Melatonin has been reported to improve the in vitro development of embryos in some species. This study was conducted to investigate the effect of melatonin supplementation during in vitro maturation (IVM) and development culture on the development and quality of porcine embryos. In the first experiment, when the in vitro fertilized embryos were cultured with different concentrations of melatonin (0, 10, 25 and 50 ng/ml) for 8 days, the blastocyst formation rate of embryos cultured with 25 ng/ml melatonin (10.7%) was significantly increased (p < 0.05) compared to the control embryos cultured without melatonin (4.2%). The proportion of DNA‐fragmented nuclei in blastocysts derived from embryos cultured with 50 ng/ml melatonin was significantly lower (p < 0.05) than that of embryos cultured without melatonin (2.1% vs 7.2%). In the second experiment, when oocytes were cultured in the maturation medium supplemented with different concentrations of melatonin (0, 10, 25 and 50 ng/ml), fertilized and then cultured with 25 ng/ml melatonin for 8 days, there were no significant differences in the rates of cleavage and blastocyst formation among the groups. However, the proportions (2.7–5.4%) of DNA‐fragmented nuclei in blastocysts derived from oocytes matured with melatonin were significantly decreased (p < 0.05) compared to those (8.9%) from oocytes matured without melatonin, irrespective of the concentration of melatonin. Our results suggest that supplementation of the culture media with melatonin (25 ng/ml) during IVM and development has beneficial effects on the developmental competence and quality of porcine embryos.  相似文献

Conversion of methane to higher hydrocarbons by its low-temperature activation without forming undesirable carbon oxides is of great scientific and practical importance. Methane can be highly activated, yielding high rates of conversion to higher hydrocarbons and aromatics (10 to 45 percent) at low temperatures (400° to 600°C), by its reaction over H-galloaluminosilicate ZSM-5 type (MFI) zeolite in the presence of alkenes or higher alkanes. The methane activation results from its hydrogen-transfer reaction with alkenes.  相似文献

The essential transition metal ions are avidly accumulated by cells, yet they have two faces: They are put to use as required cofactors, but they also can catalyze cytotoxic reactions. Several families of proteins are emerging that control the activity of intracellular metal ions and help confine them to vital roles. These include integral transmembrane transporters, metalloregulatory sensors, and diffusible cytoplasmic metallochaperone proteins that protect and guide metal ions to targets. It is becoming clear that many of these proteins use atypical coordination chemistry to accomplish their unique goals. The different coordination numbers, types of coordinating residues, and solvent accessibilities of these sites are providing insight into the inorganic chemistry of the cytoplasm.  相似文献

For more than 30 years, the relationship between net primary productivity and species richness has generated intense debate in ecology about the processes regulating local diversity. The original view, which is still widely accepted, holds that the relationship is hump-shaped, with richness first rising and then declining with increasing productivity. Although recent meta-analyses questioned the generality of hump-shaped patterns, these syntheses have been criticized for failing to account for methodological differences among studies. We addressed such concerns by conducting standardized sampling in 48 herbaceous-dominated plant communities on five continents. We found no clear relationship between productivity and fine-scale (meters(-2)) richness within sites, within regions, or across the globe. Ecologists should focus on fresh, mechanistic approaches to understanding the multivariate links between productivity and richness.  相似文献

Feline Chronic Gingivostomatitis Syndrome (FCGS) is a common disease in clinical practice. Among the therapeutic options available, long-acting corticosteroids are frequently used due to their anti-inflammatory and immunosuppressive properties. Although they may improve the clinical symptoms, they can lead to a progressive form of the disease that becomes refractory to treatment. Furthermore, their direct relationship with type II diabetes mellitus (DM) is well known. Consequently, these drugs are controversial and not recommended for routine management of FCGS. Recombinant feline interferon-omega (rFeIFN-ω) is an immunomodulatory compound. Recently, its daily oral administration has been shown to be successful in treating refractory cases of FCGS. This case study describes two clinical cases of type II DM complicated by FCGS. Both animals were calicivirus positive and they had been previously treated with long-acting corticosteroids, which may have been the major cause of DM. The two cats were treated with glargine insulin (Lantus, starting dose 1 IU/cat twice daily (BID)), achieving remission 10 and 18 weeks later respectively. Considering the difficulty with control of FCGS in these animals, an oral daily dose of rFeIFN-ω was started as an alternative to long-acting corticosteroids. In both cats oral clinical signs gradually improved and 60 days after the start of therapy the owners reported a significant relief of pain during mastication. According to the authors’ knowledge, this is the first case report that describes the successful use of rFeIFN-ω in the management of FCGS in type II diabetic cats, in which long-acting corticosteroids are contraindicated.  相似文献

本研究以人参愈伤悬浮细胞为材料,在其生长的第28天添加1×10^-3mg/L水杨酸,测定水杨酸添加后,过氧化物酶、多酚氧化酶和苯丙氨酸解氨酶等3种酶在72h内的变化及皂苷含量,结果表明:水杨酸添加后对过氧化物酶和苯丙氨酸解氨酶的活力影响最大,分别在24h和48h达到最大峰值,在18h开始影响多酚氧化酶的活力,培养物生长的第28天添加水杨酸可以明显提高人参愈伤组织中皂苷的合成。确定添加水杨酸后24h提取总RNA,进行cDNA-RDA分析,筛选差异片段。确定差异基因并在GenBank中注册,注册号为FE900130。为探讨水杨酸作为诱导子对人参次生代谢的影响奠定基础。  相似文献

The MerR protein mediates the induction of the mercury resistance phenotype in bacteria; it has been isolated in order to study the effects of metal-ion induced changes in the metabolism of prokaryotic cells at the molecular level. After DNA sequences responsible for negative autoregulation were removed, the 16-kilodalton protein was overproduced and purified to more than 90 percent homogeneity by a salt extraction procedure that yields about 5 milligrams of protein per gram of cells. Complementation data, amino terminal analysis, gel filtration, and deoxyribonuclease I protection studies demonstrate that the purified merR gene product is a dimer under nondenaturing conditions and that it binds specifically to DNA, in the presence and absence of mercury, at a palindromic site which is directly between the -10 and -35 regions of the structural genes and adjacent to its own promoter. These initial results indicate that MerR is a DNA-binding metalloregulatory protein that plays a central role in this heavy metal responsive system and they delineate an operator site in the mer operon.  相似文献

Anti‐microbial resistance is an emerging public health issue. Farmed animals may act as reservoirs and potential sources of anti‐microbial resistant Campylobacters. The aim of this study was to investigate the anti‐microbial resistance profile of cattle and environmental Campylobacter isolates from normal untreated feedlot cattle, the role of the gyrA Thr‐86‐Ile mutation in ciprofloxacin‐resistant Campylobacter jejuni isolates and the involvement of the tripartite CmeABC efflux system for multi‐resistant C. jejuni isolates. The phenotypic anti‐microbial resistance testing was carried out on 500 Campylobacter isolates (445 cattle isolates and 55 environmental isolates). In general, there was a higher level of anti‐microbial resistance for the environmental isolates compared with the animal isolates, 45% of the animal isolates were resistant to one or more of the seven anti‐microbials compared with 84% of the environmental isolates. The combined cattle and environmental Campylobacters had 34 (6.8%) isolates resistant to three or more of the seven anti‐microbials tested on all isolates and 11 (2.2%) isolates were resistant to the seven anti‐microbials. There was a substantial level of ciprofloxacin‐resistant Campylobacters in both animal (8.5%) and environmental (21.8%) isolates. The gyrA Thr‐86‐Ile mutation was only present in five of 22 ciprofloxacin‐resistant C. jejuni isolates investigated. No multi‐drug‐resistant associated mutation was detected in the CmeB or the CmeR regions investigated. In conclusion, our study observed a substantial level of Campylobacter anti‐microbial resistance, highlighting the need for an active anti‐microbial surveillance program for food animals in Ireland and the importance of the chosen sampling point can have on the findings of such a program.  相似文献

  • 1. The natural stock of large woody debris (LWD) in the afforested Douglas River (Fermoy, Co. Cork) is very low relative to old‐growth forests, which seems to arise from deficiency both of supply and retention. Woody debris is important to the ecology and physical structure of forest streams, so its abundance is relevant to aquatic conservation and the maintenance and size of salmonid fish stocks.
  • 2. The physical characteristics and fish stocks of 16 contiguous segments of two 200 m stream reaches were surveyed in spring 1998 prior to the installation of 12 partially spanning debris structures on four of the segments. This study investigated the effect of debris structures on the heterogeneity of flow and substratum, and the distribution of brown trout (Salmo trutta), and assessed the potential use of woody debris manipulation as a tool in the management of forest streams.
  • 3. Surveys of stream habitat conditions over a 2 year period following the installation of woody debris showed a change in stream architecture. This created more suitable habitat for trout through development of additional pools in which beds of fine sediment developed, and constraining the main current, increasing the amount of eddies and slack water areas.
  • 4. There were significant increases in trout density and biomass in the debris segments relative to control segments without debris dams 1 and 2 years after debris addition, although trout condition was not modified by the addition of LWD. These results suggest that the addition of woody debris offers a positive and practical management technique for enhancing fish in plantation forest streams.
