An analysis of the polar extracts from sweet potato Ipomoea batatas (Convolvulaceae) led to the isolation of seven unknown aminoacyl sugars. On the basis of 1D, 2D NMR, and mass spectrometry data, the structures of the compounds were elucidated as: beta-D-fructofuranosyl-(2 --> 1)-alpha-D-[2-O-valyl]-glucopyranoside (1), beta-D-fructofuranosyl-(2 --> 1)-alpha-D-[2-O-tyrosyl]-glucopyranoside (2), beta-D-fructofuranosyl-(2 --> 1)-alpha-D-[2-O-threonyl]-glucopyranoside (3), beta-D-fructofuranosyl-(2 --> 1)-alpha-D-[2-O-hystidyl]-glucopyranoside (4), 2-beta-D-fructofuranosyl-(2 --> 1)-alpha-D-[2-O-alanyl]-glucopyranoside (5), beta-D-fructofuranosyl-(2 --> 1)-alpha-D-[2-O-tryptophanyl]-glucopyranoside (6), and beta-D-fructofuranosyl-(2 --> 1)-alpha-D-[2-O-glycyl]-glucopyranoside (7).  相似文献

Four different polyphenolic compounds were isolated by chromatographic methods from methanolic and hydromethanolic extracts of Ipomoea batatas tuber flour. On the basis of UV, mass, and NMR analysis procedures, the structure of the isolated compounds were determined as 4,5-di-O-caffeoyldaucic acid (1), 4-O-caffeoylquinic acid (2), 3,5-di-O-caffeoylquinic acid (3), and 1,3-di-O-caffeoylquinic acid (4). To the best of our knowledge, this is the first report of isolation and characterization of compound 1. Then, we evaluated the antioxidant activity of daucic acid derivative by using DPPH and FRAP methods together with authentic antioxidant standards, l-ascorbic acid, tert-butyl-4-hydroxy toluene (BHT), and gallic acid. The activity of compound 1 in both methods was higher than that of all standards used at the same molar concentration.  相似文献

The antiradical activity of dry beans was evaluated in order to assess the validity of this test and to correlate the results with those achieved with the Folin-Ciocalteu method and with a rapid spectrophotometric method for the analysis of total flavonoids. Four landraces (12 samples) of common beans (Phaseolus vulgaris L.), collected in two regions of Italy (Tuscany and Basilicata) in three different years, were analyzed. The EC50 values ranged from 39 to 2810 mg sample/mg 1,1-diphenyl-2-picrylhydrazil radical. The phenolic content of each sample was expressed as gallic acid equivalents; it changed from 1.17 to 4.40 mg/g. The flavonoid content, expressed as mg of (+)-catechin per g of dry seeds, ranged from 0.24 to 1.43 mg/g. The qualiquantitative composition of polyphenols has been also elucidated by means of high-performance liquid chromatography (HPLC)-diode array detection and HPLC/MS. These investigations showed that rapid tests can contribute to assessing the quality of functional food.  相似文献

Sixty-seven strains of Bradyrhizobium isolated from soybean plants growing on acid soils in West Java and Sumatra, Indonesia, were examined for the effect of the pH and aluminum concentration on their growth in nutrient media, compared with 61 strains of Bradyrhizobium from soils in Japan. The results in this study indicated that the indigenous population of Bradyrhizobium in the soils of Indonesia showed a large difference in acid- and Al-tolerance from that of Japan. Eighty-five and 48% of the isolates from Japanese soils and Indonesian soils, respectively, were unable to grow in YEM broth at pH below 4.5. The acid-tolerance was correlated with AI-tolerance of the isolates on YEM agar plates at pH 4.4. Seventy-five percent of the isolates that grew in YEM broth at pH 4.5 were also resistant to 400 µM Al on the YEM plates. Acetylene reduction assay of the root nodules revealed that 3 of the acid- and Al-tolerant isolates from Indonesian soils showed a significantly high nitrogen fixation activity.  相似文献

Several methods have been proposed to advance the onset of the breeding season in horses. Most of them are based on the exposure to an artificial lighting period combined with hormonal treatments. Mares exposed to an artificial photoperiod are most often housed indoors where the ambient temperature is often higher than the outside temperature. Mares held in barns are also exposed to different daylight intensities than horses kept outside, depending on the architecture. In the current study, we evaluated the impact of ambient temperature, daylight intensity and changes in body condition score (BCS) on the timing of first ovulation after winter anestrus in mares exposed to an artificial photoperiod. Mares (n = 211) were housed in barns with different ambient temperature and daylight exposure but with the same artificial photoperiod exposure (except for a natural photoperiod control group). Artificial photoperiod as well as an increase in BCS over the winter significantly advanced the first spring ovulation. The BCS at the start and end of the anestrus period did not have an effect on the interval to first ovulation and neither did the modest increase in ambient temperature in the barn. However, a higher light intensity during the daytime significantly advanced the first spring ovulation. The results of this study suggest that exposure to more sunlight advances the onset of the breeding season. This effect is likely mediated through the biological effect of short wavelength blue light and its impact on melatonin suppression and biological rhythms. We suggest that greater/direct exposure to the blue light component of daylight improves the response to the artificial photoperiod. The results of the present study can further assist to optimize the conditions that lead to an efficient spring transition of breeding mares.  相似文献

The objective of this study was to evaluate the accuracy of a novel, portable device (iSperm® Equine for assessing concentration and motility of stallion semen). In the first experiment, semen concentration was determined by the iSperm® Equine (Aidmics Biotechnology), Androvision® (Minitube) and NucleoCounter® SP‐100? (ChemoMetec). The total motility and progressive motility were determined by the iSperm® Equine and the Androvision® using the manufacturer's guidelines. Frozen/thawed semen samples (n = 33) at various dilutions were analysed for concentration and motility with the above‐mentioned devices. There was a significant correlation between the concentrations measured with iSperm® and NucleoCounter® at all the measured dilutions. Moreover, <10% difference in concentrations was observed between the iSperm® and NucleoCounter® using the Bland–Altman test. There was also a significant correlation between iSperm® and Androvision® for total and progressive motility. In the second experiment, the parameters used in the Androvision® were modified to match those of the iSperm®. Total motility and progressive motility of frozen/thawed semen samples (n = 10) were determined, and the similarity between the Androvision® and iSperm® was confirmed by correlation studies and Bland–Altman test. The results of these experiments demonstrate that the iSperm® offers a reliable and practical alternative for the semi‐automated measurement of concentration and motility of stallion semen in the field. The iSperm® enables the practitioner to obtain objective and repeatable measurements on a variety of semen types (fresh, cooled and frozen) in the field at the time of insemination and thus acquire more insight into the quantity and quality of the provided insemination doses. This mare‐side diagnostic tool may help practitioners in identifying presumed subfertility problems more rapidly and act accordingly.  相似文献

Small ubiquitin-like modifier (SUMO)-conjugating enzymes are involved in post-translational regulatory processes in eukaryotes, including the conjugation of SUMO peptides to protein substrate (SUMOylation). SUMOylation plays an important role in improving plant tolerance to abiotic stress such as salt, drought, heat and cold. Herein, we reported the isolation of OsSCE1 (LOC_Os10g39120) gene encoding a SUMO-conjugating enzyme from rice (Oryza sativa cv. Nipponbare) and its functional validation in response to drought stress. The E2 enzyme, OsSCE1, is one of three key enzymes involved in the conjugation of SUMO to its target proteins. Activated SUMO is transferred to the cysteine of an E2 enzyme and then to the target lysine residue of the substrate, with or without the help of an E3 SUMO ligase. Expression of OsSCE1 was strongly induced by polyethylene glycol 6000 (PEG6000) treatment, which suggested OsSCE1 may be involved in the drought stress response. Overexpression of OsSCE1 (OsSCE1-OX) in Nipponbare reduced the tolerance to drought stress. Conversely, the drought tolerance was slightly improved by the knockdown of OsSCE1 (OsSCE1-KD). These results were further supported by measurement of proline content in OsSCE1-OX and OsSCE1-KD transgenic lines under induced drought stress, which showed OsSCE1-KD transgenic lines accumulated higher proline content than the wild type, whereas OsSCE1-OX line had lower proline content than the wild type. These findings suggested OsSCE1 may play a role as a negative regulator in response to drought stress in rice.  相似文献

Cyanobacteria from underexplored and extreme habitats are attracting increasing attention in the search for new bioactive substances. However, cyanobacterial communities from tropical and subtropical regions are still largely unknown, especially with respect to metabolite production. Among the structurally diverse secondary metabolites produced by these organisms, peptides are by far the most frequently described structures. In this work, liquid chromatography/electrospray ionization coupled to high resolution quadrupole time-of-flight tandem mass spectrometry with positive ion detection was applied to study the peptide profile of a group of cyanobacteria isolated from the Southeastern Brazilian coastal forest. A total of 38 peptides belonging to three different families (anabaenopeptins, aeruginosins, and cyanopeptolins) were detected in the extracts. Of the 38 peptides, 37 were detected here for the first time. New structural features were proposed based on mass accuracy data and isotopic patterns derived from full scan and MS/MS spectra. Interestingly, of the 40 surveyed strains only nine were confirmed to be peptide producers; all of these strains belonged to the order Nostocales (three Nostoc sp., two Desmonostoc sp. and four Brasilonema sp.).  相似文献
