Many fish species undergo natural starvation periods. Adaptation to starvation is possible through the activation of behavioral, biochemical and physiological mechanisms. Knowledge of the effect of dietary nutrients on the intermediary metabolism during starvation and refeeding can be useful to improve fish health and optimize aquaculture production. To analyze the effect of dietary nutrients on liver metabolism of Siberian sturgeon (Acipenser baerii) submitted to starvation and refeeding, four isoenergetic diets differing in nutrient composition were designed: LP-St (38 % protein, 12 % lipid, 36 % carbohydrate), HP-St (44 % protein, 10 % lipid, 30 % carbohydrate), LP-L (38 % protein, 18 % lipid, 25 % carbohydrate) and HP-L (44 % protein, 16 % lipid, 22 % carbohydrate). Four groups of fish were fed 3 weeks to satiety with the corresponding diet, starved for 2 weeks and then refeed 5 weeks to satiety on the same diet. Starvation mobilized the hepatic lipid store to a greater extent than glycogen. Starvation increased superoxide dismutase activity irrespective of the diet, while low protein diets (LP-St and LP-L) increased catalase activity. The oxidative damage decreased after 5 weeks of refeeding. Refeeding the starved fish on the HP-St diet promoted the greatest growth performance. In addition to reporting for the first time the effect of diet composition on growth, liver composition and antioxidant activities in Siberian sturgeon submitted to starvation and refeeding, our findings suggest that refeeding on HP-St diet stimulated the use of dietary carbohydrates and allowed a protein sparing effect in Siberian sturgeon.  相似文献

Heavy metal uptake, translocation and partitioning differ greatly among plant cultivars and plant parts. A pot experiment was conducted to determine the effect of cadmium (Cd) levels (0, 45 and 90 mg kg?1 soil) on dry matter yield, and concentration, uptake and translocation of Cd, Fe, Zn, Mn and Cu in seven rice cultivars. Application of 45 mg Cd kg?1 soil decreased root and shoot dry weight. On average, shoot and root Cd concentrations and uptake increased in all cultivars, but micronutrients uptake decreased following the application of 45 mg Cd kg?1. No significant differences were observed between 45 and 90 mg kg?1 Cd levels. On average, Cd treatments resulted in a decrease in Zn, Fe and Mn concentrations in shoots and Zn, Cu and Mn concentrations in roots. Differences were observed in Cd and micronutrient concentrations and uptake among rice cultivars. Translocation factor, defined as the shoot/root concentration ratio indicated that Cu and Fe contents in roots were higher than in shoots. The Mn concentration was much higher in shoots. Zinc concentrations were almost similar in the two organs of rice at 0 and 45 mg Cd kg?1. A higher Cd level, however, led to a decrease in the Zn concentration in shoots.  相似文献

The use of recombinant gonadotropin-releasing hormone (rGnRH) has very rarely been tested in fish to promote spawning. This study evaluated the impact of recombinant chicken gonadotropin-releasing hormone (rcGnRH) with metoclopramide on the release of sex steroids and final maturation induction in goldfish (Carassius auratus) broodstock. For this purpose, goldfish broodstock was divided into four groups and treated with 0.9% NaCl with 20 mg/kg metoclopramide (Met) (C); 10 μg/kg body weight (BW) rcGnRH with 20 mg/kg metoclopramide (rcGn10); 15 μg/kg BW rcGnRH with 20 mg/kg metoclopramide (rcGn15); and 20 μg/kg BW rcGnRH with 20 mg/kg metoclopramide (rcGn20). The capability of the rcGnRH for eliciting biological response was tested in vivo by evaluating the changes of 17β estradiol (E2), testosterone (T), and 17α, 20β-dihydroxy-4-pregnen-3-one (DHP) and the induced spawning. Blood samples were obtained at 0 h, 12 h, and 24 h after injection. The rcGn10, rcGn15, and rcGn20 treatments induced lower E2 concentration, especially 24 h post-injection. T levels were significantly higher in rcGn10, rcGn15, and rcGn20 treatments 12 h post-injection than at 0 h and then decreased at 24 h post-injection. Furthermore, the rcGnRH tested significantly enhanced DHP secretion in rcGn10, rcGn15, and rcGn20 treatments 12 h post-injection before a decline at 24 h post-injection. No significant difference between the sampling times was found in the C treatment for the 3 sex steroids tested. The results also displayed that rcGnRH at 10–20 µg/kg of body weight can trigger spawning with the highest speed and efficiency of spawning at 20 µg/kg. The obtained results represent a possible strategy for enhancing the artificial reproduction and ovulation of broodstock fish by rGnRH and further support the use of recombinant hormones to promote reproduction in aquaculture.

Background:Azo dyes are the most widely used synthetic colorants in the textile, food, pharmaceutical, cosmetic, and other industries, accounting for nearly 70% of all dyestuffs consumed. Recently, much research attention has been paid to efficient monitoring of these hazardous chemicals and their related metabolites because of their potentially harmful effect on environmental issues. In contrast to the complex and expensive instrumental procedures, the detection system based on the QDs with the superior optochemical properties provides a new era in the pollution sensing and prevention. Methods:We have developed a QD-enzyme hybrid system to probe MR in aqueous solutions using a fluorescence quenching procedure. Results:The azoreductase enzyme catalyzed the reduction of azo group in MR, which can efficiently decrease the FRET between the QDs and MR molecules. The correlation between the QDs photoluminescence recovery and MR enzymatic decolorization at the neutral phosphate buffer permitted the creation of a fluorescence quenching-based sensor. The synthesized biosensor can be used for the accurate detection of MR in a linear calibration over MR concentrations of 5-84 μM, with the LOD of 0.5 μM in response time of three minutes. Conclusion:Our findings revealed that this fluorometric sensor has the potential to be successfully applied for monitoring a wide linear range of MR concentration with the relative standard deviation of 4% rather than the other method. Knowing the effect of dietary nutrients on physiological parameters, especially oxidative stress, during fish rearing can be useful to improve fish health and optimize production in aquaculture. In this study, four iso‐energetic (2 × 2 factorial design) diets differing in nutrient composition were submitted to the Siberian sturgeon juveniles, to analyse biochemical and physiological responses. The juveniles were fed for as long as 10 weeks with the following diets: low protein–high carbohydrate (CHO): lipid ratio (LP‐St), high protein–high CHO: Lipid ratio (HP‐St), low protein‐low CHO: Lipid ratio (LP‐L) and high protein‐low CHO: Lipid ratio (HP‐L). It was shown that HP‐L diet significantly induced higher growth performance in the juveniles than LP‐L; however, there was not a significant difference between them with other treatments. HSI was increased by higher dietary CHO: L ratio. The result obviously showed the higher dietary fat led to an increase in sturgeon body fat content. In addition, dietary protein and CHO: lipid ratio significantly affected plasma metabolites (glucose, cortisol, cholesterol and triglyceride) but did not affect IGF‐1 and insulin levels. Based on enzyme activities results, incorporation of high protein (44%) in the diet induced lower amylase activity and carbohydrate digestion in the sturgeon juveniles. Moreover, antioxidant enzyme activities in this species were more influenced by lipid than other nutrients. In conclusion, using appropriate carbohydrates (~30%) in the sturgeon diet helps to spare protein. Furthermore, it is suggested to decrease the amount of lipid in high protein diet, to improve higher sturgeon fillet quality and less oxidative damage in liver.  相似文献

BACKGROUND: Anopheles culicifacies is a main malaria vector in southeastern part of Iran, bordring Afghanistan and Pakistan. So far, resistance to DDT, dieldrin, malathion and partial tolerance to pyrethroids has been reported in An. stephensi, but nothing confirmed on resistance status of An. culicifacies in Iran. METHODS: In current study, along with WHO routine susceptibility test with DDT (4%), dieldrin (0.4%), malathion (5%), permethrin (0.25%), lambadacyhalothrin (0.1%), and deltamethrin 0.025, we cloned and sequenced segment VI of domain II (SII6) in voltage-gated sodium channel (vgsc) gene of An. culicifacies specimens collected in Sistan and Baluchistan province (Iran). RESULTS: A 221-bp amplified fragment showed 91% and 93% similarity with exon I and exon II of An. gambiae. The size of intron II in An. culicifacies is 62 bp, while in An. gambiae is 57 bp. The major difference within An. culicifacies specimens and also with An. gambiae is in position 29 of exon I, which led to substitution of Leu to His amino acid. CONCLUSION: This data will act as first report on partial sequence of vgsc gene and its polymorphism in An. culicifacies. A Leu to His amino acid substitution detected upstream the formerly known knockdown resistance (kdr) mutation site could be an indication for other possible mutations related to insecticide resistance. However, the result of WHO susceptibility test carried out in Baluchistan of Iran revealed a level of tolerance to DDT and dieldrin, but almost complete susceptibility to pyrethroids in An. culicifacies. We postulate that the molecular diagnostic tool developed for detection and identification of kdr-related mutations in An. culicifacies, could be useful in monitoring insecticide resistance in Iran and neighbouring countries such as Pakistan and Afghanistan. A phylogenetic tree also constructed based on the sequence of exon I and II, which readily separated An. culicifacies populations from An. stephensi, An. fluviatilis and An. gambiae.  相似文献

In this study, a new approach consisting of chemical treatment steps followed by electrospinning process was applied to produce cellulose nanofibers from wheat straws. Wheat straws were initially pretreated by NaOH solution to open the complex structure of raw materials and remove non-cellulosic materials. Then, acid and alkali hydrolysis was separately performed to eliminate hemicellulose and soluble lignin. Also, bleaching processes were implemented to remove the insoluble lignin. Cellulose nanofibers were produced by electrospinning of various concentrations of cellulose in different solvents including sodium hydroxide/urea/thiourea, pure trifluoroacetic acid (TFA), and TFA/methylene chloride. Images obtained by Scanning Electron Microscope (SEM) showed long and uniform nanofibers produced from electrospinning of cellulose/TFA/methylene chloride solution. An epoxy based laminated composite was prepared by a lamina of cellulose microfiber and electrospun nanofiber mat using hand lay-up composite manufacturing method. The fracture surface of the epoxy nanocomposite was analyzed by SEM images. In addition, the mechanical properties of laminated epoxy composites were compared with pure epoxy by conducting tensile and impact tests. Tensile test results showed that the ultimate tensile strength, elongation, and modulus of laminated epoxy nanocomposites were significantly increased. Moreover, it was found that by adding a nanofiber lamina in the epoxy composite, the impact resistance was significantly improved as a result of crack growth prevention.  相似文献

The cotton aphid, Aphis gossypii Glover (Aphididae) is an important pest of indoor and outdoor cucumber crops in Iran. We studied the life history performance of A. gossypii on seven cucumber cultivars (Negin, Pierro, Davos II, Royal safa, Dominus (Ps), Super pretty and TN-94-203) at 27 ± 1 ºC, 50% ± 10% RH and a photoperiod of 16:8 (L:D) h. The developmental time for the nymphal stages ranged from 4.27 days on Pierro to 4.47 days on Negin, but differences among cultivars were not statistically significant. The lowest and highest survival rates for immature stages were 68.33% and 95% on Royal safa and TN-94-203, respectively. The total fecundity of A. gossypii significantly differed among cultivars, being the highest on Pierro (44.66 nymphs/female) and the lowest on Dominus (Ps) (69.89 nymphs/female). The lowest intrinsic rate of natural increase (rm) was obtained on Negin, and the highest on Super pretty. The generation time (T) was shortest on Pierro (7.41 days) and longest on Negin (8.32 days). According to our results, Super pretty and Negin were partially susceptible and resistant to A. gossypii, respectively. The findings of this study provide direction to design a more comprehensive integrated pest management program for this pest.  相似文献
