Fipronil (FPN) is a systemic insecticide that antagonizes the gamma-aminobutyric acid type A (GABAA) receptors in insects. Recently, adverse effects of FPN on mammals have been reported, but most of those were caused by high doses of FPN and additives in the products. We investigated the effects of low-dose pure FPN on the emotional behavior of mice. Nine-week-old male mice conducted behavioral tests 24 hr after FPN administration by gavage at doses of 0.05 or 5 mg/kg based on the no-observed-effect level (NOEL), showed a significant increase in locomotor activity and dose-dependent responses on the time they spent in the central zone in the open field test. Pure FPN below the NOEL dose may affect the emotional behavior of mice. 相似文献

To increase the sensitivity of an enzyme-linked immunosorbent assay (ELISA) for equine herpesvirus type 4
(EHV-4) that uses a 12-mer peptide of glycoprotein G (gG4-12-mer: MKNNPIYSEGSL) [4], we used a longer peptide consisting of a 24-mer repeat sequence (gG4-24-mer:
MKNNPIYSEGSLMLNVQHDDSIHT) as an antigen. Sera of horses experimentally infected with EHV-4 reacted much more
strongly to the gG4-24-mer peptide than to the gG4-12-mer peptide. We used peptide ELISAs to test paired sera
from horses naturally infected with EHV-4 (n=40). gG4-24-mer ELISA detected 37 positive samples (92.5%),
whereas gG4-12-mer ELISA detected only 28 (70.0%). gG4-24-mer ELISA was much more sensitive than gG4-12-mer

The gene constitution of polymorphisms of the four calpain genes (µ‐calpain, m‐calpain, p94, and µ/m‐calpain) were analyzed in South‐East Asian native chickens, White Leghorn and Broiler commercial chickens, and Red and Green jungle fowl. Polymorphisms were detected at all loci in chickens and Red jungle fowl, but only for CAPN1 (µ‐calpain gene) in Green jungle fowl. CAPN2 and CAPN1.5 are linked on chicken chromosome 3, and the genotype for these loci were treated as haplotype. Some combinations of calpain loci were tested using principal component analysis, and the best combination (CAPN1, CAPN3, and CAPN1.5) was determined. The proportion of polymorphic loci (Ppoly) and heterozygosity (H?) were 1.00 and 0.316–0.465 in domestic chickens and red jungle fowl, and 0.33 and 0.137 in Green jungle fowl, respectively. GST values suggested that the degree of subdivision among native chickens was relatively low except for Thailand, which was highest. Pair‐wise FST testing, dendrogram and principal component analysis from the results of calpain loci showed that the four South‐East Asian native and commercial chicken populations were close genetically. 相似文献

One hundred and twenty
stranding events of Stejneger’s beaked whales were reported in Japan between 1999 and
2011. The purpose of this study is to introduce pathological data and to discuss probable
causes of death for 44 Stejneger’s beaked whales among them. The significant pathological
findings were the pulmonary edema, parasitic granulomatous nephritis, emaciation,
amyloidosis, suppurative bronchopneumonia and so on. The probable causes of death were
categorized as noninfectious in 43 of the cases, which included drowning, starvation and
secondary amyloidosis. One individual was diagnosed with septicemia, which was the only
example of an infectious disease. Because we could not always perform advanced analyses,
such as microbiology tests, biotoxin examinations or contaminant analyses, the finality of
our findings may be impaired. However, the present study has broad implications on the
causes of death of Stejneger’s beaked whales of the seas around Japan, which are valuable
for the future studies and for the detection of emerging diseases. 相似文献

The aim of this study was to evaluate the prognostic value of concurrent measurement of
serum phosphorylated neurofilament heavy subunit (pNF-H) concentration and intramedullary
T2W hyperintensity in paraplegic to paraplegic dogs. Our hypothesis was that concurrent
measurement of these would provide a more accurate prediction of functional outcome in
dogs with thoracolumbar intervertebral disc herniation (IVDH). A prospective case-control
clinical study was designed using 94 dogs with acute onset of thoracolumbar IVDH. The
association of serum pNF-H concentration, T2W hyperintensity on sagittal MRI (T2H/L2),
deep pain perception and surgical outcome were evaluated with logistic regression analysis
after three months for all 94 surgically treated dogs. Sensitivity to predict
non-ambulatory outcome was compared among pNF-H and T2H/L2 and combination of both.
Logistic regression analysis indicated that serum pNF-H concentration and T2H/L2 were
significantly correlated with surgical outcome (P<0.05); however, deep
pain perception was not (P=0.41). The results of logistic regression
analysis indicated that the odds ratios of unsuccessful long-term outcome were 2.6 for
serum pNF-H concentration, 1.9 for T2H/L2 and 2.3 for deep pain sensation. The sensitivity
and specificity to predict non-ambulatory outcome for using serum parameter pNF-H>2.6
ng/ml, using T2H/L2 value of>0.84 and using both
serum pNF-H and T2H/L2, were 95% and 75.7%, 65% and 86.5%, and 90.0% and 97.5%,
respectively. Therefore, combined measurements of serum pNF-H and T2H/L2 might be useful
for predicting long-term outcome in dogs with thoracolumbar IVDH. 相似文献

The time course of GnRH pulse generator activity and plasma concentrations of energy substrates and insulin were simultaneously observed in female goats during 4-day fasting and subsequent refeeding in the presence or absence of estrogen for a better understanding of the mechanism of energetic control of gonadotropin secretion in ruminants. The GnRH pulse generator activity was electrophysiologically assessed with the intervals of characteristic increases in multiple-unit activity (MUA volleys) in the mediobasal hypothalamus. In estradiol-treated ovariectomized (OVX+E2) goats, the MUA volley intervals increased as fasting progressed. Plasma concentrations of non-esterified fatty acid and ketone body increased, while those of acetic acid and insulin decreased during fasting. The MUA volley intervals and plasma concentrations of those metabolites and insulin were restored to pre-fasting levels after subsequent refeeding. In ovariectomized (OVX) goats, changes in plasma metabolites and insulin concentrations were similar to those in OVX+E2 goats, but the MUA volley intervals were not altered. The present results demonstrated that fasting suppressed GnRH pulse generator activity in an estrogen-dependent manner. Changes in plasma concentrations of energy substrates and insulin during fasting were associated with the GnRH pulse generator activity in the presence of estrogen, but not in the absence of the steroid in female goats. 相似文献

Posttranslational modification of alpha-dystroglycan (α-DG) by the like-acetylglucosaminyltransferase (LARGE) is required for it to function as an extracellular matrix (ECM) receptor. Mutations in the LARGE gene have been identified in congenital muscular dystrophy patients with brain abnormalities. However, the precise function of LARGE remains unclear. Here we found that LARGE could act as a bifunctional glycosyltransferase, with both xylosyltransferase and glucuronyltransferase activities, which produced repeating units of [-3-xylose-α1,3-glucuronic acid-β1-].
