Vitamin C in broccoli (Brassica oleracea L. var. italica) flower buds as affected by postharvest light,UV-B irradiation and temperature

In this study, the changes in vitamin C, l-ascorbic acid (AA) and l-dehydroascorbic acid (DHA) levels in broccoli flower buds were examined during pre-storage and storage periods, simulating refrigerated transport with wholesale distribution and retail, respectively. Broccoli heads were pre-stored for 4 or 7 days at 0 °C or 4 °C in the dark and then stored for 3 days at 10 °C or 18 °C. During storage the broccoli heads were exposed for 12 h per day to three different levels of visible light (13, 19 or 25 μmol m⁻² s⁻¹) or a combination of visible light (19 μmol m⁻² s⁻¹) and UV-B irradiation (20 kJ m⁻² d⁻¹), or they were stored in the dark. The vitamin C content in broccoli flower buds during storage was significantly affected by pre-storage period and temperature. Higher vitamin C levels in flower buds after storage were observed for broccoli heads pre-stored for 4 days or at 0 °C as compared to those pre-stored for 7 days or at 4 °C. Storage temperature also affected vitamin C in broccoli flower buds, with higher levels observed for broccoli stored at 10 °C than at 18 °C. Hence, vitamin C in broccoli flower buds was demonstrated to decrease together with increasing pre-storage period, pre-storage temperature and storage temperature. AA in broccoli flower buds was influenced mainly by storage temperature and to a minor extent by pre-storage temperature. The DHA level and DHA/AA ratio were stable in flower buds of broccoli pre-stored for 7 days, whereas increasing tendencies for both DHA level and ratio were observed after pre-storage for 4 days. These results indicate a shift in the ascorbate metabolism in broccoli flower buds during storage at low temperatures, with its higher rate observed for broccoli pre-stored for shorter time. There were no effects of the light and UV-B irradiation treatments on vitamin C, AA and DHA levels in broccoli flower buds.     

球等鞭金藻三级培养过程中细菌群落多样性分析

球等鞭金藻三级培养过程中生长差异明显,在完全灭菌的一级培养中生长最佳,在未灭菌的三级培养中生长最差。为揭示培养过程中藻际细菌群落多样性与其生长差异的相关性,以球等鞭金藻(Isochrysis galbana)为研究对象,利用Illumina HiSeq平台通过高通量测序的方法对球等鞭金藻三级培养过程中细菌群落多样性进行研究。结果表明,一、二、三级培养组之间的藻际细菌群落组成差异明显。Shannon和Simpson指数分析说明,一级培养组中细菌群落多样性显著低于二级和三级培养组。MetaStat分析发现,一级和三级培养组之间存在两株丰度显著差异的细菌,其中麦氏交替单胞菌(Alteromonas macleodii)在三级培养下丰度显著高于一级,而红球菌(Rhodococcus erythropolis)相反。进一步通过2216E平板涂布法分离并鉴定了麦氏交替单胞菌等17株球等鞭金藻藻际环境细菌;系统进化树构建结果显示,整个进化树分成13个分支,分别对应13个属,包括交替单胞属(Alteromonas)、假交替单胞属(Pseudoalteromonas)、海杆菌属(Marinobacter)等。本研究结果为探索藻菌互作机理奠定了基础,有助于提高水产养殖过程中球等鞭金藻的产量和质量。     

Observations of flow patterns in a model of a marine fish larvae rearing tank

There are few studies on flow fields generated by aerators in fish larvae rearing tanks. The flow varies with aeration rates and tank proportions resulting in different larvae survival rates. The effects of aeration rate and aspect ratio AR (the ratio of liquid depth to tank radius) on overall flow patterns were investigated experimentally using flow visualization techniques. Two distinct types of flow patterns, a single-pair vortex system and a two-pair vortex system, were observed as the value of tank aspect ratio varied from about 1.0 to 2.0. In addition, corner vortex structures were observed in both the region between the free surface and the upper sidewall, and between the bottom wall and the lower sidewall of the tank. On the sidewall, reattachment and separation points were found, which were closely related with these vortex structures.     

香芋复合饮料的研制

为了确定香芋复合饮料的最佳配方，以香芋、干花和大米等为原料，通过打浆、加热研磨等
工艺，加上后期调配获得最终产品。通过正交试验以及对饮料的综合感官评价和理化分析，得到了最佳的
配方。结果如下：当香芋添加量为100 g，水的添加量为1 100 mL 时，蜂蜜和柠檬汁的添加量分别为9%
和0.07%；粳米（晚粳宁88）添加量为10 g；干茉莉花的添加量为2 g；牛奶和枸杞的添加量分别为5 mL
和1 g。该产品具有茉莉花和牛奶的清香和乳香，口感细腻，为淡乳黄色液体，具有营养保健功效。     

锦鲤保护性组织RNA提取及引物扩增研究

从养殖锦鲤各种组织中提取完整的RNA, 是分子生物学研究锦鲤应激表达、体色变异、生长发育等功能基因表达的关键实验。在不伤害养殖锦鲤个体健康的情况下, 取用鱼体的鳍条、鳞片、鳃丝、血液4种保护性组织,可以最大程度地减少对珍贵锦鲤个体的损伤, 并达到研究相关基因表达的目的。本研究利用TR izo l法提取RNA, 获得的各样品RNA条带完整、纯度高。结果表明, 4 种保护性组织提取的RNA 经过逆转录反应, 得到了高质量的CDNA。内标基因( B- actin)、热激蛋白基因( hsp70)、金属硫蛋白基因(MT)扩增得到清晰的条带, 通过NCB I数据上的BLAST比对证明, 各序列的同源性在95%以上。利用本研究的方法, 可以充分保护养殖锦鲤个体的完好性, 用于进一步的分子生物学研究。     

象山港桐照滩涂小型底栖动物丰度和生物量的研究

调查结果表明,象山港桐照滩涂小型底栖动物有11个类群,线虫在丰度上占绝对优势(93%),其次是涡虫、桡足类等.按生物量统计,线虫占53%,其次是多毛类(18%).小型底栖动物的年平均丰度为4168.66个/10 cm2,年平均生物量1828.39 μg /10 cm2.小型底栖动物在丰度和生物量上出现了明显的季节变化,其最高值分别出现在秋季和冬季.并且还出现了明显的垂直分布,超过60%的小型底栖动物分布在(0～2)cm层段,随着深度的增加数量逐渐减少.
