母源抗体的干扰是重组鸡痘病毒(FPV)活载体基因工程疫苗至今未能得到推广应用的主要原因,本试验使用FPV新的复制非必需区构建的载体pP12-18构建在高母源抗体商品鸡具有较高免疫力的基因工程疫苗.将H5亚型禽流感病毒分离株的血凝素(HA)基因和神经氨酸酶(NA)基因定向插入鸡痘病毒转移载体pP12-18中,H5A和NA基因的启动子分别为PS和PE/L,获得用不同的启动子启动不同的外源基因且两基因盒方向为背向串联的重组转移载体p12LSH5HANA.将p12LSH5HANA转染至已感染鸡痘病毒282E4疫苗株(wt-FPV)的鸡胚成纤维细胞(CEF)中.p12LSH5HANA与wt-FPV基因组DNA之间的同源重组产生了重组鸡痘病毒rFPV-12LSH5HANA.通过在含X-Gal的营养琼脂上连续挑选蓝色病毒蚀斑,获得纯化的重组病毒.经传代证实该重组病毒具有良好的遗传稳定性.用105PFU的rFPV-12LSH5HANA免疫无特定病原体(SPF)鸡,能激发机体产生有效的血凝抑制(HI)抗体.初步的动物试验表明,该重组病毒能使经滴鼻点眼攻毒的SPF鸡抵抗H5亚型AIV的致死性攻击,保护率为100%.在高母源抗体的商品鸡上,rFPV-12LSH5HANA与原有载体构建的重组疫苗rF-PV-11SH5HANA的免疫效力有显著差异,保护率分别为81.4%和45.4%.结果表明,选择FPV合适的复制非必需区构建载体是提高重组FPV在高母源抗体商品鸡免疫效力的有效策略之一. 相似文献

After‐effects of maternal soil treatment, controlling for photoperiod and weather conditions, were tested in a nursery experiment with Pinus sylvestris L. seedlings. Three full‐sib crosses were replicated in 4 different soil treatment plots of a grafted experimental seed orchard (63"54'N). Weight of filled seeds, percentage of empty seeds, first year hypocotyl and epicotyl length, and autumn frost hardiness (from artificial freeze testing) were evaluated. Systematic effects from maternal soil treatment were small compared to family differences for all characters studied. However, epicotyl length of the progenies showed a positive relationship with N needle content of the mother grafts. The results also indicated an important interaction between maternal soil treatment and autumn frost hardiness of the families. It is suggested that the altered mother tree treatment could lead to a change in progeny performance, corresponding to a 1.5 degree difference in latitudinal origin of natural stand seedlings. The results demonstrate the advantages of providing the mother tree with uniform conditions in early progeny testing. 相似文献

Pregnancy‐associated glycoproteins (PAG) constitute a large family of glycoproteins found in the outer placental epithelial cell layer of the placenta in Eutherian species. In ruminants, they are noted to be structurally closely related among the different species. This study was designed to determine PAG concentrations in maternal and fetal plasma, allantoic and amniotic fluids in buffalo species. Antisera (AS) generated in rabbits against distinct PAG molecules were used in three radioimmunoassay (RIA)‐PAG systems: RIA‐1 (antiserum raised against bovine PAG67kDa; AS#497), RIA‐2 (antiserum raised against caprine PAG55 + 62 kDa; AS#706) or RIA‐3 (antiserum raised against buffalo PAG; AS#859). Samples were collected at a slaughterhouse (n = 67). PAG concentrations determined by RIA‐2 gave significantly higher results in both allantoic and amniotic fluids (12.7 ± 2.1 ng/mL and 24.0 ± 7.3 ng/mL, respectively). Regarding maternal and fetal plasma, PAG concentrations obtained by RIA‐2 (21.8 ± 2.4 ng/mL and 20.2 ± 2.5 ng/mL, respectively) and RIA‐3 (25.0 ± 2.2 ng/mL and 21.9 ± 3.2 ng/mL, respectively) were higher than those obtained by RIA‐1 (15.5 ± 1.4 ng/mL and 16.1 ± 1.8 ng/mL, respectively). The correlation among the three systems was very high. The study clearly reveals the ability of different PAG‐RIA systems to measure PAG concentration in swamp buffalo samples. 相似文献

Variance components and genetic parameters for greasy fleece weights of Muzaffarnagari sheep maintained at the Central Institute for Research on Goats, Makhdoom, Mathura, India, over a period of 29 years (1976 to 2004) were estimated by restricted maximum likelihood (REML), fitting six animal models including various combinations of maternal effects. Data on body weights at 6 (W6) and 12 months (W12) of age were also included in the study. Records of 2807 lambs descended from 160 rams and 1202 ewes were used for the study. Direct heritability estimates for fleece weight at 6 (FW6) and 12 months of age (FW12), and total fleece weights up to 1 year of age (TFW) were 0.14, 0.16 and 0.25, respectively. Maternal genetic and permanent environmental effects did not significantly influence any of the traits under study. Genetic correlations among fleece weights and body weights were obtained from multivariate analyses. Direct genetic correlations of FW6 with W6 and W12 were relatively large, ranging from 0.61 to 0.67, but only moderate genetic correlations existed between FW12 and W6 (0.39) and between FW12 and W12 (0.49). The genetic correlation between FW6 and FW12 was very high (0.95), but the corresponding phenotypic correlation was much lower (0.28). Heritability estimates for all traits were at least 0.15, indicating that there is potential for their improvement by selection. The moderate to high positive genetic correlations between fleece weights and body weights at 6 and 12 months of age suggest that some of the genetic factors that influence animal growth also influence wool growth. Thus selection to improve the body weights or fleece weights at 6 months of age will also result in genetic improvement of fleece weights at subsequent stages of growth. 相似文献

Estimates of (co)variance components were obtained for weights at birth, weaning and 6, 9 and 12 months of age in Chokla sheep maintained at the Central Sheep and Wool Research Institute, Avikanagar, Rajasthan, India, over a period of 21 years (1980–2000). Records of 2030 lambs descended from 150 rams and 616 ewes were used in the study. Analyses were carried out by restricted maximum likelihood (REML) fitting an animal model and ignoring or including maternal genetic or permanent environmental effects. Six different animal models were fitted for all traits. The best model was chosen after testing the improvement of the log-likelihood values. Direct heritability estimates were inflated substantially for all traits when maternal effects were ignored. Heritability estimates for weight at birth, weaning and 6, 9 and 12 months of age were 0.20, 0.18, 0.16, 0.22 and 0.23, respectively in the best models. Additive maternal and maternal permanent environmental effects were both significant at birth, accounting for 9% and 12% of phenotypic variance, respectively, but the source of maternal effects (additive versus permanent environmental) at later ages could not be clearly identified. The estimated repeatabilities across years of ewe effects on lamb body weights were 0.26, 0.14, 0.12, 0.13, and 0.15 at birth, weaning, 6, 9 and 12 months of age, respectively. This study evaluated the importance of the environment and spatial distribution of juvenile fish for the adoption of alternative migratory tactics in a partially anadromous population of rainbow trout (Oncorhynchus mykiss) from the Santa Cruz River. We captured young‐of‐the‐year fish along the river during autumn 2009, 2010 and spring 2010 and determined their maternal origin (anadromous vs. resident) using strontium to calcium ratios in the otolith core. Relative proportion of anadromous offspring, modelled with logistic regression, increased towards headwaters and in areas with deeper channels and larger substrate composition. Body length, modelled with linear multiple regression, varied positively with site depth, water velocity, substrate size and anadromous maternal origin. Based on evidence for limited juvenile movements (<25 km), the spatial extent of this study (240 Rkm) and the identification of large, contrasting reaches along the river, it is likely that the observed spatial distribution of juveniles and their association to sites with coarse substrate composition reflects maternal spawning activity. Results further indicate that anadromous females breed predominantly in middle and upper river sections in areas with coarse substrate. Given that body size in this system has been positively related to propensity for anadromy, we propose that female spawning choice affects their offspring's spatial distribution, providing the adequate physical template for anadromous offspring to reach or maintain larger body sizes and display anadromy themselves. Relevance of this study is also discussed in the context of alterations in response to future dam construction in one of the latest free‐flowing rivers sustaining anadromous O. mykiss. 相似文献

Abstract – Understanding how maternal and ova traits influence offspring quality is a key issue in both ecological research and fish culture. We examined the survival and size at hatch of walleye embryos in relation to a suite of maternal and ova traits in laboratory incubation trials involving two native walleye populations. Ova were analysed for size, lipid content, fatty acid composition, soluble protein content and size composition, and mineral (Ca, Co, Cu, Fe, Mg, Mn and Zn) content. Embryonic survival (egg hatching success) varied most consistently with respect to spawning date and maternal age. Embryonic survival increased with maternal age, but other indices of maternal health, such as condition (residual mass at length) or somatic lipid content, did not account for significant amounts of variation. Indices of ova total lipid content, fatty acid composition and mineral composition explained significant variation in embryonic survival not accounted for by maternal age and spawning date, whereas indices of protein content and composition generally explained less variance. Both larval mass and length at hatch were primarily determined by ovum size, but length at hatch was also related to some aspects of ova fatty acid and mineral composition. 相似文献

本试验采用IHA试验方法,对经猪瘟灭活疫苗免疫的猪抗体产生的水平进行了监测,并对仔猪母源抗体的消长规律进行了研究.结果表明仔猪母源抗体水平与母猪抗体水平呈正相关,其首次免疫抗体增长幅度与母源抗体呈负相关,但其抗体水平与免疫次数呈正相关.主要免疫的抗体效价与免疫剂量呈正相关,而与首免日龄无关.攻毒结果表明:当IHA<6log2时,仅能抗死亡;当IHA≥7log2时,可抗感染. 相似文献
