Soil and root samples were collected from major tomato growing areas of Ethiopia during the 2012/2013 growing season to identify root-knot nematode problems. DNA-based and isozyme techniques revealed that Meloidogyne incognita and M. javanica were the predominant Meloidogyne species across the sampled areas. The aggressiveness of different populations of these species was assessed on tomato cultivars Marmande and Moneymaker. The two most aggressive populations of each species were selected and further tested on 33 tomato genotypes. The resistance screening and mechanism of resistance was performed after inoculation with 100 freshly hatched (<24 h) second-stage juveniles (J2). Eight weeks after inoculation the number of egg masses produced on each cultivar was assessed. For the resistance mechanism study, J2 penetration and their subsequent development inside the tomato roots were examined at 1, 2, 4 and 6 weeks after inoculation. On both cultivars Marmande and Moneymaker all M. incognita and M. javanica populations formed a high number of egg masses indicating highly aggressive behaviour. Populations from ‘Jittu’ and ‘Babile’ for M. incognita and ‘Jittu’ and ‘Koka’ for M. javanica were selected as most aggressive. None of the 33 tomato genotypes were immune for these M. incognita and M. javanica populations. However, several tomato genotypes were found to have a significant effect on the number of egg masses produced indicating possible resistance. For M. javanica populations there were more plants from cultivars or breeding lines on which no egg masses were found compared to M. incognita populations. The lowest number of egg masses for both populations of M. incognita was produced on cultivars Bridget40, Galilea, and Irma while for M. javanica it was on Assila, Eden, Galilea, Tisey, CLN-2366A, CLN-2366B and CLN-2366C. Tomato genotypes, time (weeks after inoculation) and their interaction were significant sources of variation for J2 penetration and their subsequent development inside the tomato roots. Differential penetration was found in breeding lines such as CLN-2366A, CLN-2366B and CLN-2366C, but many of the selected tomato genotypes resistance for the tested M. incognita and M. javanica populations were expressed by delayed nematode development. Therefore, developing a simple screening technique to be used by local farmers or extension workers is crucial to facilitate selection of a suitable cultivar.  相似文献

Multilayer microencapsulation of fish oil was evaluated by using fish gelatin (treated with or without microbial transglutaminase [MTGase]), chitosan, and maltodextrin for its ability to control lipid oxidation. This study showed that a mixture containing gelatin and maltodextrin obtained the highest percentage of core release. The oxidative stability of the formulated mixtures and the bulk oils was investigated during a period of 60 days. The peroxide value (PV) was assessed as a parameter for primary oxidation products, and p-anisidine (p-AV) and thiobarbituric acid reactive sub-stances (TBARS) was used to analyze secondary oxidation compounds. Observation of oxidation products showed that combinations of gelatin and maltodextrin made by adding MTGase and a mixture of gelatin and chitosan were able to increase the oxidative stability, and increases in PV and p-AV and TBARS were found for all oils.  相似文献

Faba bean gall (FBG) is a devastating disease of faba bean (Vicia faba) in Ethiopia. Studies were undertaken first to compare and contrast similarities between FBG disease symptoms and morphology in Ethiopia with those reported earlier in China and, secondly, to identify definitively the FBG causal agent, previously considered as Olpidium viciae, through molecular studies. Morphological studies confirmed an epibiotic phase of zoosporangia for dispersing zoospores, characteristic of Physoderma but not Olpidium, and did not show critical diagnostic characteristics of Olpidium such as presence of numerous short zoosporangial discharging tubes, or binucleate resting sporangia. Recognizing this epibiotic phase is a foundation for comprehending FBG epidemiology and will allow forecasting of zoospore release to highlight best timings for applications of chemical sprays to reduce reinfection cycles. Sequences of partial ITS1-5.8S-partial ITS2, the 18S-ITS1-5.8S-ITS2-part of 28S rRNA, and LSU (28S rRNA) derived from tissue with symptoms confirmed Physoderma, and not Olpidium, as the causal agent. Sample sequences were either close to Physoderma or the contaminant ascochyta pathogen Didymella. From symptom, morphological, and molecular data, the causal agent of FBG disease in Ethiopia is Physoderma. From observations of symptoms that Physoderma can cause, it was determined that this Physoderma crosses over between different legume host genera (e.g., Vicia, Pisum, Trifolium), highlighting the significant biosecurity risk for countries currently free of FBG.  相似文献

The aim of this study was to investigate the effects of onion on serum uric acid levels and hepatic Xanthine Dehydrogenase/Xanthine Oxidase activities in normal and hyperuricemic rats. Hyperuricemia was induced by intraperitoneal injection of 250 mg kg(-1) potassium oxonate in rats. Oral administration of onion at 3.5 and 7.0 mg kg(-1) day(-1) for 7 days was able to reduce serum uric acid levels in hyperuricemic rats with no significant effects on the level of this compound in the normal animals. In addition, onion when tested in vivo on rat liver homogeneities elicited significant inhibitory actions on the Xanthine Dehydrogenase (XDH) and Xanthine Oxidase (XO) activities. This effect resulted less potent than that of allopurinol. However, the hypouricemic effect observed in the experimental animal did not seem to parallel the change in XDH and XO activities, implying that the onion might be acting via other mechanisms apart from simple inhibition of enzyme activities. Such hypouricemic action and enzyme inhibitory activity of onion makes it a possible alternative for allopurinol, or at least in combination therapy to minimize the side-effects of allopurinol, in particular in long-term application.  相似文献

Twenty-four isolates of Fusarium oxysporum f. sp. ciceris were isolated from wilted chickpea plants obtained from different districts and ‘wilt sickplots’ of central Ethiopia to assess variability in pathogenecity of the populations. Each isolate was tested on 10 different chickpea lines and eight improved chickpea varieties. Isolates showed highly significant variation in wilt severity on the differential lines and improved varieties. Based on the reaction types induced on differential lines, isolates were grouped into four corresponding races. Of the 24 isolates, F13, F20 and F22 were the most virulent. Isolates of race 3 were found in all of the districts and ‘wilt sickplots’ studied. Improved chickpea varieties also showed differential reactions to the isolates. All varieties were resistant to isolates of race 3, while varieties Arerti and DZ-10-4 were resistant to all isolates tested, showing the lowest mean wilt severity. Varieties DZ-10-11 and Maryie were susceptible to isolates F13, F20 and F22 and showed the highest mean wilt severity. Identification of races can be useful in breeding chickpea varieties resistant to wilt. The differential reactions of the improved varieties against different races might be important in managing chickpea wilt through gene deployment.  相似文献

Fusarium basal rot caused by Fusarium oxysporum f.sp. cepae is an economic disease of shallot. Field experiments were conducted at Debre Zeit during 2006 and 2007 cropping seasons to determine effective fungicides and their method of application for the management of fusarium basal rot of shallot. The field was naturally infested with F. oxysporum f. sp. cepae and treatments were arranged in randomized complete block design in four replications. Five fungicides, Mirage 50 WP, Folicur 25 EC, Seed plus 30 WS, Penncozeb 80 WP and Ridomil Gold 68 WG were evaluated as seed bulb dressing and bulb dip treatments against basal rot in the field and storage. Bulb dressing with Mirage, and dip treatment in Seed plus reduced the disease incidence by 40% and 43%, respectively over control. These fungicides also resulted in a significant reduction in severity, basal rot affected cull bulbs on shallot. Bulb rot during three months of storage on concrete ground floor and on wire mesh shelves was also reduced by seed bulb treatment over control. The highest increase in yield was obtained from bulb dressing with Mirage (42%) and Seed plus (45%) and from bulb dip treatment in Seed plus (44%) over control. Fusarium basal rot caused 45% loss in yield and 12-30% of bulb loss in the storage. The study showed that basal rot of shallot can be managed effectively by seed bulb dressing or dip treatment in Mirage or Seed plus.  相似文献
