Sequencing of partial cox2 (part of the mitochondrial cytochrome-c-oxydase (COX) gene) was performed with samples from the oomycete genus Pustula, the white blister rusts of Asteraceae and related families. Sequence comparison uncovered several single nucleotide polymorphisms (SNPs) between P. spinulosa and host specific strains of Pustula isolated from Senecio vulgaris, Tragopogon pratensis and cultivated sunflower, Helianthus annuus. Based on these differences, specific primers were designed for PCR-based detection of white blister rust strains pathogenic to sunflower. The specificity of the primers was confirmed by cross testing with DNA from various oomycetes occurring in the same locality. The limit of detection for DNA of P. helianthicola was 10 pg. This allowed detection with DNA from single sporangia and single oospores. The PCR-based experiments allowed detection of the presence of sunflower white blister rust in soil samples from fields on which infected plants had been cultivated several months before. Moreover, the molecular tools were successfully applied to trace the pathogen in asymptomatic tissue of infected plants, demonstrating the systemic nature of Pustula on sunflower.  相似文献

Leaves of Xanthium strumarium infected with downy mildew were collected in the vicinity of a sunflower field in southern Hungary in 2003. Based on phenotypic characteristics of sporangiophores, sporangia and oospores as well as host preference the pathogen was classified as Plasmopara angustiterminalis. Additional phenotypic characters were investigated such as the size of sporangia, the number of zoospores per sporangium and the time-course of their release. Infection studies revealed infectivity of the P. angustiterminalis isolate to both X. strumarium and Helianthus annuus. Inoculation of the sunflower inbred line, HA-335 with resistance to all known P. halstedii pathotypes, resulted in profuse sporulation on cotyledons and formation of oospores in the bases of hypocotyls. Infections of sunflower differential lines often led to damping-off. Molecular genetic analysis using simple sequence repeat primers and nuclear rDNA sequences revealed clear differences to Plasmopara halstedii, the downy mildew pathogen of sunflower.  相似文献

Bioassays testing the fungicide sensitivity against metalaxyl of Peronospora tabacina isolates collected in German tobacco fields in 2005 revealed the presence of two phenotypes, resistant and sensitive. DNA fingerprints using SSR and minisatellite primers allowed separation of the samples into two groups. The differences in amplification patterns coincided with the sensitive and resistant reaction of the isolates in metalaxyl bioassays. New primers were developed which allowed PCR-based detection of P. tabacina and differentiation of the metalaxyl-sensitive and the metalaxyl-resistant phenotype, respectively. Screening of recent blue mold isolates from Germany and other European countries for metalaxyl sensitivity with leaf disk bioassays coincided completely with the PCR-based identification of the two phenotypes. In Germany, exclusively resistant isolates were found between 2002 and 2004. These still dominate. Since 2005 the co-occurrence of sensitive isolates has been shown. No similar monitoring has been done in any other European country. However, we found the resistant phenotype reaction in a French isolate of 2004 and in two out of three Italian isolates in 2007. Two isolates from Poland and Bulgaria in 2007 were sensitive to metalaxyl. For all 58 isolates tested since 2002 the metalaxyl bioassay-based resistance type and the PCR-based tests for sensitive and resistant genotype coincided. Using genotype-specific primers for future population studies may help to trace the sources of the pathogen from which yearly propagation starts.  相似文献

露天煤矿区是人类活动强扰动地区之一。该文以阿巴拉契亚煤田区韦恩县为研究区域,应用遥感时序分析法分析了像元尺度的土地损毁和复垦过程特征。得出结论:1984-2010年间,韦兹县露天开采扰动区域占采矿权范围的45.80%,其中植被恢复区域占开采范围的66.45%,开采时间越早,植被恢复像元比例越高;开采造成的地表无植被覆盖期时长中位数为6 a,均值为7 a;已充分复垦的区域,NDVI值恢复至采前水平的加权平均时长为12 a。基于像元变化轨迹的研究,除揭示土地损毁-复垦过程特征外,能较好地反映空间异质性,可以为土地复垦管理和相关政策决策提供科学依据。  相似文献

Competitive effects of dense herbaceous vegetation (ground cover) can inhibit forest restoration on mine sites. Here we review the evidence of ground cover interactions with planted tree seedlings on coal surface mines of the eastern US, discuss recent research into these interactions, and draw conclusions concerning ground cover management when restoring forests on reclaimed coal mines. Reclaimed mine sites have a high potential to support productive forests, however forest establishment is inhibited by reclamation practices that included soil compaction, and the seeding of competitive ground covers. In the first few years after tree planting, a dense ground cover of grass and legume species commonly seeded on mine sites often affect growth and survival negatively. Herbaceous vegetation providing less extensive and competitive ground coverage may either facilitate or inhibit tree establishment, depending on site conditions. The use of quality planting stock promotes the competitive ability of seedlings by improving nutrient status and the ability to capture available resources. Herbaceous species have contrasting functional characteristics, and thus compete differently with trees for available resources. Negative interactions with trees are more frequently reported for non-native cool-season grasses than for native warm-season grasses, while the effects of legumes change over time. Further research is needed to fully understand the mechanisms of tree/ground cover interactions. The development of seeding mixes that can control erosion, facilitate survival and growth of planted trees, and allow establishment by unplanted native species would aid forest restoration on eastern US, coal mines.
