The increased recognition of the importance of soil is reflected in the UN Post‐2015 Development Agenda with sustainable development goals that directly and indirectly relate to soil quality and protection. Despite a lack of legally binding legislation for soil protection, the European Commission remains committed to the objective of soil protection. However, the achievement of a legally binding framework for soil protection relies on the implementation of a soil monitoring network (SMN) that can detect changes to soil quality over time. As beneficiaries do not pay for the provision of soil information, the options for soil monitoring are limited. The use of existing data sets should be considered first. Using Ireland as an example, this research explored the opportunities for a SMN for Ireland considering three existing national data sets. The options for a SMN are considered in terms of their spatial and stratified distribution, the parameters to be measured and an economic analysis of the options proposed. This research finds that for Ireland, either a 10 or a 16 km2 grid interval stratified by land use and drainage class offers the best potential in relation to the spatial distribution of existing data sets to reflect local data at a national level. With existing data, the stratified SIS data using the 16 km2 grid offers the best value for money, with baseline costs for analysis, excluding field costs, of between €706 481 and €2.8 million. Acknowledging the impossibility of measuring all parameters with ideal frequency, this study proposes a two‐tier system for optimized monitoring frequency. Parameters must anticipate future policy requirements. Finally, the implementation of a SMN must be accompanied by standardized methods, defined thresholds and action mandates to maintain soil quality within allowable limits.  相似文献

AIM: To compare, in cows treated with an internal teat sealant, the effect of short-acting and long-acting cloxacillin-based dry-cow therapy on somatic cell counts (SCC) after calving.

METHODS: Cows from a spring-calving, pasture-based dairy farm in the Manawatu-Whanganui region of New Zealand were randomly allocated to receive either a short-acting cloxacillin and ampicillin dry-cow therapy and internal teat sealant (n=291) or a long-acting cloxacillin and ampicillin dry-cow therapy and internal teat sealant (n=288) at the end of lactation. Cows were managed on-farm with routine husbandry procedures through the dry period and following calving. A multivariable logistic regression model was used to determine the association between length of action of dry-cow therapy and the proportion of cows with a SCC >150,000?cells/mL at the first herd test after calving.

RESULTS: Age of cow, mean SCC for the preceding season and interval from calving to the first post-calving herd test were all associated with the proportion of cows with an individual SCC >150,000?cells/mL at the first herd test (p<0.001) Treatment with the short-acting dry-cow therapy was not associated with decreased odds of cows having a SCC >150,000?cells/mL at the first herd test compared with treatment with long-acting dry-cow therapy (OR=0.724; 95% CI=0.40–1.30).

CONCLUSIONS AND CLINICAL RELEVANCE: In this herd, which routinely used internal teat sealants, the use of short-acting cloxacillin-based dry-cow therapy did not result in an increased proportion of cows with elevated SSC post-calving. This was a single farm, single year study but indicates that in this herd, changing from a long-acting to a short-acting antimicrobial may have no impact on the prevalence of subclinical mastitis.  相似文献

The Less Favoured Area (LFA) scheme is a major element of the EU Rural Development Policy, aimed at supporting farming in areas with natural handicaps or low soil productivity. It has been in place since 1975 and accounts for 14% of total Community funding. In 2003, the European Court of Auditors recommended that the socio‐economic criteria on which the current scheme is based be replaced by biophysical criteria. Reviews of the proposals suggest that in Atlantic climates of Northwest Europe, the new criteria do not delineate adequately areas where agricultural productivity is constrained by the biophysical environment and that such areas are instead demarcated by the occurrence of excess soil moisture conditions. In this paper, we review the impact of excess soil moisture conditions on the sustainability of farming systems and their role in constraining strategic and tactical farm management practices. In particular, we review the scientific evidence on the impact of excess soil moisture conditions on herbage growth, herbage utilization, farm operations and environmental sustainability. On the basis of this, we propose an additional biophysical criterion for the new delineation of LFAs, namely the length of time that soil water is in excess of field capacity (‘field capacity days’). While there is no clear threshold for field capacity days above which agricultural sustainability is acutely constrained, the evidence reviewed in this paper suggests that the sustainability of intensive livestock farming and tillage systems is particularly challenging in scenarios where the 80 percentile of field capacity days exceeds 220–230 days.  相似文献

Whey protein concentrate solutions (12% w/v, pH 6.65 +/- 0.05) were pressure treated at 800 MPa for 20-120 min and then examined using size exclusion chromatography (SEC), small deformation rheology, transmission electron microscopy, and various types of one-dimensional (1D) and two-dimensional (2D) polyacrylamide gel electrophoresis (PAGE). The pressure-treated samples showed a time-dependent loss of native whey proteins by SEC and 1D PAGE and a corresponding increase in non-native proteins and protein aggregates of different sizes. These aggregates altered the viscosity and opacity of the samples and were shown to be cross-linked by intermolecular disulfide bonds and by noncovalent interactions using 1D PAGE [alkaline (or native), sodium dodecyl sulfate (SDS), and SDS of reduced samples (SDS(R))] and 2D PAGE (native:SDS and SDS:SDS(R)). The sensitivity of the major whey proteins to pressure was in the order beta-lactoglobulin B (beta-LG B) > beta-LG A > bovine serum albumin (BSA) > alpha-lactalbumin (alpha-LA), and the large internal hydrophobic cavity of beta-LG may have been partially responsible for its sensitivity to high-pressure treatments. It seemed likely that, at 800 MPa, the formation of a beta-LG disulfide-bonded network preceded the formation of disulfide bonds between alpha-LA or BSA and beta-LG to form multiprotein aggregates, possibly because the disulfide bonds of alpha-LA and BSA are less exposed than those of beta-LG either during or after pressure treatment. It may be possible that intermolecular disulfide bond formation occurred at high pressure and that hydrophobic association became important after the high-pressure treatment.  相似文献

Heat-induced aggregation of whey proteins in solutions made from two commercial whey protein concentrates (WPCs), one derived from mineral acid whey (acid WPC) and the other from cheese whey (cheese WPC), was studied using polyacrylamide gel electrophoresis (PAGE), size exclusion chromatography (SEC), and transmission electron microscopy (TEM). Heat treatment (75 degrees C) of acid WPC solutions (12.0%, w/w, pH 6.9) resulted in formation of relatively small "soluble" aggregates that were predominantly disulfide-linked. By contrast, heat treatment of the cheese WPC solutions (under the same conditions) caused formation of relatively large aggregates, containing high proportions of aggregates linked by noncovalent associations. The rate of aggregation of both beta-lactoglobulin and alpha-lactalbumin at 75 degrees C, measured as the loss of native proteins by PAGE, was higher in the cheese WPC solution than in the acid WPC solution. Cross dialysis of the two WPC solutions resulted in alteration of the mineral composition of each WPC solution and reversing their heat-induced aggregation behavior. The results demonstrated that the mineral composition is very important in controlling the aggregation behavior of WPC products.  相似文献

Locoweeds cause significant livestock poisoning and economic loss in the western United States. The toxicity of Embellisia sp. fungi isolated from locoweed was compared with locoweed toxicity using the rat as a model. Rats were fed diets containing locoweed, fungus and alfalfa, or alfalfa. Locoweed- and fungus-fed rats consumed swainsonine-containing food at approximately 1.3 mg x kg(-1) x d(-1), gained less weight (P = 0.001) and ate less than controls. Swainsonine is the principal agent responsible for inducing locoism in animals. The concentrations of alkaline phosphatase and aspartate aminotransferase enzymes were greater (P < 0.05) in serum of locoweed- and fungus-fed rats compared with control rats. Similar intracellular vacuolation was observed in renal, pancreatic, and hepatic tissues of rats that consumed either locoweed or fungus. Rats that ate locoweed or Embellisia fungi displayed indistinguishable toxicity symptoms. The Embellisia fungi from locoweed can induce toxicity without the plants. Locoism management strategies need to involve management of the Embellisia fungi.  相似文献

This study aimed to elucidate the response of diversity and activity of soil invertebrates to elevated soil metal concentrations that were a result of sewage sludge application. Field sampling of soil invertebrates was carried out from 2002 to 2004 at an experimental site established in 1982 to test the effects on crop production of metal contamination from sewage sludge applications with elevated concentrations of zinc (Zn), copper (Cu) and nickel (Ni) with certain treatments exceeding the current UK statutory limits for the safe use of sludge on land. At metal concentrations within the limits, none of the invertebrates sampled showed adverse effects on their abundance or overall community diversity (from Shannon–Weiner index). At concentrations above the limits, individual taxa showed sensitivity to different metals, but overall diversity was not affected. Earthworm abundance was significantly reduced at total Cu concentrations at and above 176 mg kg?1, while nematode and enchytraeid abundances were sensitive to Cu and high Zn concentrations. Correspondingly, litter decomposition was lower in Zn and Cu treatments although there was no direct relationship between decomposition and soil invertebrate abundance or diversity. Such enduring changes in both soil biodiversity and biological activity around the current UK regulatory limits warrant further investigation to determine whether they indicate detrimental damage to soil functioning over the long‐term.  相似文献

Background

Grazed grassland management regimes can have various effects on soil fauna. For example, effects on earthworms can be negative through compaction induced by grazing animals, or positive mediated by increases in sward productivity and cattle dung pats providing a food source. Knowledge gaps exist in relation to the behaviour of different earthworm species i.e. their movement towards and aggregation under dung pats, the legacy effects of pats and the spatial area of recruitment. The present study addressed these knowledge gaps in field experiments, over 2 years, using natural and simulated dung pats on two permanent, intensively grazed pastures in Ireland.

Results

Dung pats strongly affected spatial earthworm distribution, with up to four times more earthworms aggregating beneath pats, than in the control locations away from pats. In these earthworm communities comprising 11 species, temporally different aggregation and dispersal patterns were observed, including absence of individual species from control locations, but no clear successional responses. Epigeic species in general, but also certain species of the anecic and endogeic groups were aggregating under dung. Sampling after complete dung pat disappearance (27 weeks after application) suggested an absence of a dung pat legacy effect on earthworm communities. Based on species distributions, the maximum size of the recruitment area from which earthworms moved to pats was estimated to be 3.8 m2 per dung pat. Since actual grazing over 6 weeks would result in the deposition of about 300 dung pats per ha, it is estimated that a surface area of 1140 m2 or about 11% of the total grazing area can be influenced by dung pats in a given grazing period.

Conclusions

This study showed that the presence of dung pats in pastures creates temporary hot spots in spatial earthworm species distribution, which changes over time. The findings highlight the importance of considering dung pats, temporally and spatially, when sampling earthworms in grazed pastures. Published comparisons of grazed and cut grasslands probably reached incorrect conclusions by ignoring or deliberately avoiding dung pats. Furthermore, the observed intense aggregation of earthworms beneath dung pats suggests that earthworm functions need to be assessed separately at these hot spots.

Efficient soil P cycling is essential for promoting optimal and sustainable grassland growth. The soil biological community is regarded as an important source of available P to the plant community. However, the effects of P fertilization on the soil biota are unclear. This study aimed to investigate the effects of P fertilization on plant and soil biological communities in two intensively managed grassland sites that had been receiving mineral‐based P fertilization over a 14‐year period. Both pastures had been frequently cut and harvested for plant material and had received only inorganic fertilizers from the establishment of the trial. Both sites were sampled four times from October 2009 to April 2011 and plant and a range of soil biological parameters were determined at each sampling period. The main findings of this study showed that soil chemical measures, such as labile inorganic P and total P concentrations, and plant yield and P contents responded as expected to P fertilization. However, all soil biological parameters either showed no response or inconsistent responses to P fertilization over the experimental period. This study indicates that intensive management regimes, for example intensive plant harvesting and fertilizer regimes, appear to override the relationship between plant and soil biological communities with respect to their response to P fertilization, and thus their productivity is apparently not predicated upon biotic activity.  相似文献

The use of indicators in soil monitoring schemes to detect changes in soil quality is receiving increased attention, particularly the application of soil biological methods. However, to date, the ability to compare information from different laboratories applying soil microbiological techniques in broad-scale monitoring has rarely been taken into account. This study aimed to assess the consistency and repeatability of two techniques that are being evaluated for use as microbiological indicators of soil quality: multi-enzyme activity assay and multiple substrate-induced respiration (MSIR). Data were tested for intrinsic (within-assay plate) variation, inter-laboratory repeatability (geometric mean regression and correlation coefficient) and land-use discrimination (principal components analysis). Intrinsic variation was large for both assays suggesting that high replicate numbers are required. Inter-laboratory repeatability showed diverging patterns for the enzyme assay and MSIR. Discrimination of soils was significant for both techniques with relatively consistent patterns; however, combined laboratory discrimination analyses for each technique showed inconsistent correspondence between the laboratories. These issues could be addressed through the adoption of reliable analytical standards for biological methods along with adequate replication. However, until the former is addressed, dispersed analyses are not currently advisable for monitoring schemes.  相似文献
