Blood samples from 78 cattle were tested for hemoplasma infection using molecular methods. PCR and sequence analysis revealed that 17 cattle were infected with Mycoplasma wenyonii, while 13 were infected with 'Candidatus Mycoplasma haemobos'. Four animals were infected with both species. This is the first study to report hemoplasma species infection among cattle in Japan.  相似文献

Localization of leptin and leptin receptor in the bovine adenohypophysis   总被引:1,自引:0,他引:1  
The present study was carried out to detail the cellular localization of leptin (Lep) and the leptin receptor (LepR) in the bovine adenohypophysis. Lep immunoreactivity (Lep-ir) was found in about 30% of adenohypophysial cells in the gland. Immunochemistry of Lep and specific hormones using serial sections revealed that Lep-ir was present in 60.4% of somatotrophs, 15.9% of gonadotrophs, 6.5% of mammotrophs, 6.5% of thyrotrophs and 2.4% of corticotrophs. Both the common short isoform (OBRa) and the long isoform (OBRb) of LepR mRNA were expressed in the bovine adenohypophysis. LepR immunoreactivity (LepR-ir) was found in only 2.8% of the adenohypophysial cells and over 50% of LepR-ir cells were gonadotrophs, in which most of the cells were distributed in the zona tuberalis. The findings on Lep and LepR in the adenohypophysial cells indicate that Lep may regulate gonadotroph function through autocrine/paracrine pathway in the bovine adenohypophysis.  相似文献

A class III chitinase gene (CHI2) is induced in cucumber plants (Cucumis sativa L.) in response to infection by pathogenic microorganisms. Infection of Botrytis cinerea, causal agent of gray mold disease on cucumber, also induces CHI2 expression. To investigate whether CHI2 is involved in resistance to gray mold disease, transgenic cucumber plants were produced to overexpress the CHI2 gene. One line was analyzed in detail in terms of disease resistance. The transgenic cucumber plant (CC2) constitutively expressed CHI2 and reduced the symptoms of B. cinerea for 4 days after inoculation compared with nontransgenic plants. However, this inhibitory effect was not absolute, and CC2 eventually developed serious disease symptoms. Chitinase activity of the crude extract from CC2 leaves was higher than that from nontransgenic plants. A high-molecular-weight fraction containing CHI2 from CC2 leaves had fungistatic activity against B. cinerea. Interestingly, the low-molecular-weight fraction from CC2 leaves with CHI2 removed also had fungistatic activity against B. cinerea. Not only the introduced chitinase activity but also the endogenous defense reactions activated by overexpression of CHI2 may be involved in the enhanced gray mold disease resistance in CC2.  相似文献

The objectives of this study were to determine Global Positioning System (GPS) positional errors while moving under the forest canopy and to clarify the effects of polyline simplification on area and perimeter estimations. We used the Pathfinder Pro XR and GPSMAP 76S, which are categorized as “high-end mapping” and “general navigation” GPS receivers, respectively. The field tests were conducted in both natural and plantation forests. The results showed that the Pathfinder Pro XR, which has better multipath rejection technology, worked well, especially in the plantation forest under unfavorable conditions of higher stand density. We used analysis of variance to clarify the effects of the receiver type, positioning mode, stand type, and polyline simplification method on area and perimeter estimations. The receiver type and positioning mode were found to be significant factors that affected area estimation. The Pathfinder Pro XR estimated the area more accurately than the GPSMAP 76S, and differential GPS estimated the area more accurately than autonomous GPS. With respect to the perimeter, the receiver type, positioning mode, and polyline simplification method were found to be significant factors. The results showed that perimeter estimation was improved by using the velocity filter, and further improved by using the velocity filter and Douglas-Peucker algorithm, especially when the Pathfinder Pro XR was used. The GPSMAP 76S estimated the perimeter accurately without any filtering because its default speed filter worked well, even though the GPSMAP 76S is a general navigation GPS receiver.  相似文献

Using attached and detached leaves ofAcer palmatum Thunb. andRhaphiolepsis umbellata Makino, pulse-modulated chlorophyll fluorescence and CO2 exchange were measured. Quantum yield of photosynthesis was determined from the fluorescence parameter(Fm′−Fs)/Fm′, where (Fm′−Fs) was defined as the difference between steady state chlorophyll fluorescence (Fs) and maximum fluorescence (Fm′) elicited by a saturating light pulse. The rate of electron transport through photosystem II (total electron flow) was calculated from the product of quantum yield andA (PFD), whereA is the rate of absorbed photons as given by leaf absorptance, and PFD is the photon flux density at the leaf surface. The rate of electron transport dependant on CO2 uptake (assimilative electron flow) was calculated from the gross photosynthetic rate in a leaf. The difference between the rates of total and assimilative electron transport was denoted as the rate of non-assimilative electron transport which depends on photorespiration and oxygen reduction. Available data provided quantitative information on the rate of non-assimilative electron flow in intact leaves. When leaf photosynthesis ofA. palmatum was measured under sunlight, the rates of total and assimilative electron transport were determined to be approximately 900 and 150 μmol equiv. e/mg Chl·h, respectively. The difference (750 μmol equiv. e/mg Chl·h) was attributed to the activity of non-assimilative electron flow. The ratio of total to assimilative electron flow was found to increase gradually with rising in irradiance. The results suggest that non-assimilative electron flow occurred at much higher rate than assimilative electron flow at high irradiance. Implications of the results are briefly discussed in relation to photosynthesis limitation in tree leaves.  相似文献

We examined the accuracy of the shear properties of wood by the Iosipescu shear test using specimens whose shape was simplified. Quartersawn boards of sitka spruce (Picea sitchensis Carr.) and shioji (Japanese ash,Fraxinus spaethiana Lingelsh.) were used. Two types of specimen for the Iosipescu shear test were compared: a standard specimen whose notch angle is 90° and a keyhole type specimen, which is more easily prepared than the standard type. The shear modulus, yield shear stress, and failure shear stress of the keyhole-type specimen were compared to those of the standard specimen. Shear stress analysis was conducted using the finite element method (FEM). The results obtained were as follows: (1) The failure pattern obtained by the simplified Iosipescu shear test was similar to that seen with the standard Iosipescu shear test. (2) The shear modulus, yield stress, and failure stress obtained by the simplified Iosipescu shear test coincided with those by the standard Iosipescu shear test. (3) The principal strain angle and principal stress angle of the simplified Iosipescu shear test were about 45°. (4) It is recognized that pure stress is applied to the strain-gauge regions in the simplified Iosipescu shear test, and it is expected that the shear properties are independent of the notch angle.  相似文献

Age trends in variance components and heritability of overall wood density, earlywood and latewood density, and latewood proportion were investigated in 29-year-old trees of 19 full-sib families of hybrid larch (Larix gmelinii var. japonica × Larix kaempferi) F1. The age–age correlation and optimum selection age for these traits were also estimated and genetic and phenotypic correlations between wood density and radial growth rate were calculated for each growth ring. Intraring wood density data were obtained using X-ray densitometry. The coefficient of additive genetic variance was stable over all ages, whereas the coefficient of environmental variances gradually decreased with increasing age, resulting in increases in heritability estimates with age for overall density. The latewood proportion had the highest heritability estimates at all ages, ranging from 0.44 to 0.66. Overall density and its various components at 28 years of age showed strong genetic correlations with their respective traits at all younger ages. Optimum selection ages for the wood density traits ranged from 8 to 14 years, at which point maximum gain efficiencies per year were obtained. There were negative correlations between wood density and radial growth rate at early ages, although these relationships tended to be weaker with increasing age. These results suggest that selection at a young age is effective for wood density, but particular care must be taken in selecting trees with an improved radial growth rate because rapid growth will result in a low-density wood product, especially in the early growth period.  相似文献

Summary Electrophoretic variants of peroxidase in mulberry (Morus spp.) were demonstrated by thin-layer gel isoelectric focusing. Of these variants, three isozyme band groups were found to be controlled by codominant alleles at a single locus. The gene symbol Px 1 was given to this locus, with alleles Px 1 1 and Px 1 2 assigned to the A6-A7-A8 and A7-A8-A9 band groups, respectively. The A6-A7-A8-A9 band group proved to be controlled by the Px 1 1 and Px 1 2 heterozygote.Additional experiments showed that among the three banding types, there were no statistically significant differences in leaf blade length, leaf blade width, length-width ratio of leaf blade, internode length, phyllotaxis, leaf shape, tree vigor and resistance to powdery mildew, but there were significant differences in leafstalk length.  相似文献
