Agrobacterium-mediated transformation of the CrDAT gene and selection of transgenic periwinkle lines with a high vincristine accumulation

Catharanthus roseus contains vincristine and vinblastine, which are outstanding drugs for cancer. In the biosynthetic pathways of terpenoid indole alkaloids (TIAs) in C. roseus, deacetylvindoline 4-O-acetyltransferase (DAT) is a key enzyme that catalyses the last reaction of vindoline biosynthesis to form vinblastine and vincristine. In this study, the CrDAT transgene was transferred into the periwinkle by Agrobacterium-mediated transformation and generated transgenic periwinkle lines with an increase in vincristine accumulation. The C. roseus DAT gene was introduced into C. roseus plants and it was confirmed that CrDAT was successfully transferred into the genome of periwinkle plants and efficiently translated to synthesise recombinant DAT protein. Four transgenic periwinkle lines in T1 generation, T1-1, T1-3, T1-6, and T1-7, expressed recombinant DAT protein with the total protein content in the range of 2.86 μg.mg^?1 to 5.12 μg.mg^?1. Moreover, the vincristine contents of four transgenic lines increased by 1.63?2.48-fold compared to non-transgenic plants, ranging from 6.91 µg.g^?1 (fresh weight) to 10.53 µg.g^?1 (fresh weight). The T1-1 line had the highest vincristine content. Hence, the overexpression of the recombinant DAT protein can improve the vincristine accumulation of transgenic C. roseus plants.

2009年6月全国畜产品和饲料价格情况

据对全国470个集贸市场畜禽产品和饲料价格定点监测,6月份活猪价格止跌回升,仔猪和猪肉价格降幅趋缓,鸡蛋价格止涨下跌,生鲜乳价格持续下降,牛羊肉价格基本平稳,饲料价格继续上涨.本月猪粮比价5.7:1,比上月下降0.1个点.     

日粮色氨酸水平对仔猪生产性能和血液生化指标影响的研究

在赖氨酸/蛋氨酸+胱氨酸/苏氨酸/异亮氨酸比例为100:68:65:59的玉米—玉米蛋白粉基础日粮中,添加合成的 L-色氨酸构成5种色氨酸(TRP)水平的日粮(TRP 水平分别为0.125%、0.155%、0.180%、0.215%和0.24%)。将35头太湖母猪与长白公猪的杂交仔猪在35日龄断奶时,随机分为5组,分别饲喂上述为同 TRP 水平的日粮,饲养期长达30天。结果表明,日粮(TRP)水平显著影响仔猪的日均耗料量(ADFI)、日均增重(ADG)和饲料转化率(FCR)。TRP 水平为0.215%时,仔猪生长性能最好。TRP 水平为0.24%时仔猪的生长性能与0.215%时差异不显著(P>0.05)。TRP 水平在0.125%～0.215%范围内,仔猪 ADG、ADFI 与 TRP 水平呈正相关,回归方程分别为:ADG(g)=2.57 TRP(mg/kg)-80.21,(r=0.99,P<0.01);ADFI(g)=2.49 TRP(mg/kg)+220.31,(r=0.95,P>0.05);FCR 与日粮 TRP 水平呈显著负相关,回归方程为:FCR=2.877-5.668 TRP(%),(r=0.945,P<0.01).血清尿素氮(SUN)含量与日粮 TRP 水平呈负相关,回归方程为 SUN=19.47-34.35TRP(%),(r=0.95,P<0.05)。从仔猪生长性能和 SUN 两项指标确定 TRP 在理想氨基酸模式中的相应值分别为18和20。在 TRP 水平0.125%～0.215%范围内,血清碱性磷酸酶活性与 TRP 水平呈现正相关,在0.215%时最高;TRP 水平在0.24%时与0.215%时差异不显著(P>0.05)。日粮 TRP 水平对血清谷丙转氨酶(GPT)和谷草转氨酶(GOT)活性无显著影响(P>0.05)。日粮 TRP 水平对血清总蛋白含量无显著影响,而血清蛋白含量随 TRP 水平升高而增加,在0.215%时最高。血清蛋白质系数(A/G)在各 TRP 水平间有显著差异(P<0.05),日粮中 TRP 含量在0.125%时,A/G显著降低(P<0.05),A/G 在 TRP 水平为0.215%时最高,TRP 在0.125%～0.215%之间,A/G 呈现线性递增,回归方程为:A/G=9.34TRP(%)+0.956,r=0.98,P<0.05),而 TRP 水平为0.24%与0.215%的两组 A/G 无显著差异(P>0.05)。     

推求合理畦宽的方法及应用

为了寻求合理的节水省时畦宽,以更好地利用现有水源,在1990、1991年试验和实践的基础上,用统计回归分析方法对大城县农田畦田规格进行了分析,求出了合理畦宽,并从灌水持续时间、用电量、提水量等方面对合理畦宽的节水、节能效果进行了分析。     

Acute infection of encephalomyocarditis (EMC) virus in Syrian hamsters.

One-month-old Syrian hamsters of the APA and Std: golden strains were inoculated intraperitoneally with 10(5) PFU/head of the D variant of encephalomyocarditis (EMC) virus and examined virologically and pathologically up to 7 days after inoculation. APA hamsters developed apparent hyperglycemia due to pancreatic islet cell damage while Std:golden hamsters did not. Hamsters of both strains showed clear histopathologic changes in the testis with prominent viral replication as well as in the brain, heart and exocrine pancreas. The susceptibility to EMC virus-infection was higher in males than in females and in APA than in Std: golden hamsters.     

家蚕病原性微孢子虫孢子表面蛋白的选择性分离与总蛋白比较分析

选择性分离了Ｎｏｓｅｍａｂｏｍｂｙｃｉｓ孢子表面蛋白和总蛋白，采用ＳＤＳ－ＰＡＧＥ检测发现，孢子表面蛋白由分子量不同的３种亚基（３２０００、２６０００、２４５００Ｄａ）组成，其总蛋白至少包括２５个条带。同时将从养蚕生产中收集到的两种微粒子ＭＡ－１和ＭＤ－１以及从野外昆虫收集到的微孢子虫Ｐｈａ－Ｍ和Ｈａ－Ｍ与Ｎ．ｂｏｍｂｙｃｉｓ进行了比较，发现不同微孢子虫间蛋白组成有差异。
