用纯化的猪繁殖与呼吸综合征病毒重组N蛋白作为包被抗原,建立了检测猪繁殖与呼吸综合征病毒抗体的间接ELISA方法,并确定了ELISA最佳工作条件:抗原包被浓度为0.27μg/ml,37C1h加4C过夜,血清(1:40)和酶标免抗猪IgG(1:400)分别在37℃温育30min,底物溶液37℃显色15min。经重复性试验、交叉试验、阻断试验等试验结果表明该方法重复性好、特异性强、灵敏度高;与美国IDEXX试剂盒相比较,特异性和敏感性分别为97.6%和92.1%,无显著性差异。用已建立的方法检测临床血清样本187份,总阳性率为30.5%。 相似文献

ABSTRACT: The present study investigates the relationship between oocyte development and serum steroid hormone levels in captive Pacific herring, Clupea pallasii , during the first reproductive cycle. The process of oocyte development in Pacific herring belongs to the group-synchronous type. Maturity of the ovary was divided into six periods based on histological observation (i.e. immature (April to September), onset of vitellogenesis (August to October), progress of vitellogenesis (October to December), completion of vitellogenesis (December to March), maturation and spawning (March to April) and spent (late April)). The pattern of seasonal change in the gonadosomatic index (GSI) well reflected the ovarian maturity. Serum vitellogenin levels showed good correlation with change in GSI, which increased from September to a peak (4.2 ± 0.3 mg/mL) in March. Serum estradiol-17β (E2) levels elevated from September and reached a peak (15.8 ± 4.2 ng/mL) in December, and remained comparatively high until March, suggesting that the active vitellogenin synthesis during vitellogenesis is controlled by the high E2 level. 17,20β-Dihydroxy-4-pregnen-3-one showed a single sharp peak (2.4 ± 0.28 ng/mL) in early April of the second year, suggesting it was a maturation-inducing steroid in this species. 相似文献

用免疫组化ABC法,对发情周期中奶山羊下丘脑-垂体-卵巢轴催产素(OT)分布进行了观察研究.结果表明,下丘脑中分泌OT的神经元主要分布在室旁核和视上核,在穹窿周核、腹内侧核、腹外侧核、交叉上核、背内侧核、乳头体、下丘脑外侧区、下丘脑前核等核团也有一定数量的阳性神经元;阳性神经纤维仅见于室旁核、下丘脑前核、视上核等少数核团,在正中隆起和第3脑室室周可见到一定数量的阳性神经纤维.在垂体前叶未见到OT免疫反应阳性产物,自垂体柄和正中隆起的一侧可见到平行排列的OT阳性神经纤维断续地延伸至神经部.卵巢的卵泡及间质未见OT免疫阳性反应,,在黄体组织中存在数量较多的免疫反应阳性细胞,阳性细胞主要呈圆形、卵圆形,小梁两侧及黄体中央近腔区域的阳性细胞呈长梭形,有相当数量的阳性细胞具有突起.连续切片HE染色对照观察显示,黄体中OT主要由大黄体细胞产生,但小黄体细胞也存在OT免疫阳性反应. 相似文献

The in vitro antiproliferative, apoptotic and cell‐cycle effects of 2‐methoxyestradiol (2ME2), an endogenous oestrogen metabolite, were investigated using a variety of canine tumour cell lines. The cells were cultured under standard conditions and incubated with varying concentrations of 2ME2. Inhibition of tumour cell proliferation was evaluated using a tetrazolium‐based colorimetric assay. DNA content analysis was performed using propidium iodide staining and flow cytometry. Cytologic analysis with Leukostat staining solution and Hoechst 33342 staining and Annexin V‐fluorescein isothiocyanate (FITC) fluorescence were used to quantify cell‐cycle distribution and apoptosis induction. Tumour cell proliferation was inhibited by 50% at concentrations of 2ME2 ranging from 0.88 to 7.67 µM, depending on the cell line tested. Profound G2/M phase arrest, an increase in binucleate cells and induction of apoptosis were observed in all cell lines tested, in a dose‐dependent manner. Based on these results, this compound has potential as an agent for the treatment of canine cancer and warrants further investigation. Abstract– I collected bluntface shiners, Cyprinella camura , once or twice monthly from February to November 1991 in Morganfork Creek, southwestern Mississippi. Examinations of mean largest oocyte diameter (LOD), female mean relative gonadal index (RGI), ovarian developmental condition, and male secondary sexual characteristics indicated that C. camura spawned from late March through mid-August. There were significant positive correlations between mean LOD and mean daylength, mean maximum temperature, and mean minimum temperature (environmental variable means were calculated for the 28 day period prior to each sampling date). Mean day length and mean RGI were significantly positively correlated, but there was no significant relationship between mean maximum and minimum temperatures and mean RGI. Clutch size, RGI, and mean mature oocyte diameter (MMOD) of mature females were significantly correlated with length (standard length). Mean length, clutch size, and RGI of mature females in individual collections decreased significantly over the reproductive season. The corresponding decrease in mean MMOD was not significant. 相似文献

The changes of soluble sugar and total nitrogen content of vegetative organs in faba bean, sugar-nitrogen ratio, abscission of reproductive organs and the relationship between sugar-nitrogen ratio and abscission probability were studied using the faba bean cultivar Xichang Dabai . Plants were field grown in 1986, 1988 and 1990. The lands were separated into high yield land, middling yield land and low yield land according to grain yield at maturing stage. The soluble sugar content in vegetative organs (leaf + stem) was greatest from branching to floral initiation stages and pod-set to filling grain stages, and distinctly decreased at the onset of bloom and maturing stages. The total N content was greatest at branching stage, and gradually decreased with growth. In general, the sugar and total N contents in high yield lands were greatest, followed by that in middling yield and low yield lands. The sugar-nitrogen ratio varied depending on the soluble sugar content.
Average yield on high yield lands, middling yield land and low yield land across 3 years were 5048.77 kg/ha, 3744.01 kg/ha and 2378.22 kg/ha, respectively. The total reproductive organs abscission exceeded 90 % of total flower buds. Abscission probability averaged across 3 yield lands and 3 years were 21.6 %, 84.0 % and 32.8 % in flower buds, flowers, and pods, respectively. Node 7–12 of canopy produced more flowers, and fewer flowers and pods were produced in the upper and lower regions. Photosynthate supply has been implicated in control of abscission. Faba bean reproductive organs abscission probability increased with the sugar-nitrogen ratio of vegetative organs decreasing.
