采用未成熟SD大鼠肝细胞微粒体为试验材料,以2,3,7,8-四氯-二苯基-并-二哑噁英(2,3,7,8- tetrachlorobenzo-p-dioxin,TCDD)作为参照,利用甲氧基-异酚噁脱甲基酶(methoxyresorufin-O- demethylase,MROD)竞争性抑制动力学和荧光光谱法分析法,测定了5种多溴联苯(polybrominated diphenyl ethers,PBDEs)对7-甲氧基-异酚噁唑脱甲基反应的影响,间接研究了这些PBDEs对CYP1A2 的竞争性抑制作用。结果表明,和TCDD相比,PBDEs对MROD活性的抑制作用较弱,亦即对CYP1A2的 竞争性抑制作用较弱。  相似文献

以磺胺间甲氧嘧啶钠注射液为例,阐述永停滴定法含量测定结果的不确定度评定。通过建立数学模型,分析不确定度来源,对各个不确定度分量进行了评估,计算了扩展不确定度,含量测定结果可表述为(95.3±0.8)%(k=2)。结果表明,重复性测量、校准和温度差异引入的不确定度为测量结果不确定度的主要来源。  相似文献

A sample preparation method based on ultrasound assisted-extraction (UAE) of Ca, Mg and P from swine feed has been described. The experiment was performed to cover the variables influencing the sonication process and, the method validation using standard reference material. Final solutions obtained upon sonication were analyzed by flame atomic absorption spectrometry (for Ca and Mg) and by UV–vis spectrophotometry (for P). The best conditions for metal extraction were as follows: sample mass: 100 mg in 20 mL 0.10 mol/L HCl, a particle size: <60 μm, sonication time: 5 cycles of 10 s and ultrasound power: 102 W. The UAE method was applied in digestibility assays in different piglet feeds and their results showed that it is highly comparable (P > 0.05) to the other methods used for such purposes, as block digestion, and offered a Ca, Mg and P method of quantification limit of 10.6, 12.4 and 14 mg/kg, respectively. The major advantages of the UAE method compared to other methods are the high treatment rate, low reagent usage in the extracts and, it does not generate toxic residues that might negatively affect human health and the environment, accompanied by good precision and accuracy.  相似文献

This study aimed to evaluate the sampling method for the detection of Ascaris suum larval DNA in chicken livers using real-time PCR. Chickens were inoculated with A. suum eggs of a single dose (Group A) or repeatedly low doses (Group B). White spots (WSs) were continuously observed on liver from day 3 after the last infection in Group B and day 14 in Group A. In Group A, larval DNA was detected in WS lesions (78.6%) at a significantly higher rate than in the remaining tissue samples (31.3%). In conclusion, applying WS lesions to the assay improved the detection rate of A. suum DNA in chicken livers, especially in the case of a single infection.  相似文献

Summary Wheat seedlings, treated with the auxine 2,4-dichlor-phenoxy acetic acid (2,4-D) during germination developed only a residual root system. Root elongation was extremely restricted and root tips were deformed to thick club-shaped tumours. When 2,4-D was added in a later stage of plant growth the plants developed additional nodule-like knots along primary roots. Root and shoot dry-matter production was slightly repressed in all 2,4-D treatments and N translocation from roots to shoots was repressed as well. When transferred to an auxine-free growth medium, the 2,4-D-affected roots were not capable of complete recovery. In plants inoculated gnotobiotically with Azospirillum brasilense, either with the wild type or with the NH 4 + -excreting mutant strain C3, a 2,4-D addition increased rhizosphere acetylene-reduction activity at pO2 1.5 kPa. The O2 sensitivity of root-associated nitrogenase activity tended to be reduced. The number of root-colonizing bacteria, at approximately 108 colony-forming units (cfu) per g dry root, was similar in the 2,4-D treatments and untreated controls. Plant treatment with high concentrations of the chemical isomer 3,5-dichlor-phenoxy acetic acid (3,5-D) did not have comparable effects, either on plant development or on rhizosphere-associated nitrogenase activity. Root-tumour tissue inhabited by A. brasilense showed purple staining when subjected to a tetrazolium chloride solution, which may indicate intensive local nitrogenase activity in this tissue. Exposed to an 15N2-enriched atmosphere, plants treated with 2,4-D and with A. brasilense incorporated significantly higher amounts of 15N than untreated controls. In all cases the highest values of 15N enrichment were found following inoculation with the NH 4 + -excreting mutant strain C3. Present address: F. A. Summary Previous laboratory and greenhouse studies have shown that phages significantly reduce soil populations of homologous rhizobia. Reductions in nodulation and N2 fixation have also been observed. The purpose of the current study was to examine the effect of a phage specific ofBradyrhizobium japonicum USDA 117 on nodulation, nodule occupancy, N2 fixation and soybean growth and yield under field conditions. The phage was inoculated in combination withB. japonicum USDA 117 and/orB. japonicum USDA 110 (resistant strain) into a rhizobia-free sandy loam soil and planted toGlycine max (L.) Merr. Williams. When the phage was applied to soil inoculated withB. japonicum USDA 117 alone, significant reductions in nodule weight and number, shoot weight, foliar N, nitrogenase activity, and seed index were observed. When, however, the soil also contained the non-homologous strain,B. japonicum USDA 110, no significant effects on any of these parameters were found. Nodule occupancy by competing strains ofB. japonicum USDA 110 and USDA 117 was also affected by the phage. In soil which did not contain the phage, 46% and 44% of the identified nodules were occupied by USDA 110 and 117, respectively. When the phage was present in the soil, nodule occupancy byB. japonicum USDA 117 was reduced to 23%, while occupancy byB. japonicum USDA 110 was increased to 71%. These results suggest that nodulation by selected strains of rhizobia can be restricted and nodulation by more effective, inoculated strains can be increased through the introduction of a homologous phage to soils.  相似文献

The artificial chromophoric substrate analog of phytic acid, 5-O-[6-(benzoylamino)hexyl]-d-myo-inositol-1,2,3,4,6-pentakisphosphate (T-IP5), may prove useful in measuring soil phytase activity. This chemical probe allows for direct measurement of phytase-catalyzed dephosphorylation (i.e., hydrolysis of the phosphoester bond) using high-performance liquid chromatography with UV detection. Before T-IP5 can be used to measure phytase activity in environmental samples (soil, stream sediment, manure) refinement of the T-IP5 probe methodology is required. Using 31P nuclear magnetic resonance (NMR) spectroscopy, we identified 5-O-[6-(benzoylamino)hexyl]-d-myo-inositol-trisphosphate (T-IP3) as the key intermediate that accumulates during phytase-catalyzed dephosphorylation of T-IP5. An improved HPLC method for separation of reactants is also presented.  相似文献
