水稻淀粉合成代谢相关基因与千粒重QTLs的连锁分析   总被引:6,自引:1,他引:6  
稻米淀粉的合成和积累是水稻千粒重形成的重要基础之一。在水稻千粒重QTLs定位分析基础上,对本研究克隆的14个水稻胚乳淀粉合成与积累相关基因及已报道的和预测的42个相关基因进行了电子定位分析,比较水稻淀粉合成代谢相关基因与千粒重QTLs的连锁关系。发现有38个基因位点与千粒重QTLs连锁,说明千粒重形成与胚乳淀粉积累与淀粉结构形成代谢密切相关。本研究结果将为进一步研究水稻产量形成的分子机理提供参考。  相似文献

Excised leaves of four rice varieties, ‘Facagro 57’, ‘Facagro 76’, ‘Kirundo 3’, ‘Kirundo 9’, were chilled for six days at 10/5°C day/night, along with 11 somaclonal families arising from three of these varieties, which had been selected over four generations for improved chilling tolerance. At the end of the chilling treatment, electrolyte leakage from leaf pieces was followed for 12 h and compared with an unchilled control. With unchilled leaves, no varietal differences nor a rise in leakage over time were observed. Chilling induced higher leakages in all varieties, except in ‘Kirundo 3’ at the time of first measurement, and a continuous rise over time was found in all varieties. Varietal differences were detected, ‘Kirundo 3’ being the least affected, followed by ‘Kirundo 9’ and ‘Facagro 76’, and finally ‘Facagro 57’, which coincides with their ranking for chilling tolerance established through chilling survival tests. The leakages were generally lower in the somaclonal families than in the variety they originated from in ‘Kirundo 9’ and ‘Facagro 76’, while the opposite was observed in ‘Kirundo 3’. The significance and usefulness of electrolyte leakage measurement for chilling tolerance screening, and the potentials of somaclonal variation for chilling tolerance improvement are discussed.  相似文献

利用来自抗旱性较好的供体亲本(BG300和BG304)、具有两种遗传背景(IR64和特青)的水稻高代回交(BC2)抗旱选择导入系,通过人工接种的方法鉴定纹枯病抗性,考察纹枯病抗性与抗旱性之间可能存在的遗传重叠。通过与受体亲本的纹枯病抗性表现比较发现,具有特青背景的抗旱选择导入系倾向于纹枯病抗性的降低,而IR64背景的抗旱选择导入系则倾向于纹枯病抗性的增强。基于基因型与表型的方差分析共鉴定到6个与纹枯病抗性相关的位点,其中QSbr6在不同供体和背景的两个群体中分别检测到,而QSbr10则在同一供体的两个遗传背景下均检测到;有3个位点(QSbr6、QSbr8和QSbr10)与同一群体中检测到的抗旱性位点位置相近,很可能是两种抗性重叠的遗传基础。尽管方差分析的方法在选择导入系的非选择目标性状相关位点的鉴定中存在相当程度的偏低估计,本研究所检测到的纹枯病抗性位点,特别是那些与抗旱性重叠位点的分子标记以及相关的抗性株系仍将为进一步的水稻纹枯病抗性和抗旱性的多抗性育种和深入的遗传重叠研究提供有用的信息和材料。  相似文献

Following anther culture of various F1 hybrids of indica restorer (R) lines/wide compatible varieties (WCVs) and japonica R lines/WCVs, the homozygous diploid plants (2n) generated were test-crossed with indica WA type cytoplasmic-genetic male sterile (CMS) line Zhanshan 97A (WA), indica testers Nanjing 11 and Nante, japonica BT CMS line Hanfeng A (BT), and japonica testers Balilla and Akihikari to identify widely compatible restorer lines. The results of this study showed that among the diploid pollen plants generated from F1 hybrids of indica R lines/WCVs, 36.7% and 64.7% possessed normal fertility restoration ability (rate of seed-setting > 80%) to Hanfeng A and Zhanshan 97A, respectively. 71.3% and 32.3% had normal fertility restoration ability to Hanfeng A and Zhanshan 97A, respectively, in diploid pollen plants derived from the japonica R lines/WCVs F1's anther culture. Several widely compatible R lines were selected from anther culture of F1 hybrids of indica R lines/WCVs and japonica R lines/WCVs. These widely compatible R lines derived from diploid pollen plants showed good wide compatibility and restoration ability both to WA and BT type CMS lines. Strong standard heterosis of major agronomic traits and yield traits was observed in F1 test-crosses of widely compatible R lines with Hanfeng A and Zhanshan 97A. The wide compatibility in widely compatible R lines H17, H158 and H281 was studied by a set of three-way crosses. Segregation of the fertile plants with seed-setting rate > 70% and semi-fertile plants with seed-setting rate > 69.9% agreed with a ratio of 1:1, indicating that wide compatibility in these widely compatible R lines is governed by a pair of major genes. 利用两组5×5双列杂交,以 Griffing模型Ⅰ分析籽粒蛋白质、游离氨基酸含量的配合力与杂种优势表现。结果表明:1.蛋白质、游离氨基酸含量均受加性和非加性基因效应的共同作用,但蛋白质以加性效应为主,游离氨基酸以非加性效应为主;2.蛋白质不存在显著的胞质效应,而游离氨基酸则存在极显著的胞质效应;3.蛋白质杂种优势较小,其最强的组合超中亲优势值仅为13.75%,而游离氨基酸存在强大的杂种优势,最高的组合可达155.17%;4.蛋白质、游离氨基酸一般配合力间存在极显著线性关系,在一般配合力改良上可同步进行; 5.京香1号、培矮64、IR58、Mcp231-2具有较高的一般配合力和较大的特殊配合力方差,可作为优良亲本利用。  相似文献

Protoplasts were isolated from young inflorescence-derived suspension cultures of a japonica rice cultivar ‘Taipei 177’. The isolated protoplasts which were cultured either in liquid, agar on Sea plaque agarose underwent sustained division. Maximum plating efficiency of 1.06% occurred in a medium containing macroelements of KM, microelements and vitamins of B5, 0.5 % Sea plaque agarose, 1.0 mg/l of 2,4-D, and glucose as an osmotic stabilizer. Green and albino plants were regenerated from the protocalli in MS semisolid medium containing 4 mg/l BAP, 0.5 mg/l NAA and 500 mg/l casein hydrolysate (MS18–2).  相似文献

In a backcrossing programme to transfer desirable characters from wild Avena maroccana Gdgr. to cultivated oats, A. sativa L., meiotically stable plants in BC1F3 and BC2F2 progenies were isolated. The recovery of stable genotypes with 2n = 6×= 42 chromosomes indicated that two backcrosses are enough for such a programme. The cytological observations in various backcross generations are presented and discussed.  相似文献

The brown planthopper (BPH) is one of the most destructive insect pests of rice. Resistant varieties have proved to be one of the most economic and effective measures for BPH management. In this study, an indica rice ‘DV85’ showed resistance to biotype 2 of BPH by bulked seedling test, and a recombinant inbred line (RIL) population derived from a cross between a susceptible rice ‘Kinmaze’ and ‘DV85’ was phenotyped to map genetic factors conferring BPH resistance in ‘DV85′. Composite interval mapping revealed that one quantitative trait locus (QTL) with a LOD score of 10.1 was detected between XNpb202 and C1172 on chromosome 11. This QTL was designated as Qbph11. Qbph11 explained 68.4% of the phenotypic variance of BPH resistance in this population. The allele from the resistant parent ‘DV85’ at Qbph11 reduced the damage caused by BPH feeding and would be very useful in breeding resistant rice varieties via marker‐assisted selection.  相似文献

水稻重要农艺性状的基因定位研究在育种上具有重要意义。2004年在海南陵水县种植两个完全测序水稻品种日本晴与9311的F2群体及双亲,分别考察了其单株分蘖数、穗数、有效分蘖数、穗长、主穗长、抽穗期、株高和剑叶8个农艺性状3次重复的平均值。用已构建的连锁遗传图谱(Nipponbare/9311-F2遗传图谱)及Excel 2000和Mapmaker/QTL 1.1b软件对这8个性状间的相互关系和基因位点进行了分析。结果在LOD>2.0和P<0.005的条件下共检测到41个QTLs,它们分布在水稻所有染色体上,单个QTL对性状表型贡献率11.0%~46.4%,其中大于20%的有22个。对选用已测序材料为亲本构建图谱来探讨水稻农艺性状的分子基础及其育种意义进行了讨论。  相似文献

Two molecular marker approaches [amplified fragment length polymorphism (AFLP) and simple sequence repeat (SSR)] were employed to study genomic relationship among 96 rice cultivars. These included most of the best reputed Italian accessions. AFLP produced 461 fragments, 248 (53%) of which were polymorphic, SSR produced four to 11 alleles in the 12 genomic loci investigated. Genomic similarity was estimated independently for the two molecular marker techniques. Both AFLP and SSR dendrograms agree in splitting the cultivars into two main clusters: a small one, comprising four exotic accessions, and a larger one which could be split into four subgroups. These were also analysed on the basis of historical and pedigree information. This is the first report on the application of DNA polymorphism analysis to reveal genomic relationship among cultivated Italian rice germplasm. Results will be useful for breeding programmes.  相似文献
