Abstract Only a few studies have been conducted in which regenerability of barley has been examined. In the current study, 17 barley genotypes (nine two-row malting type: Aster, Emon, Ruen, Jubiley, PV101, Körten, Krassi 2, Perun and Igri, and eight six-row feed types: Karnobat, Hemus, Jerun, Veslets, Aheloi 2, Diana, Panagon, and Izrgev) were evaluated for tissue-culture response from seedlings during a three-year period. Regenerable calli were obtained from all tested genotypes. Although there was much variation in regeneration capacity among the cultivars, plants were obtained from all cultivars. The majority of green plants grown to maturity were fertile and normal in appearance. The frequency of embryogénie callus induction and regenerability was influenced by genotype and growth conditions of the donor plants. Genotype was the most important determinant of the in vitro response. The best in vitro performance, based on ability to form regenerable calli and to regenerate plants was observed for Ruen, Aster, and Emon. 相似文献

Callus induction and plant regeneration of Mniochloa abersend via lateral shoots were conducted in this study. Murashige and Skoog (MS) medium supplemented with 1.0 mg/L 2,4-dichlorophenoxyacetic acid (2,4-D) and 1.0 mg/L naphthaleneacetic acid (NAA) was effective for compact callus induction. Remarkably, calli on the MS medium with 0.1 mg/L 2,4-D yielded the highest folds of proliferation (8.01), and showed a high potential capacity to differentiate 1 year after subculture. In addition, the compact calli possessed 100% differentiation rate and generated more shoots that were green and strong in 1.0 mg/L kinetin and 1.0 mg/L NAA. Vigorous roots were generated in the 1/2MS supplemented with 0.5 mg/L indole-3-butyric acid, and the resultant plantlets exhibited 90% survival rate after they were hardened and transplanted. 以喷油助燃再生方式为例,基于柴油机微粒捕集器(DPF)过滤体孔道热再生模型,对喷油助燃再生装置在不同工作参数下即喷油压力、喷油率、补气量等单因素变化时对再生过程过滤体排气背压、孔道内微粒层厚度及壁面峰值温度等变化规律进行了研究与分析.结果表明:燃烧器的工作参数喷油压力、喷油率、补气量等对再生过程有显著影响.随着喷油压力的提高,过滤体孔道再生壁面峰值温度升高,加快再生速率提高喷油率可加大燃烧器的功率,再生过程壁面峰值温度增幅较大,从过滤体的安全耐热温度来看应加以控制,但喷油率提高到一定程度后,使排气中氧含量不足,燃烧器燃烧效率下降,且影响微粒的氧化速度,导致再生速率随之下降.适当补气可增加排气中氧含量,可加快再生过程,但进一步加大补气量后由于对流传热散失作用加强将减缓再生过程. 相似文献

本试验对小麦两类不育材料幼胚愈伤诱导及分化进行了研究,结果表明:①N6培养基愈伤组织诱导率最高95%~100%;不同材料平均出愈率由高到低依次为雄性不育1号、雄性不育2号、雌性不育1号;②接种18~30dMS培养基最早产生不定芽分化。最早可见不定芽分化的材料为雄性不育1号,分化进程较雌性不育幼胚提早7~10d;雄性不育1号和雌性不育1号幼胚愈伤组织分别培养45,52d分化率达峰值均为100%,雄性不育2号材料在培养112d分化率达峰值67.4%;③单独加入2,4 D并随2,4 D浓度降低不定芽分化率有增加的趋势。2,4 D与KT配合使用不定芽分化率最高,达100%,以低浓度2,4 D(0.25~0.50mg·L-1)与KT(0.1~0.2mg·L-1)配合为宜。④分化不定芽在不同培养基中生长增殖不同,以C17为最佳同时附加KT0.2mg·L-1 2,4 D0.1mg·L-1分化苗可增殖6倍;再生植株移栽成活率80%以上;接种至移栽历时4个月。 相似文献

The widespread adoption of the sediment fingerprinting approach to guide catchment management has been limited by the cost and the difficulty to prepare and process samples for geochemical and radionuclide analyses. Spectral properties have recently been shown to provide a rapid and cost‐efficient alternative for this purpose. The current research objective was (i) to quantify the sediment source contributions in a 1∙19‐km2 rural catchment of Southern Brazil by using mid‐infrared (MIR) spectroscopy and (ii) to compare these results with those obtained with geochemical approach and near‐infrared and ultraviolet–visible spectroscopy methods. The sediment sources to discriminate were cropland surface (n = 20), unpaved roads (n = 10) and stream channel banks (n = 10). Twenty‐nine suspended sediment samples were collected at the catchment outlet during nine significant flood events. The sources could be distinguished by MIR spectroscopy. Cropland and channel bank sources mainly differed in their clay mineral contents, but their similar organic matter content complicated the MIR‐model predictions. Unpaved road contributions were discriminated from the other sources by their lower organic carbon content. When the results of the current research based on MIR spectroscopy are compared with those obtained using other sediment fingerprinting approaches, based on geochemistry and near‐infrared and ultraviolet–visible spectroscopy, an overestimation of channel banks contribution and an underestimation of cropland and unpaved road contributions is found. These results suggest that MIR spectroscopy can provide a useful tool that is non‐destructive, rapid and cheap for tracing sediment sources in rural catchments and for guiding the implementation of soil and water conservation measures. 以10个不同基因型黄瓜为试材进行游离小孢子培养,通过对成胚条件的系统研究,从‘7447’和‘Poinsett97’中获得了子叶形胚和再生植株。研究结果表明:基因型和小孢子发育时期是限制黄瓜游离小孢子成胚的关键因子。不同品种间胚状体产量差异显著,每皿产胚1.5~33.4个。单核靠边期是进行黄瓜游离小孢子培养的最佳时期。低温预处理有利于胚状体的诱导,4 ℃预处理2~4 d为宜,以处理2 d时胚状体产量最高。外源激素在诱导胚状体时并非必需,但低浓度的外源激素(0.5 mg·L-1 2,4-D、0.2 mg·L-1 6-BA)更能促进小孢子成胚。NLN和B5两种基础培养基在诱导胚状体上无显著差异。子叶形胚易分化成苗,而其它类型的胚状体不能获得再生植株。 相似文献
