The present study was aimed to investigate the possible interaction of the standardized extract of Acorus calamus (AC) with Cytochrome P450 enzyme, quantitative determination of the α-asarone in the AC rhizome was performed by RP-HPLC method. In vitro interaction of the plant extract was evaluated by CYP450-carbon monoxide complex (CYP450-CO) assay. Effect on individual isoforms such as CYP3A4 and CYP2D6 isozymes were analyzed through fluorescence product formation and respective IC50 values were determined. CYP450-CO assay showed moderate interaction potential. Extract showed higher IC50 values (46.84 ± 1.83-32.99 ± 2.21 μg/ml) comparing to the standard inhibitors and lower IC50 value than α-asarone (65.16 ± 2.37-42.15 ± 2.45 μg/ml).  相似文献

ABSTRACT

“Lona ilish” is a salt fermented fish product prepared exclusively from fatty Indian shad (Tenualosa ilisha). Despite the presence of salt and metals coming from the container used for fermentation, rancidity is not observed as long as the products are kept immersed in the fermenting brine. To understand the technical principles of this indigenous preservation method, the traditional preparation was followed along with analyses of chemical and microbiological changes. The biochemical and microbiological changes were followed at 15 day intervals during the fermentation period of 150 days. From the 45th day of fermentation onwards, the microbial flora in the product was composed of only two species, tentatively identified as Bacillus licheniformis and Micrococcus kristinae. Because Bacillus licheniformis showed some atypical biochemical reactions, it was tentatively identified as Bacillus licheniformis var. III. These two bacterial species either singly or collectively were involved in the fermentation process.  相似文献

The objectives of the present study were to investigate phytochemical screening and to assay cytotoxicity and antibacterial activities of ethanolic extracts of leaves of two medicinal plants, Aglaonema hookerianum Schott (Family: Araceae) and Lannea grandis Engl. (Family: Anacardiaceae) available in Bangladesh. The brine shrimp lethality bioassay showed that the ethanolic extracts of Aglaonema hookerianum and Lannea grandis possessed cytotoxic activities with LC50 5.25 (microg mL(-1)) and 5.75 (microg mL(-1)) and LC90 10.47 (microg mL(-1)) and 9.55 (microg mL(-1)), respectively. Two extracts obtained from leaves were examined for their antibacterial activities against some gram positive bacteria such as Bacillus subtilis, Bacillus megaterium and Staphylococcus aureus, also gram negative strains of Pseudomonas aeruginosa, Escherichia coli, Shigella dysenteriae, Salmonella typhi, Salmonella paratyphi and Vibrio cholerae. Agar disc diffusion method was applied to observe the antibacterial efficacy of the extracts. Results indicated that both plant extracts (500 microg disc(-1)) displayed antibacterial activity against all of the tested microorganisms. These results were also compared with the zones of inhibition produced by commercially available standard antibiotic, Amoxicillin at concentration of 10 microg disc(-1). Observed antibacterial properties of the ethanolic extract of Aglaonema hookerianum Schott and Lannea grandis Engl. showed that both plants might be useful sources for the development of new potent antibacterial agents.  相似文献

Transition proteins (TNPs) are essential in chromatin condensation during spermiogenesis, and hence, they are the candidate genes for identifying sperm motility markers. Coding and in silico predicted promoter regions of these genes were investigated in crossbred and purebred cattle, and also, their mRNA quantification was done to explore its use as a diagnostic tool of infertility. PCR‐SSCP analysis revealed two band patterns in fragment III of TNP1 and fragment II of TNP2 gene. Sequence analysis revealed a deletion of “G” nucleotide in 3′UTR region of TNP1 and C>T SNP in intronic region of TNP2 gene. Least square analysis of variance did not reveal any significant influence of nucleotide deletion on any sperm motility parameters in both crossbred and purebred cattle. However, C>T SNP had a significant effect on initial progressive motility (p < 0.05) in purebred cattle and post‐thaw motility in overall cattle population. RT‐qPCR analysis did not reveal any significant variation in TNP1 and TNP2 gene expression among poorly motile and good quality spermatozoa of Vrindavani bulls.  相似文献

Ganges River dolphins (Platanista gangetica spp. gangetica) are air-breathing, warm-blooded mammals endemic to the Ganges and Karnaphuli rivers of the Indian subcontinent. Nevertheless, very little basic histomorphological research has been conducted on this endangered species. Therefore, this study aimed to describe the morphological features of different organs of P. gangetica. Despite becoming aquatic animals, they showed similarities with terrestrial mammals, such as the pair of lungs and an apical bronchus in the respiratory system, which are pretty standard in ruminants and pigs. However, unlike the terrestrial animal, the tracheobronchial tree was stiffer due to circularly arranged anastomosing plates of the hyaline cartilaginous ring in the trachea, cartilaginous plates in the bronchiole, and thick alveolar septa. The digestive system showed a three-chambered mechanical and glandular stomach similar to the artiodactyles. However, the intestine showed smaller caecum like the monogastric mammal. The urogenital system showed lobulated kidneys, a urinary bladder, a fibroelastic penis with sigmoid flexure, and a long urethral process similar to some terrestrial ruminants. Considering the aquatic environment, all those modifications, unlike terrestrial mammals, are necessary for their adaptation. Thus, this research will broadly help our clinicians and conservationist to take further steps toward disease diagnosis and monitoring of marine health of this endangered species.  相似文献

Macrophomina phaseolina (Tassi) Goid. is the most important pathogen of jute and primarily causes seedling blight, leaf spot and stem rot. The pathogen was detected from field samples by a simple method of direct PCR (dPCR) which obviates the steps of DNA extraction. The leaf bits were treated with a lysis buffer at 65°C for 25 min, whereas the stem pieces were initially incubated at 65°C for 5 min followed by incubation at 60°C for 25 min and the lysate was used as PCR template. Based on the type of tissue, the composition and concentration of lysis buffer systems were optimized. For leaf samples the optimized buffer system composed of 20 mmol l -1 tris (hydroxymethyl aminomethane (Tris)-Cl (pH 8.0), 1.5 mmol l -1 ethylene diamine tetra acetate (EDTA) (pH 8.0), 1.4 mol l -1 sodium acetate and 200 μg/ml proteinase K. Further, 3% PVP (w/v) and β-mercaptoethanol (1% w/v) were added into the buffer. In case of stem samples, PVP was not applied and higher concentrations were used for other components. M. phaseolina could be detected from both leaf and stem samples generating amplicon of 350 bp. This is the first report of detecting M. phaseolina by a direct PCR method without DNA extraction.  相似文献

Several Ayurvedic plants are known to have activity against diverse urinary crystals. The traditional knowledge of Ayurveda, collective clinical experience in arthritis and the earlier experimental studies on urinary crystals led to the selection of three plants, viz. Rotula aquatica, Commiphora wightii Bhandari syn. C.mukul. and Boerhaavia diffusa for screening anticrystal activity against basic calcium phosphate (BCP), calcium pyrophosphate (CPPD) and monosodium urate monohydrate (MSUM). The effects of each plant were assayed on microcrystals in 24-well microplates in vitro. Our results show that the aqueous extracts of only R. aquatica and C. wightii have shown crystal dissolving activity against MSUM.  相似文献

Beauveria bassiana (Balsamo) Vuillemin (Ascomycota: Hypocreales), an entomopathogenic fungus, was introduced through seed inoculation with spore suspension as an endophyte in white jute (Corchorus capsularis L.), a bast fiber crop. Out of nine B. bassiana strains, seven, viz., ITCC 6063, ITCC 4512, ITCC 4563, ITCC 5562, ITCC 4796, ITCC 5408 and ITCC 4705, became established as endophytes. The endophytic colonization was initially detected by cultivation on selective medium. Colonization was confirmed by polymerase chain reaction (PCR) using sequence characterized amplified region (SCAR) marker. Endophytic B. bassiana strains colonized in leaves of all the plants grown from treated seeds. However, the colonization frequency varied among the strains. The highest colonization frequency (70.09%) was recorded in ITCC 6063 followed by ITCC 5562 (67.67%) and ITCC 5408 (64.17%); ITCC 4512 exhibited the lowest (42.54%) colonization. ITCC 4925 and ITCC 4644 did not show any colonization. Endophytic colonization of B. bassiana reduced stem weevil infestation under pot culture. ITCC 5408 and ITCC 6063 were found most efficient, with only 10.44% and 14.06% infested plants, respectively.  相似文献

Antioxidant activity of piper betel leaf extract and its constituents   总被引:6,自引:0,他引:6  
The 1,1-diphenyl-2-picrylhydrazyl (DPPH) assay of the ethanol extracts of three varieties (Bangla, sweet, and Mysore) of Piper betel (pan) revealed the Bangla variety to possess the best antioxidant activity that can be correlated with the total phenolic content and reducing powers of the respective extracts. Column chromatography of the extract of the Bangla variety led to the isolation of chevibetol (CHV), allylpyrocatechol (APC), and their respective glucosides. The HPTLC analyses of the extracts revealed similar chemical profiles in all three P. betel varieties, although the concentrations of CHV and APC were significantly less in the sweet and Mysore varieties. Among the isolated compounds, APC showed the best results in all the in vitro experiments. It could prevent Fe(II)-induced lipid peroxidation (LPO) of liposomes and rat brain homogenates as well as gamma-ray-induced damage of pBR322 plasmid DNA more efficiently than CHV. The superior anti-LPO and radioprotective activities of APC vis-à-vis those of CHV could not be explained by their respective Fe(II) chelation and .OH radical scavenging capacities. The better ability of APC to scavenge O2-. radicals and H2O2 might account for the results.  相似文献
