Summary Individual tracheid lengths were measured in macerated tissue samples of Pinus merkussii Jungh. and de Vries, P. kesiya Royle ex Gordon (syn. P. khasya Royle) and P. patula Schiede and Deppe. The sample trees were selected from stands of different ages, treatments and sites in central Africa.A sampling unit of 50 tracheids, measured for convenience on two microscope slides, was chosen on the basis of an earlier study. The variation in individual tracheid length and the variation of within-slide variance was studied by means of analysis of variance and variance components. This indicated the nunber of samples required in the different sampling categories to give maximum sampling efficiency. Sampling categories included both random factors (individual tracheids, number of slides, radii in discs) and fixed factors (annual rings, tree classes, thinning treatments, etc.).While large effects were attributable to fixed factors more than half the total variation was explained by random factors. The effect of the number of slides was rarely important and two slides need be used only to facilitate the location of the required number of tracheids and to provide a check on operator consistency. The sample unit of 50 tracheids can detect differences in tracheid length of 0.20 ... 0.34 mm thus confirming the routine used in this laboratory as a practical and meaningful procedure.Laboratory work was carried out in Kitwe, computing at Oxford. Methods based on liquid chromatography-mass spectrometry (LC-MS) and protein trap mass spectrometry (trap-MS) were developed to determine the complement of pathogenesis-related (PR) proteins in grape juice. Trap-MS was superior to LC-MS in terms of simplicity, rapidity, and sensitivity. Proteins with a wide range of masses (13--33 kDa) were found in the juices of 19 different varieties of grape (Vitis vinifera) and were identified as mostly PR-5 type (thaumatin-like) and PR-3 type (chitinases) proteins. Although the PR proteins in juices of grapes are highly conserved, small consistent differences in molecular masses were noted when otherwise identical proteins were compared from different varieties. These differences persisted through different harvest years and in fruits grown in different Australian locations. With the definition of four different masses for PR-5 proteins (range = 21,239--21,272 Da) and nine different masses of PR-3 proteins (range = 25,330--25,631 Da) and using statistical analysis, the methods developed could be used for varietal differentiation of grapes grown in several South Australian locations on the basis of the PR protein composition of the juice. It remains to be seen whether this technology can be extended to grapes grown worldwide and to wine and other fruit-derived products to assist with label integrity to the benefit of consumers. 相似文献

Thaumatin-like proteins and chitinases, which are pathogenesis-related (PR) proteins, were the major soluble protein components of grapes from five cultivars of Vitis vinifera. This dominance of PR proteins was apparent at berry softening (véraison) and then throughout berry development for the Muscat of Alexandria, Sultana, and Shiraz cultivars and in the berries of the Sauvignon Blanc and Pinot Noir cultivars examined at commercial maturity. The M(r) of the major thaumatin-like protein from Muscat of Alexandria grapes was 21 272, and those of the three major chitinases from this cultivar, ChitB, ChitC, and ChitD, were 25 588, 25 410, and 25 457, respectively. The vines in the study were irrigated and showed no obvious signs of disease. Shiraz vines that had not been irrigated throughout the season were clearly water stressed, but had levels of PR proteins in the berry similar to vines that had been fully irrigated. It appears that the production of PR proteins that cause protein instability in wines by grapes may be little influenced by environmental conditions. 相似文献

Little information currently exists regarding the risk of low-back disorders among youth who perform physically demanding farm activities. Thus, afield study was conducted in which children and adolescents who engage in farm work were recruited, fitted with a lumbar motion monitoring system, and then observed performing their usual chores. The lumbar motion monitor was used to record the trunk movements required while youth were performing routine manual material handling tasks on a farm. Workplace factors and motions from both males and females were recorded on over 40 farm tasks, such as feeding animals, lifting bales of hay and straw, and other miscellaneous farm chores. Although the sample size and number of observations in this study were small, the results showed that the magnitude of several work-related factors, such as weight and horizontal moment arm, and trunk motions for many farm activities were equal to or greater than those associated with high injury risk jobs previously assessed in industrial workplaces. In this study, we quantified the physical demands of tasks performed by children and adolescents on farms. In addition, the specific farm chores more likely to load the spines of youth and thereby contribute to musculoskeletal injury were identified. 相似文献

Protein haze formation in white wine is dependent on the presence of both wine protein and other unknown wine components, termed factor(s) X. The ability to reconstitute protein haze upon heating artificial model wine solutions (500 mg/L thaumatin, 12% ethanol, 4 g/L tartaric acid) to which candidate components were added was employed to identify factor(s) X. No protein haze was formed in the absence of additives. The individual or combined addition of caffeic acid, caftaric acid, epicatechin, epigallocatechin-O-gallate, gallic acid, or ferulic acid at typical white wine concentrations did not generate protein haze. However, PVPP fining of commercial wines resulted in a reduction in protein haze, suggesting that phenolic compounds may play a modulating role in haze formation. To elucidate the nature of the unknown factor(s) wine was fractionated and fractions were back-added to model wine and tested for their essentiality. Wine fractions were generated by ultrafiltration, reverse-phase chromatography, and mixed-mode anion-exchange and reverse-phase chromatography. The only purified fraction containing the essential component(s) was free of phenolic compounds, and analysis by mass spectrometry identified sulfate anion as the dominant component. Reconstitution with KHSO4 using either commercially available thaumatin or wine proteins confirmed the role of sulfate in wine protein haze formation. The two main wine proteins, thaumatin-like protein and chitinase, differed in their haze response in model wines containing sulfate. Other common wine anions, acetate, chloride, citrate, phosphate, and tartrate, and wine cations, Fe(2+/3+) and Cu(+/2+), when added at typical white wine concentrations were not found to be essential for protein haze formation. 相似文献

Long-term culture of bone marrow derived stromal colony forming cells (S-CFC) in matrix and nutrient defined agar medium resulted in stromal cell colonies that pass sequentially through three distinct morphological stages: firstly, aggregated loose syncytium of round to ovoid cells (stage I), a second developmental stage of large branching colonies in which the cells become enlarged, elongated with cytoplasmic projections forming a loosely anastomized network with adjacent cells (stage II), and finally cells become dissociated, loosing their long, thin cytoplasmic filaments and breaking their contacts with one another, but remain large and retain a bi-polar nature (stage III). Cells were also grown in liquid medium in a culture microenvironment closely resembling conditions of haemopoiesis in vitro. Using a panel of well defined monoclonal antibodies reactive against the rat, rabbit and human growth hormone receptors, this study found immunochemical evidence of the presence and localization of binding sites of growth hormone (GH) in the cell membrane and extra-nuclear Golgi area of long-term bone marrow derived human stromal cells in liquid and semi-solid nutrient agar mediums. GH-receptor immunoreactivity was present in small proliferating progenitor cells, myofibroblast-like cells, large reticular fibroblast cells, adipocytes and endothelial cells. Only MAb known to be reactive against human tissue resulted in strong immunoreactivity. The expression of GH-receptors not only on small proliferating, but also on the well differentiated cells, indicates a role for growth hormone on non-progenitor cells. GH-receptor immunoreactivity on differentiating and/or differentiated cells suggests that GH is also necessary for, or has a trophic function in differentiation. We propose that direct GH action is necessary not only for differentiation of progenitor cells as implied by the dual effector hypothesis, but also their subsequent clonal expansion, differentiation and maintenance. 相似文献

The Nimbus 5 microwave spectrometer has been used to measure thermal radiation in five frequency bands between 22.235 and 58.8 gigahertz, and has yielded both the temperature profile and, over ocean, the vapor and liquid water content of the terrestrial atmosphere, even in overcast conditions. Information has also been obtained on geophysical parameters that affect the surface emissivity, such as ice type, sea roughness, and snow cover. The experiment demonstrates the considerable potential of passive microwave sensing of meteorological and geophysical parameters. 相似文献
