The microbial community structure and function under forest in tropical peatlands are poorly understood. In this study, we investigated the microbial community structure and diversity in natural peat swamp forest soil, disturbed peat soil and mineral soil in Central Kalimantan, Indonesia, using 454 pyrosequencing. The results showed that the natural peat soil had the greatest fungal species richness (Chao1), which was significantly (< .05) larger than that in the other two soils. Community structure of both fungi and bacteria in natural peat soil differed significantly from that in the disturbed peat soil (= .039 and = .045, respectively). Ascomycota (40.5%) was the most abundant phylum across the three soils followed by Basidiomycota (18.8%), Zygomycota (<0.1%) and Glomeromycota (<0.1%). The linear discriminant analysis with effect size (LEfSe) showed that Ascomycota (< .05) and genus Gliocephalotrichum (< .05) dominated in natural peat soil. Functionally, pathotrophs were more abundant in disturbed peat soil (< .05). Proteobacteria (43.8%) were the most abundant phylum followed by Acidobacteria (32.6%), Actinobacteria (9.8%), Planctomycetes (1.7%). Methylocystis, Telmatospirillum, Syntrophobacter, Sorangium and Opitutus were the more abundant genera in disturbed peat soil, whereas Nevskia and Schlesneria were more abundant in mineral soil and natural peat soil, respectively. The natural peat forest soil supported a more diverse microbiology; however, the land use of such a soil can change its microbial community structure. The results provide evidence that the disturbance of tropical peat land could lead to the introduction and spread of a large number of fungal diseases  相似文献

Lamb carcasses (n = 278) were selected immediately after slaughter and fat thickness was measured with the SP2 Hennessy grading probe (HP) at the interface of the 12th and 13th ribs, 3.8 cm from the backbone. After a 24-h chilling period, carcasses were graded by a USDA grader and probed with the HP to obtain a fat thickness measure on the chilled carcass. One hundred sixty-five carcasses were fabricated into wholesale cuts (.64 cm of external fat trim), and 113 carcasses were fabricated into tray-ready retail cuts (.25 cm of external fat trim). Carcass weight, fat thickness (metal probe), adjusted fat thickness, hot and chilled carcass HP fat measures, as well as kidney and pelvic fat percentage and USDA yield grade, were highly correlated to cutting yield for both fabrication methods. Regression models developed to predict wholesale cut yields using HP or grader-collected measures were similar with respect to predictive accuracy. Fat thickness explained most of the variation in wholesale and tray-ready cut yields among the variables collected by the grader. Kidney and pelvic fat accounted for more of the variation in yield of wholesale cuts during stepwise regression to determine HP equations, but for predicting tray-ready yields, fat thickness taken with the HP accounted for the largest amount of variation. Equations developed to predict tray-ready retail cut yields using the HP or USDA grader-collected carcass measures were similar in the amount of variation explained. Kidney and pelvic fat percentage must be included in equations to maximize predictive accuracy when this depot site is left in carcasses.  相似文献

Progeny from reciprocal F1 crosses and F1 backcrosses between fenvalerate-resistant and fenvalerate-susceptible Colorado beetles, Leptinotarsa decemlineata (Say), and between carbofuran-resistant and carbofuran-susceptible Colorado beetles were bioassayed to investigate the mode of inheritance of resistance to these chemicals. Bioassays of progeny from these crosses indicate that resistance to fenvalerate is inherited in a semi-recessive, sex-linked manner and carbofuran resistance is inherited in a partially dominant autosomal fashion. Log concentration/probit mortality lines and chi-square tests, however, indicate that multiple genes may be involved in resistance to both insecticides.  相似文献

An improved ability to quantify differences in the fabrication yields of beef carcasses would facilitate the application of value-based marketing. This study was conducted to evaluate the ability of the Dual-Component Australian VIASCAN to 1) predict fabricated beef subprimal yields as a percentage of carcass weight at each of three fat-trim levels and 2) augment USDA yield grading, thereby improving accuracy of grade placement. Steer and heifer carcasses (n = 240) were evaluated using VIASCAN, as well as by USDA expert and online graders, before fabrication of carcasses to each of three fat-trim levels. Expert yield grade (YG), online YG, VIASCAN estimates, and VIASCAN estimated ribeye area used to augment actual and expert grader estimates of the remaining YG factors (adjusted fat thickness, percentage of kidney-pelvic-heart fat, and hot carcass weight), respectively, 1) accounted for 51, 37, 46, and 55% of the variation in fabricated yields of commodity-trimmed subprimals, 2) accounted for 74, 54, 66, and 75% of the variation in fabricated yields of closely trimmed subprimals, and 3) accounted for 74, 54, 71, and 75% of the variation in fabricated yields of very closely trimmed subprimals. The VIASCAN system predicted fabrication yields more accurately than current online yield grading and, when certain VIASCAN-measured traits were combined with some USDA yield grade factors in an augmentation system, the accuracy of cutability prediction was improved, at packing plant line speeds, to a level matching that of expert graders applying grades at a comfortable rate.  相似文献

An investigation on the presence of larval cestodes in musk rats (Ondatra zibethicus) was carried out in two regions of the Netherlands (east Groningen and south Limburg) where in a earlier study foxes with Echinococcus multilocularis were found. A total of 1726 musk rats were dissected (1200 in Groningen, 526 in Limburg). Larval stages of Taenia taeniaeformis were most frequently found (total 44.8%: Groningen 42%, Limburg 51.3%), followed by T. martis (total 6.1%: Groningen 0.7%, Limburg 18.6%). Infections with T. crassiceps (total 0.3%: Groningen 0%, Limburg 1.0%), T. polyacantha (total 0.2%; Groningen 0.3%, Limburg 0%) and E. multilocularis (0.1%: Groningen 0.1%, Limburg 0%) were rare. Infections with T. taeniaeformis were more frequent in adults (71.8%) than in juveniles (34.2%). The same was found for T. martis: adults 15.3%, juveniles 2.5%. This difference was also reflected in the relation between weight of the animals and presence of infection. Heavier animals (>1000 g) were more often infected with T. taeniaeformis (74.1%) than animals less than 1000 g (34.8%). In musk rats weighing less than 500 g (n=155) only 5.2% were infected, but above 1200 g, 82.6%. The highest number of T. taeniaeformis was 28, of T. martis 13, of T. crassiceps >1000 and of T. polyacantha 24. The E. multilocularis was in a very young stage, a few white spots in the liver. Although E. multilocularis infections were exceptional, it is expected that with a rise in the number of infected foxes in the Netherlands the number of infected musk rats will increase.  相似文献

The general objective of this study was to evaluate a crude Ostertagia ostertagi antigen enzyme-linked immunosorbent assay (ELISA) for monitoring gastrointestinal parasites in lactating dairy cattle. A longitudinal study of gastrointestinal parasites in lactating dairy cows was carried out in 38 herds in four provinces of Canada (Prince Edward Island, Quebec, Ontario and Saskatchewan) from September 1999 to October 2000. Bulk tank milk, cow milk, serum and fecal samples were collected monthly or quarterly from all these farms. Information on herd management factors was collected by a standard questionnaire and individual cow production data were obtained from an electronic database. The overall mean optical density ratio (ODR) was 0.30 and ranged from -0.05 to 1.55. Although a clear seasonal pattern was not observed, the ODR values tended to decrease during the housing period and start increasing in the spring before the cows went out to pasture. The second and third or greater lactation cows had significantly higher ODR values compared with first lactation animals. The individual cow ODR had a very low correlation with individual squared root fecal egg counts but showed a reasonably high correlation when herd averages values were computed (r=0.73). A moderate correlation (r approximately 0.50) between the bulk tank and herd average ODR was observed. Milk yield was negatively associated with individual cow milk ODR and a quadratic effect on ODR was observed for days in milk. Twenty-eight of the herds participated in a clinical trial of eprinomectin (Ivomec Eprinex) treatment at calving. The cow level ODR values determined late in the previous lactation had a marginally significant effect (P=0.07) on treatment response, suggesting that high OD cows responded better to the anthelmintic treatment. However, because of the small sample size available in this model, more research is needed to better understand this relationship. In conclusion, the indirect ELISA using an O. ostertagi crude antigen appears useful as a technique for monitoring gastrointestinal parasite burdens in adult dairy cows and holds promise as a potential predictor of response to anthelmintic treatment.  相似文献

This study evaluated effects of four uniquely applied beef carcass electrical stimulation (ES) treatments on USDA grade factors, muscle color, subprimal purge loss, cooked steak weight loss, and cooked steak tenderness. One side of each (n = 284) beef carcass was subjected to ES using one of four treatments (medium voltage for medium duration, MVMD; medium voltage for long duration, MVLD; high voltage for medium duration, HVMD; or high voltage for long duration, HVLD) and was compared to its corresponding non-ES control side. Electrical stimulation of beef sides was applied focusing on middle meats while preventing severe contraction of the round and chuck. From matched (ES and control) sides of 120 carcasses (10 each of Select, low Choice, and upper two-thirds of Choice in each of the four ES treatments), longissimus steaks (2.5 cm thick) were cooked and used for Warner-Bratzler shear force (WBS) analysis. Mean marbling scores (n = 284) for stimulated sides did not differ (P = .923) from those for control sides within ES treatment classes. Mean values for CIE L*, a*, and b* of lean color (n = 284) were higher (P < .05) for MVMD, MVLD, HVMD, and HVLD treated sides than for the respective control sides. When WBS values for steaks were adjusted to an equal visual degree of doneness, WBS values (n = 120) were lower (P < .05) for ES treated sides than for control sides for all four types of ES application treatments. Treatment responses were not influenced by USDA Quality Grade group. For those carcasses for which the control sides had WBS values greater than 4.5 kg, matching sides treated with MVMD, MVLD, HVMD, or HVLD had WBS values less than 4.5 kg 50, 88, 60, and 75% of the time, respectively. Mean cooked steak weight loss (n = 120), adjusted to an equal visual degree of doneness, and mean purge loss (n = 24) did not differ with ES treatment.  相似文献

We studied habitat selection and breeding success in marked populations of a protected seabird (family Alcidae), the marbled murrelet (Brachyramphus marmoratus), in a relatively intact and a heavily logged old-growth forest landscape in south-western Canada. Murrelets used old-growth fragments either proportionately to their size frequency distribution (intact) or they tended to nest in disproportionately smaller fragments (logged). Multiple regression modelling showed that murrelet distribution could be explained by proximity of nests to landscape features producing biotic and abiotic edge effects. Streams, steeper slopes and lower elevations were selected in both landscapes, probably due to good nesting habitat conditions and easier access to nest sites. In the logged landscape, the murrelets nested closer to recent clearcuts than would be expected. Proximity to the ocean was favoured in the intact area. The models of habitat selection had satisfactory discriminatory ability in both landscapes. Breeding success (probability of nest survival to the middle of the chick rearing period), inferred from nest attendance patterns by radio-tagged parents, was modelled in the logged landscape. Survivorship was greater in areas with recent clearcuts and lower in areas with much regrowth, i.e. it was positively correlated with recent habitat fragmentation. We conclude that marbled murrelets can successfully breed in old-growth forests fragmented by logging.  相似文献
