Serum samples of five goats inoculated with Toxoplasma gondii were analyzed using the enzyme linked immunosorbent assay (ELISA), indirect hemagglutination (IHA) and western blotting (WB). Antibodies detected by ELISA peaked between 19 and 62 days after inoculation and persisted throughout the experiment with no association to parasitaemia. Using WB, the main antigens detected had molecular weights of approximately 68, 62, 50, 48, 42, 34, 28, 26, 22 and 19 kDa. Antibody titers of between 1:256 and 1:32000 were observed using IHA, with a significant drop in activity after treatment with 2-mercapto-ethanol between days 12 and 48. This coincided with the parasitaemic period that occurs between 5 and 64 days after inoculation. 相似文献

The coffee berry borer, Hypothenemus hampei (Ferrari), is an important devastating coffee pest worldwide. Both trypsin and chymotrypsin enzyme activities from H. hampei larval midgut can be inactivated by proteinaceous enzyme-inhibitors. A serine proteinase inhibitor belonging to the Bowman-Birk class was purified from a wild accession of Phaseolus coccineus L. seeds. The inhibitor (PcBBI1) is a cysteine-rich protein that is heat-stable at alkaline pH. MALDI-TOF/MS analysis showed that PcBBI1 occurs in seeds as a monomer (8689 Da) or dimer (17,378 Da). Using in vitro inhibition assays, it was found that PcBBI1 has a high inhibitory activity against H. hampei trypsin-like enzymes, bovine pancreatic chymotrypsin, and trypsin. According to this, PcBBI1 could be a promising tool to make genetically modified coffee with resistance to coffee berry borer. 相似文献

This study was conducted to determine the age and liveweight at puberty of 120 crossbred beef heifers submitted to four diets to achieve predetermined weight gains (kg/day): 0.5 (G500; n?=?32), 0.75 (G750; n?=?32), 1.0 (G1000; n?=?29), and 1.25 (G1250; n?=?27). Animals were classified depending on their level of crossing between Nelore (N) and Hereford (H): 25%N-75%H, 37.5%N-63.5%H, 43.7%N-56.7%H, 50%N-50%H, and 75%N-25%H. Reproductive evaluation was performed at the beginning and at the end of the experimental period by ultrasonography and hormone analysis. The average age and liveweight at puberty were 388.0?±?1.9 days and 331.4?±?1.3 kg, respectively. Animals from the 25%N-75%H group reached puberty earlier than heifers from other genetic groups supplemented with G1250 diet (P?<?0.05). Heifers with higher degree of Nelore (75%N-25%H) fed with G1000 diet showed estrus 42 days prior to mating, but only 57 % reached puberty at mating (P?<?0.05). Heifers with follicles of higher diameter reached puberty (P?<?0.05) earlier. Higher average daily weight gain showed a positive effect on follicular diameter and IGF-I level at puberty (P?<?0.01). Concentrations of GH were lower in heifers fed G1250 compared to G1000 diet (P?<?0.05). There was a significant interaction between nutritional level and insulin levels at puberty (P?<?0.01). We demonstrated the relationship between IGF-I and average daily gain on the onset of puberty in heifers. Most materials shrink laterally like a rubber band when stretched, so their Poisson's ratios are positive. Likewise, most materials contract in all directions when hydrostatically compressed and decrease density when stretched, so they have positive linear compressibilities. We found that the in-plane Poisson's ratio of carbon nanotube sheets (buckypaper) can be tuned from positive to negative by mixing single-walled and multiwalled nanotubes. Density-normalized sheet toughness, strength, and modulus were substantially increased by this mixing. A simple model predicts the sign and magnitude of Poisson's ratio for buckypaper from the relative ease of nanofiber bending and stretch, and explains why the Poisson's ratios of ordinary writing paper are positive and much larger. Theory also explains why the negative in-plane Poisson's ratio is associated with a large positive Poisson's ratio for the sheet thickness, and predicts that hydrostatic compression can produce biaxial sheet expansion. This tunability of Poisson's ratio can be exploited in the design of sheet-derived composites, artificial muscles, gaskets, and chemical and mechanical sensors. 相似文献

Purpose
Polycyclic aromatic hydrocarbons (PAHs) are unintentional products that are classified as persistent toxic substances. The goal of the present study was to generate data on the presence of 15 priority PAHs that are found in surface sediment and core sediment in the region of the Turvo/Grande watershed, São Paulo State, Brazil, which is an area of expanding sugarcane cultivation, and to correlate these data with the sources of these PAHs and the guiding values for sediment quality analysis.Materials and methods
Surface sediments and sediment cores were sampled during the rainy and dry seasons in February and July 2010. The extraction of PAHs from sediments was performed using a Soxhlet extractor, and then the extract was cleaned according to the methods of the US EPA 3630C (US EPA 1996) using a silica gel column. Quantification was performed using high performance liquid chromatography with fluorescence detection.Results and discussion
The concentrations of all 15 PAHs decreased as the depth of the sediment core increased. Overall, the concentrations decreased along the sediment core; however, the RTURARG (region predominantly used for planting sugarcane and livestock) during the rainy season and the CAPRP sampling site (located at part of the Preto River dam) during the dry season showed increased concentrations in the first few sediment fractions, and then the concentrations decreased. Higher concentrations were observed in urban locations, and the concentration of naphthalene was higher than the probable effect level (PEL) determined by the Canadian environmental agency. The obtained diagnostic ratios indicate that the sediment from areas with an abundance of sugarcane was a pyrolytic source of PAHs, which indicates a contribution from burning straw to the PAH concentrations in those areas.Conclusions
For all sampling sites and all PAHs, we found a decreasing trend in PAH concentrations with increasing sediment core depth, and the locations, such as CAPRP, that experienced a higher level of human activity had the highest total concentrations of PAHs. These locations were the only areas in which the PAH naphthalene was found in higher concentrations than the PEL. The diagnostic ratios reveal that regions with sugarcane plantations had predominantly pyrolytic sources of PAHs, indicating the contribution of PAHs from sugarcane straw burning.7.
The organophosphorus insecticides, parathion and azinphos (10?5-10?4M), significantly stimulate the Ca2+-pump activity of sarcoplasmic reticulum, while malathion has a limited effect. The rates of Ca2+ translocation and ATP hydrolysis are both stimulated and, apparently, the ratio is improved. Parathion and azinphos maximally increase this ratio by 26 and 14%, respectively. The organochlorine compounds, DDT and aldrin, also stimulate the Ca2+ pump, and lindane has a reduced effect. These effects are smaller than those observed for parathion and azinphos. The order of effectiveness is similar to the toxicity of the compounds to mammals and can be described as follows: parathion > azinphos > DDT ≈ aldrin > malathion ≈ lindane. 相似文献

1. Microbial β-1,3-1,4-glucanases improve the nutritive value of barley-based diets for poultry by effectively decreasing the degree of polymerisation of the anti-nutritive soluble β-glucans. Glycoside hydrolases (GHs) acting on recalcitrant polysaccharides display a modular architecture comprising a catalytic domain linked to one or more non-catalytic Carbohydrate-Binding Modules (CBMs). 2. GHs and CBMs have been classified in different families based on primary structure similarity (see CAZy webpage at http://www.cazy.org). The role of CBMs is to anchor the appended GHs into their target substrates, therefore eliciting the efficient hydrolysis of structural carbohydrates. 3. Here we describe the biochemical properties of the family 16 GH from Clostridium thermocellum, termed CtGlc16A. CtGlc16A is a thermostable enzyme that specifically acts on β-1,3-1,4-glucans with a remarkable catalytic activity (38000?U/mg protein). 4. CtGlc16A, individually or fused to the family 11 β-glucan-binding domain of cellulase CtLic26A-Cel5E of C. thermocellum, was used to supplement a highly viscous barley-based diet for broilers. 5. The data showed that birds fed on diets supplemented with the recombinant enzymes displayed an improved performance when compared with birds given diets not supplemented with exogenous enzymes. However, inclusion of the non-catalytic CBMs had no influence on the capacity of CtGlc16A to reduce the anti-nutritive effects of soluble β-1,3-1,4-glucans. 6. Organic additives are recently being used in animal diets owing to their ability to control metabolic issues and result in better animal performance. Specifically, the organic additive Fator P® presents an additional advantage that is to cause a lesser greenhouse gas emission. This study evaluated whether Fator P® intake changes ruminal parameters or animal performance of beef cattle. Evaluations were carried out in a feedlot experiment divided into growing (46 days; two diets [control mix—CM and standard mix—SM] and finishing (lasted 83 days; four diets: CM, SM, Fator P® + virginiamycin, and Fator P® alone [FP]) trials. Animal performance study involved 48 animals allocated to 12 collective pens in completely randomized experimental design. Ruminal parameters were evaluated in separate metabolism study developed carried out using individual pen with four steers. During growing trial, FP diet resulted in higher (p < 0.05) dry matter intake (DMI) and ruminating time. In the finishing trial, diets containing Fator P® resulted in higher DMI than obtained with CM. Most of the ruminal parameters did not differ (p > 0.05) among dietary treatments. Therefore, Fator P® represents a viable and safe strategy for supplementation to beef cattle finished using high‐concentrate diet in feedlot systems. 相似文献
