This experiment was conducted to investigate the effect of coconut oil and garlic powder supplementation on digestibility of nutrients, rumen fermentation, rumen ecology, rumen microorganism and methanogen diversity. Four, 3-year old, rumen fistulated swamp buffalo bulls were randomly assigned in a 4 × 4 Latin square design to receive four dietary treatments; 7% coconut oil, 7% coconut oil with 50 g/day of garlic powder, 7% coconut oil with 100 g/day of garlic powder and non-supplemented (control). During the experiment, concentrate was offered at 0.5% of BW while rice straw was given on ad libitum basis. It was found that supplementation of 7% coconut oil had significantly influenced on total DM intake, OM, NDF and ADF digestibilites while supplementation of 7% coconut oil with garlic powder (50 and 100 g/day) were not significantly different when compared with the control. Dietary supplementations did not affect on rumen pH, NH3–N concentrations. Blood urea nitrogen (BUN) was significantly higher in supplemented groups. Total VFA concentration, proportion of acetate and acetate to propionate ratio were reduced by supplementation. Proportion of propionate was increased (P < 0.05) when supplemented with 7% coconut oil and 7% coconut oil with 100 g/day of garlic powder. Methane production was dramatically reduced (P = 0.005) in supplemented treatments and was 10% reduced in 7% coconut oil supplementation. Amylolytic and proteolytic bacteria were increased (P = 0.007 and P = 0.024) while protozoal population decreased by 68–75% (P < 0.01) by supplementation. Total bacterial population was increased by supplementation while total fungi and total methanogens were not significantly different among treatments. Percentage of cellulolytic bacterial population was not different among treatments. However, dietary supplementation reduced (P < 0.001) percentage of F. succinogenes population. However, methanogen diversity was not changed using PCR–DGGE as technique. Thirteen separate bands were found in the DGGE gel. Based on this study, supplementation with 7% coconut oil plus 100 g/day of garlic powder could be efficiently utilized in the rumen and thus, could provide good fermentation end products and improve rumen ecology for the host swamp buffaloes particularly in reducing 9% methane gas production without changing nutrient digestibilities.  相似文献

Elephant endotheliotropic herpesvirus (EEHV) infection is one of the most common diseases in young elephants, causing severe fatal hemorrhagic disease. Subclinical infection was previously described; however, information about the factors associated with virus shedding and reactivation were scarce. To identify the biological and environmental factors related with EEHV detection, blood and oral swab samples were collected from nine captive Asian elephants in Thailand for one year and tested for EEHV presence using real-time PCR. Data including hematological values, management, environmental temperature, and serum cortisol levels were also recorded and analyzed. Results showed that the viral detection frequency ranged from 0–25%. The highest detection frequency was found in the two youngest elephants, aged less than 15 years. Three types of viruses, EEHV1, EEHV4, and EEHV5, were found in this study, which also detected mixed infection in five elephants. Additionally, the study found that sample type, changes in hematological values, management and health issues, and serum cortisol levels were not associated with herpesvirus detection in the elephants. However, EEHV detection percentage was significantly increased in the summer (mid-Feb to mid-May), possibly due to body fitness reduction from food source limitation and low nutrient content. To obtain a broad aspect of EEHV management, long-term EEHV monitoring is highly recommended in every captive elephant herd.  相似文献

The objective of this study was to compare the number of recovered spermatozoa, in different parts of the uterine horn and oviduct in gilts, after insemination with fractionated (experiment) and non-fractionated (control) liquid stored semen. The number of spermatozoa and volume of backflow was also investigated. Twenty three cross-bred gilts were used in the study. They were divided into 2 groups, a control group (non-fractionated liquid stored semen, n=10) which were inseminated with 100 ml of liquid stored semen containing 3,000 million spermatozoa per dose and an experimental group (fractionated liquid stored semen, n=10) which were inseminated with 50 ml of liquid stored semen, with 3,000 million spermatozoa per dose and followed by another 50 ml of semen dilutor (Beltsville Thawing Solution, BTS). Thereafter, backflow semen was collected and measured every 15 min for a period of 1 hr. Three or 12 hr after insemination, 5 gilts from each group had the uterus, the horn of the uterus, the oviducts and the ovaries removed under general anaesthesia. The horn of uterus and the oviducts were seperated by ligation into 6 segments. All 6 segments were flushed with BTS to collect all spermatozoa within the segment. Recovered spermatozoa were counted, using a haemocytometer and the volume recorded. It was seen that the percentage of spermatozoa in the backflow semen in the experimental group was less than in the control group. The difference was not significant in the gilts that were operated on 3 hr after insemination, the mean number of spermatozoa in the uterine horn and the utero-tubular junction (UTJ) was more in the experimental than in the control group, but less in the isthmus and the ampulla of the oviduct. The gilts which were operated on 12 hr after insemination, had relativity more ovulating gilts in the control group than in the experimental group (3 of 4 gilts compare to 3 of 5 gilts). The control group had more spermatozoa in the oviduct than the experimental group, but less in UTJ and in the horn of the uterus. Again the difference was not significant. It can be concluded that fractionated (experimental) or non-fractionated (control) insemination of semen with the same number of spermatozoa provides no significant difference in the number of spermatozoa either in the horn of the uterus, the UTJ or the oviduct of gilts.  相似文献

To determine the relationships among vascularity, expression of angiogenic factors, and selected intrafollicular factors in dominant and nondominant follicles of the first follicular wave, ovaries were obtained on d 3 of the estrous cycle from mature cross-bred beef heifers (n = 8) after a synchronized estrus. Follicular fluid (FF) was collected from all follicles > or = 3 mm for determination of estradiol-17beta (E), progesterone (P4), vascular endothelial growth factor (VEGF), and IGFBP concentrations. The ovaries were then perfusion-fixed and used for histochemical detection of lectin BS-1 (a marker of endothelial cells and thus vascularization) binding, and immunolocalization of VEGF, endothelial nitric oxide synthase (eNOS), and proliferating cell nuclear antigen, followed by image analysis of selected follicles. Follicles were classified, based on E and P4 concentrations in FF, as dominant, estrogen-active (EA; E:P4 > or = 1) or nondominant, estrogen-inactive (EI; E:P4 <1). Concentrations of E and VEGF in FF, the area of positive staining for lectin BS-1, VEGF, and eNOS, and the labeling index (an index of the percentage of cells proliferating) in granulosa and theca layers were greater (P < 0.05) in the EA than in the EI follicles, but concentrations of P4 and IGFBP in FF were less (P < 0.05) in EA than in EI follicles. In addition, vascularity was positively correlated (P < 0.05) with VEGF and eNOS protein expression, and tended (P < 0.1) to be positively correlated with the E:P4 ratio in FF but tended (P < 0.1) to be negatively correlated with IGFBP and P4 concentrations in FF. These data highlight the importance of vascularity, angiogenic factors, and IGFBP in the health of the dominant follicle in heifers, and indicate that the FF concentrations of E, VEGF, IGFBP, and P4, and the E:P4 ratio can be used as markers of dominant follicles.  相似文献

In the traditional shifting cultivation system practiced by the Karen people in northern Thailand, the effects of burning on the content of extractable organic matter, microbial biomass, and N mineralization process of the soils were studied. Five plots (5×5 m2 quadrat) with 0, 10, 20, 50, and 100 Mg ha-1 of slashed materials were arranged and burned. Ten to 20 Mg ha-1 of slashed biomass corresponded to the amount commonly burned by the Karen people. During the burning process, the soil temperature at the depth of 2.5 cm in the 100 Mg ha-1 plot almost evenly increased to 300°C while the temperature in the 10 to 50 Mg ha-1 plots increased with large variations from 50 to 300°C. Burning caused a conspicuous increase in the contents of organic C and (organic + mineral)-N extracted at room temperature and a simultaneous decrease in the contents of microbial biomass C and N, especially in the soil of the 100 Mg ha-1 plot. In the rainy season, the values of the changes induced by burning reverted to the values recorded before burning, except for the microbial biomass in the 100 Mg ha-1 plot, which still remained lower. Based on an incubation experiment, N mineralization rate was higher in the soils taken just after burning, especially in the 100 Mg ha-1 plot, than in the soils taken during the rainy season. However, the soil in the 100 Mg ha-1 plot was considered to have the lowest ability to supply mineral N among the soils in the rainy season. Burning of 10 to 20 Mg ha-1 biomass corresponding to the values recorded in Karen peoples' shifting cultivation system was more compatible with soil ecology in terms of N supply at the initial stage of crop growth and of microbial biomass recovery during the rainy season, compared to the burning of 100 Mg ha-1 biomass corresponding to the value recorded in a natural forest. Thus, the shifting cultivation system implemented by the Karen people can be considered to be a well-balanced agricultural system.  相似文献

An in vitro gas technique trial was conducted to investigate the effect of coconut oil (Co), garlic powder (G) and their mixtures on in vitro fermentation. Incubation was carried out using rumen fluid obtained from swamp buffaloes. The experimental design was a completely randomized design (CRD). The dietary treatments were ratio of Co and G supplementation at 0:0, 16:0, 8:4, 4:8 and 0:16 mg with rice straw as a roughage source. Cumulative gas production was recorded at 0, 2, 4, 6, 8, 12, 18, 24, 36, 48, 60 and 72 h of incubation. In vitro true digestibility (IVTD) was determined after 48 h incubation. Cumulative gas production at 72 h was significantly lowest (P < 0.05) at Co:G, 16:0 mg. Garlic powder supplementation at 16 mg decreased (P < 0.05) NH3–N concentration and increased (P < 0.05) in vitro true digestibility (IVTD) while supplemented coconut oil at 16 mg decreased (P < 0.05) IVTD. Total volatile fatty acids (VFAs) were lowest (P < 0.05) by garlic powder supplementation at 16 mg. However, supplementation of Co:G, 8:4, 4:8 and 0:16 mg tended to increase the proportion of propionate, decrease C2:C3 ratio and reduce (P < 0.05) methane (CH4) production. Protozoal population was significantly lowest (P < 0.05) at Co:G, 8:4 mg. Moreover, application of quantitative PCR to quantify predominant cellulolytic bacteria (16S rRNA) and fungi (18S rRNA) targets revealed that treatments did not have an effect on Ruminococcusflavefaciens and total fungi population. However, it was found that supplementation of Co:G at 8:4 mg increased Ruminococcusalbus population (P < 0.05). Based on this study, it suggests that supplementation of Co:G at 8:4 and 0:16 mg could improve ruminal fluid fermentation in terms of volatile fatty acid profile, reduced methane losses and reduced protozoal population.  相似文献

Green sea turtles are widely distributed in tropical and subtropical waters. Adult green sea turtles face many threats, primarily from humans, including injuries from boat propellers, being caught in fishing nets, pollution, poaching, and infectious diseases. To the best of our knowledge, limited pharmacokinetic information to establish suitable therapeutic plans is available for green sea turtles. Therefore, the present study aimed to describe the pharmacokinetic characteristics of ceftriaxone (CEF) in green sea turtles, Chelonia mydas, following single intravenous and intramuscular administrations at two dosages of 10 and 25 mg/kg body weight (b.w.). Blood samples were collected at assigned times up to 96 hr. The plasma concentrations of CEF were measured by liquid chromatography tandem mass spectrometry. The concentrations of CEF in the plasma were quantified up to 24 and 48 hr after i.v. and i.m. administrations at dosages of 10 and 25 mg/kg b.w., respectively. The Cmax values of CEF were 15.43 ± 3.71 μg/ml and 43.48 ± 4.29 μg/ml at dosages of 10 and 25 mg/kg, respectively. The AUClast values increased in a dose‐dependent fashion. The half‐life values were 2.89 ± 0.41 hr and 5.96 ± 0.26 hr at dosages of 10 and 25 mg/kg b.w, respectively. The absolute i.m. bioavailability was 67% and 108%, and the binding percentage of CEF to plasma protein was ranged from 20% to 29% with an average of 24.6%. Based on the pharmacokinetic data, susceptibility break‐point and PK‐PD index (T > MIC, 0.2 μg/ml), i.m. administration of CEF at a dosage of 10 mg/kg b.w. might be appropriate for initiating treatment of susceptible bacterial infections in green sea turtles.  相似文献

To determine the relationships among vasculature, mitotic activity, and expression of endothelial nitric oxide synthase (eNOS) of antral follicles in Bos indicus, bovine ovaries were obtained on day 6 of the estrous cycle from 10 crossbred (Brahman to Thai native cows) after a synchronized estrus with prostaglandin F2α analogue. Ovaries were fixed, paraffin-embedded, and used for immunofluorescence detection of factor VIII (a marker of endothelial cells). Immunostaining of eNOS and proliferating cell nuclear antigen (PCNA) were performed with specific monoclonal antibodies. Vasculature and positive staining of eNOS and PCNA were quantitatively evaluated with the image analysis. Follicles were classified by size (small, medium, and large) and by structure as healthy and atretic follicles (n = 82). The expression of factor VIII and eNOS were detected greater in the blood vessels of the theca layers of the healthy follicles than those in atretic follicles. The labeling indices (LIs) in granulosa and theca cells were greater (P < 0.05) in the healthy small and medium follicles than in the healthy large follicles. Vasculature, capillary area density, and capillary number density were positively correlated with eNOS expression and the LIs of granulosa and theca cells but were negatively correlated with the healthy follicle size. During the growing phase of antral follicle in Bos indicus, relationships among vasculature, mitotic activity, and eNOS were observed predominantly in healthy antral follicles. Thus, these data highlight the importance of vasculature, cell proliferation, and eNOS expression of growing and atretic follicles in the first follicular wave.  相似文献

To determine the effects of Follicle Stimulating Hormone (FSH) treatment and subsequent withdrawal on uterine proliferation and estrogen receptor (ESR), Brahman crossbred heifers (n = 12) were twice daily injected with FSH (4, 3, and 2 mg/injection) on Days 17–19 of the estrous cycle (FSH 3 days) and (4 and 3 mg/injection) on Days 17–18 (FSH 2 days) and withdrawal with saline on Day 19 and (4 mg/injection) on Day 17 (FSH 1 day) and withdrawal with saline on Days 18–19. Uterine tissue was subjectively collected on Day 20 and microscopically classified to four regions: endometrial stroma (ES), surface endometrial gland (EG), deep endometrial gland (DG), and myometrium (Myo). The cell proliferation marker, Ki-67, was quantified as labeling index (LI) in uterine regions, and tissues were immunostained to detect ESR2 followed by image analysis. The LI of ES, EG, and DG was greater (P = 0.0018, P = 0.0005, and P = 0.0103; respectively) in heifers received FSH for 3 days. The expression of ESR2 protein on ES and EG was greatest (P < 0.0001 and P = 0.0036, respectively) in FSH 3 days-treated group. Thus, FSH administration during proestrus stimulates uterine cell proliferation, and ESR2 expressions are affected by FSH during proestrus and differentially distributed in the uterine regions.  相似文献
