The objective of this study was to evaluate the efficiency of Ovum Pick Up (OPU) in cycling (n = 5) and lactating, postpartum, swamp buffaloes (n = 6) with and without gonadotropin stimulation. The OPU was performed every two weeks in all groups of animals, for a total of six sessions. Thirty collections were performed in five cycling buffaloes and 36 collections in six lactating postpartum buffaloes. Buffaloes that received hormonal stimulation were given a total of 400 mg, follicle stimulating hormone (FSH), administered twice daily over 3 days in decreasing doses, together with 100 microg of GnRH, 24 h after the last FSH injection. Following a resting period of 1 month, the two groups of buffaloes, were subjected to the same OPU regimen, but without any hormonal treatment for an additional six OPU sessions. The number of aspirated follicles recorded from the hormonal stimulated, cycling animals and lactating, postpartum buffaloes was not significantly different, 7.2 +/- 3.7 and 9.0 +/- 3.2, respectively (p > 0.05). Recovered oocytes collected from the two groups of hormonally stimulated animals were also not statistically different: 3.7 +/- 2.7 in the cycling and 5.9 +/- 3.5 in the lactating postpartum group (p > 0.05). In the two groups of buffaloes not receiving hormonal stimulation, the number of aspirated follicles was not significantly different: 2.1 +/- 1.4 and 1.4 +/- 0.7 in cycling and lactating postpartum buffaloes respectively (p > 0.05). Recovered oocytes in the non-treated groups were also similar: 1.4 +/- 1.3 vs 0.7 +/- 0.8 in cycling and lactating buffaloes (p > 0.05). Among stimulated buffaloes, most aspirated follicles were small in size (< or =5 mm), whereas they were mostly medium and large sizes in the non-treated buffaloes. The oocyte recovery rate in both the groups, cycling and lactating postpartum, were 51.6% and 69.5% in stimulated groups and 55.0% and 53.1% in non-stimulated groups (p > 0.05). The majority of recovered oocytes were single- and multi-layered, and the number was greater in the cycling than in the lactating, postpartum buffaloes. The number and quality of recovered oocytes was similar in all groups of buffaloes whether they were received or did not receive hormonal stimulation. Moreover no difference was found in multi- and single-layered oocytes between cycling and lactating, postpartum buffaloes. In conclusion, OPU can be performed successfully in swamp buffalo in different reproductive status and FSH administration was shown to increase the number of aspirated oocytes in both cycling and lactating, postpartum buffaloes.  相似文献

The objectives of this study were to compare covariance functions (CF) and estimate the heritability of milk yield from test‐day records among exotic (Saanen, Anglo‐Nubian, Toggenburg and Alpine) and crossbred goats (Thai native and exotic breed), using a random regression model. A total of 1472 records of test‐day milk yield were used, collected from 112 does between 2003 and 2006. CF of the study were Wilmink function, second‐ and third‐order Legendre polynomials, and linear splines 4 knots located at 5, 25, 90 and 155 days in milk (SP25–90) and 5, 35, 95 and 155 of days in milk (SP35–95). Variance components were estimated by restricted maximum likelihood method (REML). Goodness of fit, Akaike information criterion (AIC), percentage of squared bias (PSB), mean square error (MSE), and empirical correlation (RHO) between the observed and predicted values were used to compare models. The results showed that CF had an impact on (co)variance estimation in random regression models (RRM). The RRM with splines 4 knots located at 5, 25, 90 and 155 of days in milk had the lowest AIC, PSB and MSE, and the highest RHO. The heritability estimated throughout lactation obtained with this model ranged from 0.13 to 0.23.  相似文献

The objective of the present study was to retrospectively investigate causes of variation on litter size at birth (total number of piglets born per litter (TB) and number of piglets born alive per litter (BA)) of Landrace (L) and Yorkshire (Y) sows in swine nucleus herds in Thailand. The data included sows farrowed during a four-year period from January 1998 to December 2001. The analyzed data set included observations on 8020 litters from 2199 L sows and 6919 litters from 1680 Y sows. Analysis of variance (ANOVA) was applied for statistical analyses using General Linear Mixed Model (MIXED) procedure of SAS. No breed difference was found for both TB and BA. Farrowing months significantly influenced TB and BA (P<0.001). Sows farrowed in August and September had a lower BA than sows farrowed from November to June (P<0.05). Effect of farrowing months on both TB and BA was more pronounced in primiparous compared with multiparous sows. Average minimum daily temperature during gestation negatively correlated with both TB and BA, average maximum daily temperature during gestation negatively correlated with BA and average daily humidity during gestation negatively correlated with both TB and BA. This study was conducted to compare the developmental competence of somatic nuclear transfer (NT) embryos, after either ionomycin or ethanol activation, in locally bred goats. Donor cells were prepared from the ear skin fibroblasts of a female goat. Cells, at passage 3-8, starved by culturing in 0.5% FCS for 4-8 d, were used for NT. Immature oocytes were obtained from FSH-stimulated goats and matured for 22 hr before enucleation and NT. After fusion, the reconstructed embryos were activated with either ionomycin or ethanol followed by culturing in 6-dimethylaminopurine (6-DMAP) and cytochalasin B (CB), for 3 hr. In experiment I, the fused NT embryos (n=63, ionomycin and n=68, ethanol treatments, respectively) were cultured in B2 with a Vero co-culture system and their developmental competence was evaluated through to Day 9. In experiment II, the NT embryos at the 2-4 cell stage on Day 2 derived from each treatment (ionomycin n=46, and ethanol n=37), were transferred into 10 synchronous recipients. There were no significant differences between the NT embryos derived from the ionomycin and ethanol groups, in fusion (86.3% versus 82.9%), cleavage (90.5% versus 82.4%) and for morula/blastocyst development rates (9.5% versus 5.9%). Sixty percent (3/5) of the recipients from ionomycin became pregnant by midterm (2.5 mts) while only 20% (1/5) from ethanol treatment was pregnant by Day 45. The results demonstrate that activation with either ionomycin or ethanol in combination with 6-DMAP-CB treatment does not affect the development of cloned goat embryos.  相似文献

In 2011, a catastrophic flood disaster in Thailand affected not only humans but also took animal lives. Data on livestock losses, including death, loss, and decreased production, were collected in Nakhon Sawan province. The time-series map of the flooded area from August to December 2011 was available online from the Geo-informatics and Space Technology Development Agency. To evaluate the high-density areas of livestock loss, a spatial hot spot analysis was performed. The Getis-Ord Gi statistic with weighted zone of indifference and the Euclidean distance measurement were employed to identify spatial clusters of species that were affected by the flood. The results indicated that the majority of livestock losses were from poultry and swine farms. The density of poultry and swine loss was significantly different between sub-districts with clusters of high-density loss alongside the river, particularly in Chum Saeng and Kao Liew. Using spatial hot spot analysis as a tool to classify and rank the areas with high flood risks provides an informative outline for farmers to be aware of potential flood damage. To avoid unexpected loss from flooding, poultry and swine farms in risk areas should be properly managed, particularly during the flooding season between August and December.  相似文献

The present experiments were designed to study the effect of adding the detergent Equex-STM® to freezing extender, and of straw volume (0.25 ml vs 0.5 ml), on boar sperm quality after cryopreservation. Three ejaculates from each of four purebred boars (three Landrace and one Yorkshire) were collected and frozen with a lactose-egg yolk extender containing glycerol with or without 1.5% Equex-STM®. The extended semen was loaded into either 0.25- or 0.5-ml straws. The straws were placed in liquid nitrogen (LN2) vapour approximately 3 cm above the level of LN2 for 20 min and then were plunged into LN2. Thawing was achieved in warm water at 50°C for 12 s and then was incubated in a 38°C water-bath for 30 min before evaluating sperm quality. Results showed that the individual motility, viability and acrosomal normal apical ridge (NAR) were improved (p < 0.001) when Equex-STM® was added to the freezing extender. There was no difference (p   =   0.48) in sperm motility between 0.25- and 0.5-ml straws when Equex-STM® was added. The percentages of viable and of NAR sperm in 0.5-ml straws were higher than those in 0.25-ml straws (p   =   0.02, p   =   0.0003 respectively). The percentages of membrane intact sperm evaluated using the short hypo-osmotic swelling test were not affected by straw volume or the adding of Equex-STM® (p   >   0.05). The results of these investigations suggested that Equex-STM® exerts a beneficial effect on the quality of cryopreserved boar semen and this cryopreservation protocol was favourable for a 0.5-ml straw.  相似文献

The present study was conducted to evaluate non-return rate (NR), farrowing rate (FR), and number of total pigs born/litter (TB) of weaned sows after intra-uterine insemination (IUI) using low numbers of frozen–thawed (FT) spermatozoa. Semen from 6 boars was cryopreserved individually in a 0.5-ml straw, at a concentration of 1 × 109 spermatozoa/ml. A total of 40 multiparous sows with weaning-to-estrus interval of 3 to 7 days were included. The sows were detected for standing estrus twice daily and randomly assigned to two groups: I) spontaneous ovulation (n = 20) and II) induced ovulation (n = 20) which the sows were given 750 IU human chorionic gonadotrophin (hCG) i.m. immediately at estrus detection. Ovulation was determined every 12 h using transrectal ultrasonography. FT semen containing 1 × 109 motile spermatozoa/dose was used to IUI. In group I, the sows were inseminated at 24 h after the detection of estrus and repeated every 12 h until ovulation. In group II, the sows were inseminated at 36, 42 and/or 48 h after hCG treatment. The results showed that the interval from standing estrus to ovulation (EOI) differed significantly between group I (40.2 h) and group II (35.6 h; P = 0.01). Variation of EOI among sows within each group seemed to be lower in group II (4.5 h SD) than in group I (5.5 h SD; P = 0.5). The number of IUI per sow was 2.9 ± 0.6 times in group I and was 2.4 ± 0.5 times in group II. There were no significant differences (P > 0.05) in the NR (80 vs 85%), FR (60 vs 65%) and the TB (8.0 ± 2.8 vs 9.4 ± 3.7 piglets/litter) between the groups. These results indicated that multiple doses of IUI with a low number of FT boar spermatozoa provided a fairly good NR, and reasonable FR and TB both in spontaneous and induced ovulating sows. The number of inseminations required for attaining acceptable fertility tended to be lower in the weaned sows with induced ovulation.  相似文献
