Embryos obtained via superovulation are necessary for mammalian artificial reproduction, and viability is a key determinant of success. Nonfreezing storage at 4 C is possible, but currently used storage solutions can maintain embryo viability for only 24–48 h. Here we found that 10 mg/ml antifreeze protein (AFP) dissolved in culture medium 199 with 20% (v/v) fetal bovine serum and 25 mM HEPES could keep bovine embryos alive for 10 days at 4 C. We used a recombinant AFP isolated from the notched-fin eelpout (Zoarces elongatus Kner). Photomicroscopy indicated that the AFP–embryo interaction was enhanced at 37 C. Embryos pre-warmed with the AFP solution at 37 C for 60 min maintained high viability, whereas those that were not pre-warmed could live no longer than 7 days. Thus, short-term storage of bovine embryos was achieved by a combination of AFP-containing medium and controlled pre-warming. 相似文献

The applicability of the recombinant LipL32 for serodiagnosis of leptospiral infection in field rodents was
assessed in this study. An immunodominant region of LipL32 was determined by monoclonal antibodies, and then,
truncated LipL32 (tLipL32) was designed to contain the region (87–188th amino acid). The tLipL32 was compared
between two recombinant expression hosts Escherichia coli and Pichia
pastoris in ELISA. With field rat sera, tLipL32 expressed by P. pastoris
(tLipL32p) had high antigenicity without background reactions, while tLipL32 expressed by E.
coli (tLipL32e) showed high background reactions, which were reduced by pre-adsorption of sera with
E. coli. To evaluate tLipL32-ELISA, field rat sera were tentatively divided into a
Leptospira infection positive (12 sera) and a negative group (12 sera) based on the results
from flaB gene PCR of kidney samples and WB with whole Leptospira cell.
Consequently, the sensitivity of tLipL32p-ELISA for field rat sera was 83% . A similar result was obtained
from tLipL32e-ELISA with adsorbed sera, (92%). However, sensitivity of tLipL32e-ELISA using sera without an
adsorption treatment was 50%. Regardless of the expression host, tLipL32-ELISA had 100% specificity and
sensitivity in experimentally infected laboratory rats. These results suggest that recombinant LipL32
expressed by P. pastoris is more applicable for serodiagnosis in field rats due to a lack of

In a previous study, we found that a 70% aqueous ethanol extract of the fresh aerial tissue of Geranium carolinianum L. showed antimicrobial activity against the pathogen causing soil rot of sweet potato. As the appropriate time for cultivation of sweet potato and the growing period of G. carolinianum do not overlap in Okinawa Prefecture, Japan, the fresh aerial tissue is available in order to control soil rot of sweet potato. Thus, we examined the control effect of fresh aerial tissue against soil rot of sweet potato. The various trials (a single repetition of 20 m2 ) were performed in fields that had undergone 8 years of continuous cropping of sweet potato at Okinawa Prefectural Agricultural Experiment Station, Horticultural branch. After harvest, when the disease severity was evaluated by determining the necrotic area of the storage root, the incorporation of fresh aerial tissue (5000 kg 1000 m-2 ) into the soil was considered to be highly effective, with a protective value of 75.4. This result shows that G. carolinianum could be used as a biological agent for the control of soil rot of sweet potato. 相似文献

Fresh aerial tissue and roots of 14 wild plants in Okinawa prefecture were investigated for their antimicrobial activity against Ralstonia solanacearum , which causes bacterial wilt of potato. A 70% aqueous ethanol extract of fresh aerial tissue of Geranium carolinianum L. showed strong antimicrobial activity against R. solanacearum . This extract also showed antimicrobial activity against the pathogens causing common scab of potato and soil rot of sweet potato. The antimicrobial substance could be extracted with hot water, and was effective against R. solanacearum in soil. In the field test, a treatment combining incorporation of dried aerial tissue into the soil and solarization was highly effective for control of bacterial wilt of potato. These findings suggest that G. carolinianum L. could be used as a biological agent for the control of bacterial wilt of potato. 相似文献

Commercially available tomato cultivars were hydroponically cultured for inoculation, with Ralstonia solanacearum (K-101), which causes bacterial wilt, by pouring an inoculum suspension into the nutrient solution. Cultivar susceptibility
to the bacteria was evaluated, based on the highest percentage of wilting. Because the length of time for wilt appearance
varied among cultivars, some cultivars appeared to be suppressive to the translocation and/or multiplication of the invading
pathogen. Thus, this hydroponic inoculation system is effective for examining levels of susceptibility in tomato cultivars
to bacterial wilt.

Environmental variability and growth‐rate histories from hatching to capture were investigated for larval Japanese sardine (Sardinops melanostictus) and Japanese anchovy (Engraulis japonicus). Larvae collected around the front of the Kuroshio Current were examined using otolith microstructure analysis, and their movement was estimated from numerical particle‐tracking experiments. Sardine larvae collected inshore of the Kuroshio front originated from a coastal area near the sampling site, while those collected in the offshore area originated from an area 500–800 km west‐southwest of the sampling site. Anchovy larvae collected both inshore and offshore had been transported from widely distributed spawning areas located west of the sampling area. At the age of 13–14 days for sardine and 19–20 days for anchovy, the offshore group exhibited significantly higher mean growth rates than did the inshore group. Although the offshore area was generally warmer than the inshore area, temporal variations in growth rate are not attributable solely to fluctuations in environmental temperature. While previous studies have examined the relationship between larval growth rates and environment based solely on data at capture, the methods used in the present study, combining otolith analysis and numerical particle‐tracking experiments, utilize data up until hatching. Although the relationship between growth rate and environment was not fully confirmed, this approach will greatly advance our understanding of fish population dynamics. 相似文献
