Catecholamines signal through the beta2-adrenergic receptor by promoting production of the second messenger adenosine 3',5'-monophosphate (cAMP). The magnitude of this signal is restricted by desensitization of the receptors through their binding to beta-arrestins and by cAMP degradation by phosphodiesterase (PDE) enzymes. We show that beta-arrestins coordinate both processes by recruiting PDEs to activated beta2-adrenergic receptors in the plasma membrane of mammalian cells. In doing so, the beta-arrestins limit activation of membrane-associated cAMP-activated protein kinase by simultaneously slowing the rate of cAMP production through receptor desensitization and increasing the rate of its degradation at the membrane.  相似文献

The study was performed on the area of the three countries involved in the MAGPROX project (Poland, Czech Rep., Germany). In the first stage the basic map of magnetic susceptibility based on the field topsoil measurement was compiled with an average grid density of 10 km. In the second stage of the study, vertical distribution of magnetic susceptibility of ca. 600 topsoil cores (0.3 m long), collected over central Europe, was examined with respect to the anthropogenic or lithogenic influence on magnetic susceptibility of the soil surface, with the focus being on the interpretation of the areas showing high surface susceptibility values. The maximum values were mostly observed in depths of 3–4 cm. In general seven different types of susceptibility profiles from forest areas and one typical for urban soil are presented. They are a result of a combination of natural (litho- and pedogenic) and anthropogenic contributions. Our results prove that soil profiles, dominated by anthropogenic influence due to atmospherically deposited dust, are characterized by magnetically enhanced humuous layer, corresponding to layer with the highest concentration of heavy metals. Moreover, forest soils show in these cases higher magnetic susceptibility than soils in open areas (grassland, meadows or arable soils).  相似文献

During this study, we investigated the mineralogical characterization of technogenic magnetic particles (TMPs) contained in alkaline industrial dust and fly ash emitted by coal burning power plants and cement plants. The reaction of tested dust samples varied between values of pH 8 and pH 12. Their magnetic properties were characterized by measurement of magnetic susceptibility (χ), frequency dependence of magnetic susceptibility (χfd), and temperature dependence of magnetic susceptibility. Mineralogical and geochemical analyses included scanning electron microscopy with energy dispersive spectroscopy, microprobe analysis and X-ray diffraction. The TMPs in fly ash from hard coal combustion have the form of typical magnetic spherules with a smooth or corrugated surface as well as a skeletal morphology, composed of iron oxides (magnetite, maghemite, and magnesioferrite) that occurred in the form of incrustation on the surface of mullite, amorphous silica, or aluminosilicate particles. The TMPs observed in fly ash from lignite combustion have a similar morphological form but a different mineralogical composition. Instead of magnetite and magnesioferrite, maghemite and hematite with lower χ values were the prevailing magnetic minerals, which explains the much lower magnetic susceptibility of this kind of ash in comparison with the ash from hard coal combustion, and probably results from the lower temperature of lignite combustion. Morphology and mineralogical composition of TMPs in cement dust is more diverse. The magnetic fraction of cement dust occurs mostly in the form of angular and octahedral grains of a significantly finer granulation (<20 μm); however, spherules are also present. A very characteristic magnetic form for cement dust is calcium ferrite (CaFe3O5). The greatest impact on the magnetic susceptibility of cement dust results from iron-bearing additives (often waste materials from other branches of industry), which should be considered the most dangerous to the environment. Stoichiometric analysis of micro-particles confirmed the presence of heavy metals such as Pb, Mn, Cd, and Zn connected with TMPs, which are carriers of magnetic signals in atmospheric dust. Therefore, in some cases, their presence in topsoil when detected by magnetic measurement can be treated as an indicator of inorganic soil contamination.  相似文献

The stromal microenvironment of tumors, which is a mixture of hematopoietic and mesenchymal cells, suppresses immune control of tumor growth. A stromal cell type that was first identified in human cancers expresses fibroblast activation protein-α (FAP). We created a transgenic mouse in which FAP-expressing cells can be ablated. Depletion of FAP-expressing cells, which made up only 2% of all tumor cells in established Lewis lung carcinomas, caused rapid hypoxic necrosis of both cancer and stromal cells in immunogenic tumors by a process involving interferon-γ and tumor necrosis factor-α. Depleting FAP-expressing cells in a subcutaneous model of pancreatic ductal adenocarcinoma also permitted immunological control of growth. Therefore, FAP-expressing cells are a nonredundant, immune-suppressive component of the tumor microenvironment.  相似文献

Lake Zywiec is a man-made lake constructed in 1965 on the Sola River, Beskidy Mountains (South Poland). The lake is the largest one in a flood control reservoir system known as the Sola Cascade. In May and September 1970, separated by a big flood event in July, the lake was sampled for rock magnetic, chemical and granulometric studies. In 85% of the September samples, susceptibility decreases by 10 to 40% compared to the value measured in May. Seasonal changes are also observed in the total Fe content and some heavy metals. A general decrease in Zn, Pb, Cd, Cu and Cr content during the summer season was observed in the whole lake. The content of other metals such as Mn and Ni increased in samples taken in September. The changes observed in the lake sediments could be only an accidental result of the flood or may indeed result from seasonal changes fostered by the flood effect.  相似文献
