The hypothesis that an altered expression of CD11/CD18 on bovine circulating monocytes, polymorphonuclear leukocytes (PMN), or both, contributes to an increased mastitis susceptibility in periparturient cows was tested. Expression of CD18 and CD11a, -b, -c on bovine monocytes and PMN were assessed in 8 Friesian-Holstein cows by flow cytometry from 2 wk before calving to 5 wk after calving. Minor changes in adhesion molecule expression levels were detected throughout the experimental period. Compared with PMN, monocytes exhibited an expression level that was similar for CD18, higher for CD11a and CD11c, but lower for CD11b. Differences in density may reflect the relative importance of these adhesion molecules on both leukocyte types. In this study, the decreased number of milk resident macrophages and PMN observed during the periparturient period could not be attributed to changes of CD11/CD18 levels on circulating leukocytes.  相似文献

Although there has been increasing research on the adoption of agroforestry technologies over the last decade, few such studies have assessed uptake over a long period and many are based on a single snapshot in time. Furthermore, most of these studies have mainly looked at non-adopters and adopters: only recently have social scientists considered testers. A further category of users neglected in adoption studies has been re-adopters of technologies. Studying this group provides an interesting and more nuanced understanding of adoption and re-adoption. Methodologically, most adoption studies use quantitative methods and fail to link their findings to wider socio-economic, political and institutional settings. This paper presents a study of the dynamics of improved tree fallow use by farmers in Siaya and Vihiga districts of western Kenya over a period of eight years. It uses both qualitative and quantitative data to critically discuss the motivations of adopters, testers/rejecters and re-adopters. The results show that the process of adoption is highly dynamic and variable with farmers planting improved fallows and discontinuing or re-adopting them due to a whole range of factors, of which soil fertility improvement is just one. These factors included incentives from projects, the tying of adoption to credit programmes, prestige, participation in seminars/tours and the availability of a seed market from projects promoting improved fallows. Farmers planting improved fallows for such reasons may be termed ‘pseudo-adopters’. There were significant differences in adoption between the two districts, with more farmers in Siaya planting improved fallows than in Vihiga. A majority of farmers in Vihiga (53%) who were given seed never planted improved fallows, even though they had been exposed to the technology. Some 40% of farmers in Siaya and 38% in Vihiga planted improved fallows but later rejected them. This has some important implications for research and development. For improved fallow technologies to be attractive to farmers, they must provide other tangible economic benefits besides soil fertility improvement. This presents a challenge to researchers who must better attune themselves to the needs and demands of farmers if they wish to see their research findings widely adopted.  相似文献

In gold mining regions, the risk of soil pollution by mercury is a major environmental hazard, especially in tropical areas where soil microflora plays a major part in soil functioning, major bio-geochemical cycles and carbon turn-over. The impact of mercury pollution on soil microflora should thus be carefully assessed in such environments while taking into consideration the specificities of tropical soils. The aim of this study was to compare the effects of mercury (0, 1 and 20 μg of inorganic mercury per gram of soil) on the functional diversity and genetic structure of microbial communities in a tropical soil. We investigated the effects of mercury on tropical soil microflora using soil microcosms spiked with mercury and incubated at 28 °C for 1 month. Microcosm flora, its biomass and its activity, as well as its functional and genetic structure, were followed by cultural methods, measures of respiration, ECOLOG plates, and DGGE (denaturing gel gradient electrophoresis), respectively. Fate of total and bioavailable mercury was estimated by CVAFS (cold vapor atomic fluorescence spectrometry). Results obtained for the microcosms enriched with only 1 μg g?1 mercury were indistinguishable from controls. Conversely, in the presence of high mercury contents (20 μg g(1), an immediate effect was measured on soil respiration, functional diversity (ECOLOG plates) and genetic structure (DGGE), although no significant effect was observed on plate counts or microbial biomass. In addition, whereas microbial activities (respiration and functional diversity) rapidly regained control values, a lasting effect of the high mercury concentration was observed on the genetic structure of the soil microbial community. These modifications took place during the first week of incubation when total mercury concentration was declining and bioavailable mercury was at its highest.This multiple approach study is one of the first attempts at investigating the effects of mercury on soil microbial communities in tropical soils. Our results demonstrate that in the tropical soil under study, mercury affects the soil microbial communities in a different manner than was previously reported in temperate soils. Furthermore, mercury toxicity on soil microbes may be modulated by typical tropical soil characteristics.  相似文献

The successful application of an interstitial brachytherapy course is reported in a 14-month-old male Anglo European Studbook (AES) foal for the treatment of a recurrent dermal vascular hamartoma at the dorsal aspect of the pastern on the right hindlimb. The lesion was irradiated four times over 4 days to a total dose of 28 Gy. Progressive tumour shrinkage and necrosis were observed over 2 months. No significant acute side effect was reported at the end of the treatment, with excellent and long-lasting cosmetic results observed. The only reported late effect was permanent leukotrichia within the treated area, which remained otherwise tumour-free 1224 days after treatment. Results of this report suggest the potential use of radiation therapy (and specifically brachytherapy techniques), as an effective and safe treatment modality for vascular malformations in horses.  相似文献

Logistic regression models integrating disease presence/absence data are widely used to identify risk factors for a given disease. However, when data arise from imperfect surveillance systems, the interpretation of results is confusing since explanatory variables can be related either to the occurrence of the disease or to the efficiency of the surveillance system. As an alternative, we present spatial and non-spatial zero-inflated Poisson (ZIP) regressions for modelling the number of highly pathogenic avian influenza (HPAI) H5N1 outbreaks that were reported at subdistrict level in Thailand during the second epidemic wave (July 3rd 2004 to May 5th 2005). The spatial ZIP model fitted the data more effectively than its non-spatial version. This model clarified the role of the different variables: for example, results suggested that human population density was not associated with the disease occurrence but was rather associated with the number of reported outbreaks given disease occurrence. In addition, these models allowed estimating that 902 (95% CI 881–922) subdistricts suffered at least one HPAI H5N1 outbreak in Thailand although only 779 were reported to veterinary authorities, leading to a general surveillance sensitivity of 86.4% (95% CI 84.5–88.4). Finally, the outputs of the spatial ZIP model revealed the spatial distribution of the probability that a subdistrict could have been a false negative. The methodology presented here can easily be adapted to other animal health contexts.  相似文献

Powdery mildew is one of the most common diseases of oaks in Europe. After alarming reports in the beginning of the twentieth century following the presumed introduction of the invasive fungus, the disease has become familiar to foresters. However, its impact may vary greatly according to intrinsic and extrinsic factors.

? Aims

We aimed at providing updated and synthesised information on the impact of powdery mildew on oak and on the effects of environment on disease.

? Methods

A comprehensive literature review was performed, including old reports of the early epidemics to more recent data.

? Results

Tree growth patterns are of critical importance to explain the severity of the disease and the differences between juvenile and mature trees. A critical element, especially for infection of mature trees, is the availability of spores during the production of the first leaf flush. High disease impact is often related to modified growth patterns, either by environmental factors (insects or frost) or silvicultural practices (e.g., coppicing).

? Conclusion

Powdery mildew can have important impacts in natural oak regenerations and a significant role in decline of mature trees. Climate change might influence the disease severity mainly by altering the host pathogen phenological synchrony. Process-based models are required for reliable predictions.  相似文献

Robinia pseudoacacia L. heartwood is characterized by a very high natural durability. However, a significant difference was observed between the mature and juvenile heartwood, the latter presenting less durability against fungi decay, which could be attributed to lower extractive content. In order to elucidate this idea, extractives from mature and juvenile heartwoods of black locust trees were investigated. Results showed that extractive and phenolic contents were higher in mature than in juvenile heartwoods. The identification of phenolic compounds by UPLC–DAD–MS/MS revealed, for the first time, the presence of resveratrol and piceatannol. These two stilbenes as well as the flavonoid dihydrorobinetin were present at the highest level in mature heartwood, and as they are known antifungals, they could account for the great durability of mature heartwood. The stilbenes were detected in significant amounts particularly in mature heartwood where piceatannol reached a level tenfold higher than that reported for Japanese knotweed roots, the primary natural source of these stilbenes, whereas resveratrol level was comparable with reported values. As resveratrol and piceatannol receive increasing demand for nutraceutical, cosmetic and, possibly, pharmaceutical purposes, due to their beneficial health effects, this study underlines the use of R. pseudoacacia as a promising sustainable and economical source of resveratrol and piceatannol.  相似文献

Except where nitrate is added to the soil artificially, nitrate is leached from forest soils only if it is produced. Although the factors influencing nitrification have been widely studied, nitrification activity still cannot be simply predicted from ecosystem characteristics. In France, about half of the present forest area was agricultural in 1850. Previous work suggested that former cultivation could be a major factor influencing nitrogen availability in forest soils. Using laboratory incubations, we compared the net production of ammonium and nitrate in soils from formerly manured lands planted with conifers 70–100 years ago with that in soils of surrounding ancient coniferous forests. Net nitrate production, available P content, and natural abundance of nitrogen 15, δ15N, were greater in soils from formerly manured plots than other land, whereas the C:N ratio of the soil was less. The difference in net nitrate production between previously manured sites and adjacent ancient forests was related to differences in δ15N values in the soil but not evidently to other soil properties. Because soil δ15N increases with the intensity of organic manuring, these results suggest that nitrification in forest soils depends on previous manurial practices under agriculture. In this context, the soil δ15N might be used as an indicator of both previous agricultural land use and potential nitrification. Because a significant proportion of West European forests grow on previously cultivated soils, past land use should be taken into account when evaluating the risks of nitrate leaching from forests.  相似文献
