‐trans‐echinenone, lutein and isozeaxanthin. Carotenoid composition contributed to the yellow/orange colour components of the gonads, but was not correlated with the darker roe coloration observed in some E. chloroticus individuals. Our results highlight important considerations relating sea urchin gonad coloration to carotenoid content and as such will provide useful information for the future aquaculture and processing of these two species.  相似文献

Azaspiracid (AZA) poisoning has been reported following consumption of contaminated shellfish, and is of human health concern. Hence, it is important to have sustainable amounts of the causative toxins available for toxicological studies and for instrument calibration in monitoring programs, without having to rely on natural toxin events. Continuous pilot scale culturing was carried out to evaluate the feasibility of AZA production using Azadinium spinosum cultures. Algae were harvested using tangential flow filtration or continuous centrifugation. AZAs were extracted using solid phase extraction (SPE) procedures, and subsequently purified. When coupling two stirred photobioreactors in series, cell concentrations reached 190,000 and 210,000 cell · mL(-1) at steady state in bioreactors 1 and 2, respectively. The AZA cell quota decreased as the dilution rate increased from 0.15 to 0.3 day(-1), with optimum toxin production at 0.25 day(-1). After optimization, SPE procedures allowed for the recovery of 79 ± 9% of AZAs. The preparative isolation procedure previously developed for shellfish was optimized for algal extracts, such that only four steps were necessary to obtain purified AZA1 and -2. A purification efficiency of more than 70% was achieved, and isolation from 1200 L of culture yielded 9.3 mg of AZA1 and 2.2 mg of AZA2 of >95% purity. This work demonstrated the feasibility of sustainably producing AZA1 and -2 from A. spinosum cultures.  相似文献

Soybean rust, caused by Phakopsora pachyrhizi Sydow, is a severe foliar disease of soybean [Glycine max (L.) Merr.] that occurs throughout most soybean producing regions of the world. The objective of this research was to evaluate selected soybean genotypes for resistance to soybean rust in Vietnam. Five field experiments in Vietnam were completed from 2006 to 2009. The area-under-the-disease-progress-curve (AUDPC) was calculated for each soybean genotype based on four disease assessments taken during the reproductive growth stages. AUDPC units among soybean genotypes in each experiment differed (P < 0.05). Over the five experiments, the resistant check DT 2000 was most often the genotype with the lowest AUDPC units while the sources of rust resistance (Rpp1-5) did not always have low AUDPC units in each experiment, although PI 230970 (Rpp2) appeared to be more stable. A few genotypes with non-characterized genes for resistance, such as PI 398998, PI 437323, and PI 549017, had the lowest AUDPC units in at least one of the experiments. These genetic resources may be useful for host plant resistance studies and breeding soybeans for rust resistance in Vietnam and other locations like Brazil and the United States that have more recently been inundated with soybean rust. A significant (P < 0.001) experiment × genotype interaction was found when the AUDPC data of 14 soybean genotypes tested in Experiments 1, 2, and 3 were combined and analyzed. This result indicates the potential importance of changing fungal races and/or biotypes that occur in the rust population.  相似文献
