During the last decade, Piscine orthoreovirus was identified as the main causative agent of heart and skeletal muscle inflammation (HSMI) in Atlantic Salmon, Norway. A recent study showed that PRV-1 sequences from salmonid collected in North Atlantic Pacific Coast (NAPC) grouped separately from the Norwegian sequences found in Atlantic Salmon diagnosed with HSMI. Currently, the routine assay used to screen for PRV-1 in NAPC water and worldwide cannot differentiate between the two groups of PRV-1. Therefore, this study aimed at developing a real-time polymerase chain reaction (RT-qPCR) assay to target the PRV-1 genome segments specific for variants associated with HSMI. The assay was optimized and tested against 71 tissue samples collected from different regions including Norway, Chile and both coast of Canada and different hosts farmed Atlantic Salmon, wild Coho Salmon and escaped Atlantic Salmon collected in British Columbia, West Coast of Canada. This assay has the potential to be used for screening salmonids and non-salmonids that may carry PRV-1 potentially causing HSMI.  相似文献

A bottleneck of crustacean larval culture concerns nutrition and associated cannibalism in communal rearing systems, which impact on larval survival, development and growth. For early‐stage European lobster, Homarus gammarus larvae, feeding ecology and body composition are largely unknown. We initiated four progressive feeding experiments (novel feed types, feeding regime and feed size and cannibalism effects) on growth and survival, to inform and update husbandry protocols. Performance of larvae offered a dry commercial feed was not significantly different compared with a conventional wet plankton feed of the same ration and size grade (both within 600–1,000 μm). Further experiments found that the same ration of dry feed offered six times daily improved development and growth, over the conventional regime of three times daily. Small‐grade dry feed (particles: 250–360 μm) improved larval performance compared with a larger feed (360–650 μm). Larvae were also fed different proportions of dry feed and/or conspecifics in both communal and individual rearing systems (the latter preventing cannibalism via segregation). Individually reared larvae, fed only dead conspecifics, displayed the greatest survival (80%) to postlarvae. This underlines the impact of cannibalism on survival and nutrition in H. gammarus larviculture. A final experiment analysed H. gammarus zoea 1 composition, identifying deficiencies in ash and carbohydrate in lobster feeds. This suggests a need for a species‐specific, formulated dry feed for H. gammarus larviculture. Our research represents the first investigation of H. gammarus larval composition and dietary requirements and highlights decreased growth potential associated with providing nutrition solely from generic commercial feed.  相似文献

A total of 360 bark-to-bark-through-pith wood strips were sampled at breast height from 180 trees in 30 open-pollinated families from two rotation-aged genetic trials to study inheritance, age-age genetic correlation, and early selection efficiency for wood quality traits in radiata pine. Wood strips were evaluated by SilviScan® and annual pattern and genetic parameters for growth, wood density, microfibril angle (MFA), and stiffness (modulus of elasticity: MOE) for early to rotation ages were estimated. Annual ring growth was the largest between ages 2–5 years from pith, and decreased linearly to ages 9–10. Annual growth was similar and consistent at later ages. Wood density was the lowest near the pith, increased steadily to age 11–15 years, then was relatively stable after these ages. MFA was highest (35°) near the pith and reduced to about 10° at age 10–15 years. MFA was almost unchanged at later ages. MOE increased from about 2.5 GPa near the pith to about 20 GPa at ages 11–15 years. MOE was relatively unchanged at later ages. Wood density and MOE were inversely related to MFA. Heritability increased from zero near the pith and stabilised at ages 4 or 5 for all four growth and wood quality traits (DBH, density, MFA and MOE). Across age classes, heritability was the highest for area-weighted density and MFA, lowest for DBH, and intermediate for MOE. Age-age genetic correlations were high for the four traits studied. The genetic correlation reached 0.8 after age 7 for most traits. Early selection for density, MFA and MOE were very effective. Selection at age 7–8 has similar effectiveness as selection conducted at rotation age for MFA and MOE and at least 80% effective for wood density.  相似文献

In experiment 1, juvenile sea urchins (n = 80, 0.088 ± 0.001 g wet weight and 5.72 ± 0.04 mm diameter) were held individually and fed ad libitum one of three semi‐purified formulated diets (n = 16 individuals treatment?1). In the diets, protein was held constant (310 g kg?1 dry, as fed) and carbohydrate level varied (190, 260, or 380 g kg?1 dry, as fed). Wet weights were measured every 2 weeks. Total wet weight gain was inversely proportional to dietary carbohydrate level and energy content of the respective diet. In experiment 2, sea urchins (5.60 ± 0.48 g wet weight, n = 40) fed 190 g kg?1 carbohydrate consumed significantly more dry feed than those fed 260 g kg?1, but not more than those fed 380 g kg?1 carbohydrate. Based on differential feed intake rates, sea urchins that consumed more feed also consumed higher levels of protein and had the highest weight gain. Consequently, protein content and/or protein: energy ratio may be important in determining feed utilization and growth among sea urchins in this study. The average digestible energy intake was approximately 70 kcal kg?1 body weight day?1, suggesting daily caloric intake of juvenile Lytechinus variegatus is lower than in shrimp and fish.  相似文献

One approach to ongrow juvenile European lobster, Homarus gammarus, is to utilize land based rearing systems, incorporating automated feeding, individual culture and provision of stable pelleted feeds, preferably using sustainable ingredients. We initiated three feeding experiments to investigate the general suitability of ingredients produced from seafood by‐products as novel feeds for H. gammarus, in terms of promoting survival, development and growth of post‐larval lobsters from post‐larvae (PL) stage IV to the first juvenile stage (stage V). The first experiment was designed to screen an array of candidate, locally produced, novel protein sources on growth performance parameters. This initial experiment revealed that PL reared on a raw (i.e. wet, unprocessed shrimp) feed used as a reference showed superior performance to those reared on experimental feeds containing fishmeal, herring protein isolate or mussel meal; however, a novel type of shrimp meal, produced by flocculation from waste water, promoted the best PL performance of any experimental feed. A second experiment was designed to test the effect of drying method and to optimize the form of a wet shrimp reference feed used by lobster hatcheries. This showed that the performance of PL reared on experimental freeze‐dried shrimp feed was not significantly different to those reared on the wet, unprocessed shrimp used as a reference feed. However, lobsters offered experimental oven‐dried shrimp feed (with or without an immune supplement) resulted in significantly lower survival or growth performance. A third and final experiment was designed in an attempt to improve a candidate herring‐based protein source, by supplementing with nutrients found in shrimp. However, the results showed that PL reared on the wet reference shrimp feed still showed superior growth and survival than those reared on a herring feed alone, or supplemented with additives found in shrimp meal (either glucosamine, astaxanthin or both supplements combined). The high survival and growth, low incidence of moulting problems and high availability of waste shrimp material, suggest that non‐heat‐treated shrimp products are a promising feed ingredient for post‐larval European lobsters.  相似文献

Samples of inbred lines of Hordeum vulgare produced by pedigree inbreeding, doubled haploidy and single seed descent have been compared for a range of agronomic characters. Differences between the means of the in bred s produced by single seed descent and pedigree breeding were due to differing levels of inbreeding. The present study confirmed chat recombinant inbred families which out-perform the higher scoring parent may be produced by either method. Caution should, however, be exercised when simply inherited characters controlled by major genes are fixed in early generations. Differences between the doubled haploid and single seed descent samples for the character thousand grain weight were attributable to coupling linkages involving epistatic genes. The production and evaluation of random inbred lines should be considered as an alternative to pedigree methods of barley breeding.  相似文献

It is likely that in the near future sequence information from sequencing programmes and EST libraries will generate an abundance of genic microsatellite markers. This study is focused on the assessment of their likely impact and performance vis-à-vis their genomic counterparts. Microsatellites from two sources were used to assess the genetic diversity in 56 old and new varieties of bread wheat on the UK Recommended List. A set of 12 microsatellite markers generated from genomic libraries and 20 expressed sequence tag (EST)-derived microsatellites were used in the study, and the performance of both marker sets assessed. The EST-derived or genic microsatellites delivered fingerprints of superior quality, amplifying clear products with few stutter bands. Diversity levels as revealed bygenic microsatellites are similar to the few published results. The PIC values for the genic markers were generally lower than those calculated for the genomic microsatellites, though advantages of both marker classes for variety identification applications are discussed. Summary The regular meiotic behaviour of the cultivated oat Avena sativa (2n=6x=42) is genetically controlled. The factors which control the diploid-like meiotic behaviour also restrict the amount of pairing that occurs between alien chromosomes and their homoeologues in A.sativa, and hence increases the difficulties of introducing desirable variation from wild species into the cultivated oat. A genotype of the diploid species A.longiglumis which interferes with the regular meiotic behaviour of A. sativa can be used to induce pairing between alien chromosomes and their corresponding chromosomes in A. sativa. Using this procedure the dominant gene conferring mildew resistance has been transferred from the tetraploid weed species A. barbata into the cultivated oat.  相似文献
