Performance Evaluation of the ADS 1000 Ventilator Using a Lung Model

A laboratory evaluation was performed to evaluate the performance characteristics of a new veterinary ventilator. The ventilator studied was configured according to manufacturer's directions and attached to a test lung via a pneumotachograph and differential pressure transducer interfaced to a pulmonary mechanics analyzer system. Constant resistance (R=10 cm H₂O/L/sec) and compliance (C=3 ml/cm H₂O) factors were maintained for all trials. The ventilator operated at the manufacturer's preprogrammed parameters. In the first trial, body weight was the only variable. In the second trial, an endotracheal tube was placed in series between the ventilator's breathing circuit and the pneumotachograph. Body weights from 1–20 kgs were evaluated. Mean values for respiratory rate (RR), minute ventilation (VE), inspiratory time (Ti), peak inspiratory pressure (PIP), and peak inspiratory flow (Fpki) displayed on the ventilator control panel; tidal volume (VT), calculated from the displayed minute volume, and identical parameters measured by the pulmonary mechanics system at each body weight, were compared using a two factor analysis of variance. Significant differences (P< 0.05) were found between mean displayed and measured values for RR, PIP, and Fpki.     

Glycine max and Glycine soja are capable of cold acclimation

Soybean has been considered a cold intolerant species; based largely upon seed germination and soil emergent evaluations. This study reports a distinct acquisition of cold tolerance, in seedlings, following short acclimation periods. Diversity in cold responses was assessed in eight cultivars of Glycine max and six accessions of G. soja. All varieties of soybean significantly increased in freezing tolerance following acclimation. This study indicates soybean seedlings are indeed capable of sensing cold and acquiring cold tolerance. Germination rates after cold imbibition were negatively correlated with maturity group, but positively correlated with cold acclimation potential in G. soja. Seed fatty acid composition was varied between the species, with Glycine soja accessions containing about 2‐times more linolenic acid (18:3) than G. max. Furthermore, high levels of linoleic acid (18:2) in seeds were positively correlated with germination rates following cold imbibition in G. soja only. We suggest that domestication has not impacted the overall ability of soybean to cold acclimate at the seedling stage and that there is little variation within the domesticated species for ability to cold acclimate. Thus, this brief comparative study reduces the enthusiasm for the “wild” species as an additional source of genetic diversity for cold tolerance.

Approaches to pathogen-mediated resistance breeding against plum pox potyvirus in stone-fruit trees1

In a programme for developing systems which allow the transfer of foreign genes into apricot cultivars, we have tested cotyledons of immature embryos, somatic embryos and leaf discs. Apricot plants have been transformed, and then regenerated, with Agrobacterium tumefaciens strain LBA 4404 containing various binary plasmids: pBinGUSint, carrying the marker gene β-glucuronidase (GUS), and pBinPPVm, carrying the coat-protein gene of plum pox potyvirus (PPV). The marker gene GUS was used for visual evaluation of the efficiency of the transformation system. The coat-protein gene was used in the hope of introducing coat protein-mediated resistance to one of the most important stone-fruit pathogens in Europe and the Mediterranean area.     

Effect of whole-body vibration in the vertical axis on cortisol and adrenocorticotropic hormone levels in piglets

Vibration, being a consequence of motion during transport, may impair the welfare of pigs. Therefore, the primary objectives of this study were 1) to evaluate during transport simulation the use of ACTH and cortisol plasma levels, which are part of a basic adaptation mechanism of pigs and 2) to define comfort conditions for pigs related to the frequency and acceleration of vibration. Pigs with a body weight between 20 and 25 kg were vibrated in the vertical direction for 2 h at 2, 4, 8, and 18 Hz, in combination with root mean square acceleration magnitudes of 1 or 3 m/s2. Blood was sampled at regular intervals before, during, and after vibration as the pig's behaviors were recorded. Data on ACTH, cortisol, and behavior could be collected from 104 vibrated pigs and 21 controls. In addition, eight animals (3 controls, 5 vibrated) were treated with 0.1 mg of dexamethasone/kg BW, eight animals (3 controls, 8 vibrated) with 0.1 mg naloxone/kg BW, and six (2 controls, 4 vibrated) with a physiological salt solution. Blood samples were taken and products were administrated via an intravenous catheter. The pigs spent less time lying during both hours of vibration treatment than during control conditions. Compared with 2 and 4 Hz, time spent lying was 10 times shorter at 8 Hz and 18 times shorter at 18 Hz. At 1030, ACTH levels were significantly higher than basal levels in animals vibrated at 2 (P < 0.0001), 4 (P < 0.002), and 18 Hz (P < 0.0006). After 1 h, levels returned to basal values. Cortisol levels increased very rapidly after the beginning of vibration (P < 0.0001) and remained higher until 1 h after cessation of vibration (P < 0.003). An inferrence of the lines of equal responses for ACTH and cortisol indicated that, in the beginning of vibration exposure, pigs were extremely susceptible to vibrations at lower frequencies (2 and 4 Hz), whereas at the end of vibration exposure the responses were higher at 18 Hz. The application of dexamethasone and naloxone underpinned the emotional component of the response strategy of pigs to vibration. Hence, vibration during transport should be minimized in order to enhance the adaptive capacities of pigs.
